This study describes the isolation and characterization of a novel fungus, Aspergillus flavus BS1 and its cellulolytic activities with special emphasis on endoglucanase production. Preliminary screening studies showed that A. flavus BS1 was a potent strain for the production of cellulase. To study the cellulolytic activities in detail by submerged fermentation (SmF), productions of endoglucanase, exoglucanase, and β-glucosidase were estimated from the basal salt medium (BSM) supplemented with 1 % carboxy methyl cellulose (CMC). CMC medium supported the maximum yield of endoglucanase (2,793 U/ml) on day 5 of incubation at 28 °C and 150 rpm, which was higher than that obtained with naturally available supplements (flour) from banana, tapioca, potato, or banana peel. During cellulase production by solid-state fermentation, 10 % (w/w) tapioca flour in sawdust (teak wood) moisturized with BSM (1:2, w/v) supported maximum cellulase yield (5,408 U/g dry substrate) on day 3 at 28 °C, which was 2-fold higher than that obtained during SmF. The active cellulase was qualitatively estimated by polyacrylamide gel electrophoresis (PAGE). Native-PAGE (0.25 % CMC impregnated on the 10 % gel) activity staining with congo-red showed a clear zone for CMCase activity, whereas SDS-PAGE showed a distinct band. In conclusion, this study showed that A. flavus strain BS1 is a potent strain for the production of cellulase on lignocellulosic media, the hot enzyme for bioethanol production from the lignocellulosic biomass by SSF.  相似文献

This study illustrates a novel strain (designated as BPU5) of Bacillus thuringiensis (Bt) isolated from the rumen of Malabari goat, capable of producing polymorphic δ‐endotoxin crystals concomitantly with sporulation in Luria–Bertani medium (LB), and the δ‐endotoxin was efficient to combat Tetranychus macfarlanei, a devastating mite. Polymorphic δ‐endotoxin crystals produced were assessed by scanning electron microscopy and monitored its production concomitantly with sporulation in LB with or without sugar supplements. Toxicity of the δ‐endotoxin was assessed on T. macfarlanei using leaf disc bioassay method. Mortality rate was determined by comparing the survival of mites on the diet (prepared in 10% sucrose and powdered rice husk) containing different concentrations (1–10 mg/ml) of 72‐h‐old crude pellet (dried mixture of δ‐endotoxin (17 mg/g pellet), endospores and a few vegetative cells) or control diet with autoclaved pellet. The maximum production (1.39 mg/ml) of δ‐endotoxin was observed at 72 h in LB. Among the sugars (glucose, sucrose, maltose or lactose) tested as additional carbon source, glucose (8 g/l) enhanced (1.82 mg/ml) the production of δ‐endotoxin by 30%. The lethal concentration (LC50) required to kill 50% mites was estimated as 8.024 mg/ml. The δ‐endotoxin produced by B. thuringiensis BPU5 is shown to efficiently combat T. macfarlanei, a devastating mite infesting agricultural fields.  相似文献

It is unknown which factors determine the changes in cerebrospinal fluid (CSF) pressure inside the craniospinal system during the changes of the body position. To test this, we have developed a new model of the CSF system, which by its biophysical characteristics and dimensions imitates the CSF system in cats. The results obtained on a model were compared to those in animals observed during changes of body position. A new model was constructed from two parts with different physical characteristics. The "cranial" part is developed from a plastic tube with unchangeable volume, while the "spinal" part is made of a rubber baloon, with modulus of elasticity similar to that of animal spinal dura. In upright position, in the "cranial" part of the model the negative pressure appears without any measurable changes in the fluid volume, while in "spinal" part the fluid pressure is positive. All of the observed changes are in accordance to the law of the fluid mechanics. Alterations of the CSF pressure in cats during the changes of the body position are not significantly different compared to those observed on our new model. This suggests that the CSF pressure changes are related to the fluid mechanics, and do not depend on CSF secretion and circulation. It seems that in all body positions the cranial volume of blood and CSF remains constant, which enables a good blood brain perfusion.  相似文献

The present study reports the length‐weight relationships (LWRs) of two clupeoid fish species, Thryssa malabarica (Bloch, 1795) and Thryssa dayi Wongratana, 1983 from Kerala waters, southwest coast of India during the period October 2015 to September 2016. Altogether 88 specimens of T. malabarica and 97 T. dayi were collected on weekly basis from gill nets (mesh size, 2.6–5.0 cm) and ring seines (mesh size, 0.8–2.6 cm) during the period. The value of parameter b in the equation of LWRs was estimated as 3.237 and 3.248 for T. malabarica and T. dayi respectively.  相似文献
