Human immunodeficiency virus protease activity can be regulated by reversible oxidation of a sulfur-containing amino acid at the dimer interface. We show here that oxidation of this amino acid in human immunodeficiency virus type 1 protease prevents dimer formation. Moreover, we show that human T-cell leukemia virus type 1 protease can be similarly regulated through reversible glutathionylation of its two conserved cysteine residues. Based on the known three-dimensional structures and multiple sequence alignments of retroviral proteases, it is predicted that the majority of retroviral proteases have sulfur-containing amino acids at the dimer interface. The regulation of protease activity by the modification of a sulfur-containing amino acid at the dimer interface may be a conserved mechanism among the majority of retroviruses.  相似文献

Rotation in rats was employed as an assay of the central dopaminergic activity of 3,4-methylenedioxymethamphetamine (MDMA). This agent was observed to possess predominantly amphetamine-like actions at low doses. However, at higher doses it also appears to stimulate the dopamine receptor directly. Following a third dose of MDMA, a significant decrease in rotation was evident to this drug and to amphetamine, suggesting a neurotoxic or long-term suppressive action of MDMA.  相似文献

In the past two decades, our ability to study cellular and molecular systems has been transformed through the development of omics sciences. While unlimited potential lies within massive omics datasets, the success of omics sciences to further our understanding of human disease and/or translating these findings to clinical utility remains elusive due to a number of factors. A significant limiting factor is the integration of different omics datasets (i.e., integromics) for extraction of biological and clinical insights. To this end, the National Cancer Institute (NCI) and the National Heart, Lung and Blood Institute (NHLBI) organized a joint workshop in June 2012 with the focus on integration issues related to multi-omics technologies that needed to be resolved in order to realize the full utility of integrating omics datasets by providing a glimpse into the disease as an integrated “system”. The overarching goals were to (1) identify challenges and roadblocks in omics integration, and (2) facilitate the full maturation of ‘integromics’ in biology and medicine. Participants reached a consensus on the most significant barriers for integrating omics sciences and provided recommendations on viable approaches to overcome each of these barriers within the areas of technology, bioinformatics and clinical medicine.  相似文献

The zona pellucida is an extracellular matrix consisting of three glycoproteins that surrounds mammalian eggs and mediates fertilization. The primary structures of mouse ZP1, ZP2, and ZP3 have been deduced from cDNA. Each has a predicted signal peptide and a transmembrane domain from which an ectodomain must be released. All three zona proteins undergo extensive co- and post-translational modifications important for secretion and assembly of the zona matrix. In this report, native zonae pellucidae were isolated and structural features of individual zona proteins within the mixture were determined by high resolution electrospray mass spectrometry. Complete coverage of the primary structure of native ZP3, 96% of ZP2, and 56% of ZP1, the least abundant zona protein, was obtained. Partial disulfide bond assignments were made for each zona protein, and the size of the processed, native protein was determined. The N termini of ZP1 and ZP3, but not ZP2, were blocked by cyclization of glutamine to pyroglutamate. The C termini of ZP1, ZP2, and ZP3 lie upstream of a dibasic motif, which is part of, but distinct from, a proprotein convertase cleavage site. The zona proteins are highly glycosylated and 4/4 potential N-linkage sites on ZP1, 6/6 on ZP2, and 5/6 on ZP3 are occupied. Potential O-linked carbohydrate sites are more ubiquitous, but less utilized.  相似文献

Spondylosis in the desert sand rat (Psammomys obesus) has been studied as a model for intervertebral disc degeneration. Reducing sugars, which react with protein amino groups to form a diverse group of moieties with fluorescence and cross-linking properties, have been implicated in the structural and functional alterations of proteins that occur during aging and long-term diabetes. This study was undertaken to determine the changes in two matrix cross-links of the intervertebral disc and to study their association with aging. Two types of cross-links were studied: the physiological cross-link, pyridinoline, which is initiated by lysyl oxidase; and the non-enzymatically initiated cross-link, pentosidine. A significant increase in pentosidine, but not pyridinoline, was observed in the intervertebral disc with aging. Radiological, histological and biochemical findings support a hypothesis that subchondral bone responses, marked by increased bone density, contribute to alterations in the intervertebral disc. Cross-link changes in the structural proteins of the disc may contribute to the progressive fibrocartilage degradation typical of intervertebral disc disease as an effect of age.  相似文献

Recent experimental studies suggest a role for estrogens in chondrocyte metabolism and the development and treatment of osteoarthritis. Type I cytoplasmic estradiol receptors were found in articular chondrocytes. The presence of these receptors supports a possible mechanism for previous observations on estrogen suppressive effects on cartilage proteoglycan synthesis and offers a basis for further study on the role of estrogens in the physiology of cartilage and in the pathogenesis and treatment of osteoarthritis.  相似文献

Background

HIV/AIDS and Diabetes Mellitus are the diseases’ known to supress cell mediated immunity and predispose patients for opportunistic infections. Hence, we conducted a study to compare the common opportunistic infections (OIs) between People Living with HIV with DM (PLHIV-DM) and PLHIV without DM (PLHIV).

Methodology

PLHIV with DM and without DM (1:1) were prospectively included in the study from January 2011 to January 2012 at a tertiary care hospital in Mangalore city. Patients were classified as Diabetic if their fasting plasma glucose was ≥ 7.0mmol/l (126mg/dl) or 2–h plasma glucose was ≥11.1mmol/l (200mg/dl). Standard procedures and techniques were followed for diagnosis of OIs as per WHO guidelines. The data was entered and analyzed using Statistical Package for Social Sciences (SPSS) version 11.5.

Findings

The study included 37 PLHIV with DM and 37 PLHIV without DM and both groups were treated with Anti-Retroviral Therapy (ART). The median age was 47 years (IQR: 41-55years) for PLHIV-DM as compared to 40 years (IQR: 35–45.5 years) for PLHIV (p<0.0001). PLHIV-DM had median CD4 counts of 245 (IQR: 148–348) cells/μl compared to 150(IQR: 70–278) cells/μl for PLHIV (p = 0.02). Common OIs included oral candidiasis (49% of PLHIV-DM and 35% of PLHIV); Cryptococcal meningitis (19% of PLHIV-DM and 16% of PLHIV); Pneumocystis jiroveci pneumonia (5% of PLHIV-DM and 18% of PLHIV); extra pulmonary tuberculosis (22% of PLHIV-DM and 34.5% of PLHIV); and Cerebral toxoplasmosis (11% of PLHIV–DM and 13.5% of PLHIV). Microbiological testing of samples from PLHIV- DM, C krusei was the most common Candida species isolated from 9 out of 18 samples. Out of six pulmonary TB samples cultured, four grew Non-tuberculosis mycobacteria (NTM) and two Mycobacterium tuberculosis complexes.

Conclusions

Study did not identify any significant difference in profile of opportunistic infections (OIs) between PLHIV with and without Diabetes.  相似文献

Mitochondrial dysfunction and consequent energy depletion are the major causes of oxidative stress resulting to bring alterations in the ionic homeostasis causing loss of cellular integrity. Our previous studies have shown the age-associated interactive effects in rat central nervous system (CNS) upon co-exposure to chlorpyrifos (CPF) and cold stress leading to macromolecular oxidative damage. The present study elucidates a possible mechanism by which CPF and cold stress interaction cause(s) mitochondrial dysfunction in an age-related manner. In this study, the activity levels of Krebs cycle enzymes and electron transport chain (ETC) protein complexes were assessed in the isolated fraction of mitochondria. CPF and cold stress (15 and 20 °C) exposure either individually or in combination decreased the activity level of Krebs cycle enzymes and ETC protein complexes in discrete regions of rat CNS. The findings confirm that cold stress produces significant synergistic effect in CPF intoxicated aging rats. The synergism between CPF and cold stress at 15 °C caused a higher depletion of respiratory enzymes in comparison with CPF and cold stress alone and together at 20 °C indicating the extent of deleterious functional alterations in discrete regions of brain and spinal cord (SC) which may result in neurodegeneration and loss in neuronal metabolic control. Hence, co-exposure of CPF and cold stress is more dangerous than exposure of either alone. Among the discrete regions studied, the cerebellum and medulla oblongata appears to be the most susceptible regions when compared to cortex and SC. Furthermore, the study reveals a gradual decrease in sensitivity to CPF toxicity as the rat matures.  相似文献

ASAP1 is an Arf GAP with a PH domain immediately N-terminal to the catalytic Arf GAP domain. PH domains are thought to regulate enzymes by binding to specific phosphoinositide lipids in membranes, thereby recruiting the enzyme to a site of action. Here, we have examined the functional relationship between the PH and Arf GAP domains. We found that GAP activity requires the cognate PH domain of ASAP1, leading us to hypothesize that the Arf GAP and PH domains directly interact to form the substrate binding site. This hypothesis was supported by the combined results of protection and hydrodynamic studies. We then examined the role of the PH domain in the regulation of Arf GAP activity. The results of saturation kinetics, limited proteolysis, FRET and fluorescence spectrometry support a model in which regulation of the GAP activity of ASAP1 involves a conformational change coincident with recruitment to a membrane surface, and a second conformational change following the specific binding of phosphatidylinositol 4,5-bisphosphate.  相似文献
