Summary Red blood cell esterase D (ESD) polymorphism was studied in a French-Canadian population from Quebec city, Canada, by means of high voltage electrophoresis on agarose gel followed, in heterozygotes for ESD1, by IEF to reveal the possible allele ESD*5. Frequencies of the ESD alleles in 904 unrelated individuals were ESD*1: 0.888, EDS*2: 0.095 and ESD*5: 0.017. Chromosomal anomalies associated with the use of the organic phosphate pesticide guthion were investigated in Chinese hamster cells (line CHO-K1). Most commonly observed were chromatid breaks and exchanges. Infrequently, mild failure of condensation, despiralization, secondary constriction, gaps, pulverization, ring and dicentric chromosomes were noted. The mean number of chromosome breaks per cell was significantly higher in treated cells than in control cells. Autoradiographic studies revealed that while higher dosages of the chemical (80–120 μg/ml) arrested cells and prevented their movement out of S phase, a lower dosage (60 μg/ml) caused a progression delay. In relation to the relative length of no. 1 and no. 2 chromosomes, no apparent difference existed in the incidence of total breaks between them. However, significant differences in the nonrandom distribution of breaks in no. 1 and no. 2 chromosomes indicated linear differentiation of breakage susceptibility along the chromosomes. An increased concentration of breaks was observed in the long arms of both no. 1 and no. 2 chromosomes. The experimental results suggest that guthion, as well as inducing chromosomal anomalies, may produce a viable mutant. 相似文献
An update of the human obesity gene map up to October 1996 is presented. Evidence from Mendelian disorders exhibiting obesity as a clinical feature, single-gene mutation rodent models, quantitative trait loci uncovered in crossbreeding experiments with mouse, rat, and pig models, association and case-control studies with candidate genes, and linkage studies with genes and other markers is reviewed. All chromosomal locations of the animal loci are converted into human genome locations based on syntenic relationships between the genomes. A complete listing of all these loci reveals that only 4 of the 24 human chromosomes are not yet represented, i.e., 9, 18, 21, and Y. Several chromosome arms are characterized by the presence of several putative loci. The following arms include at least three such loci: 1p, 1q, 3p, 4q, 6p, 7q, 8p, 8q, 11p, 11q, 15q, 20q, and Xq. Stand-scale gap-phase dynamics is generally viewed as the main driver of development in mesic deciduous forests of the temperate biome. Soil charcoal of temperate forests in eastern North America are unnoticed in most surveys, thus explaining why fire is undervalued as a driver of forest succession. The extent to which gap-phase, fire, or other processes are responsible for the regeneration and maintenance of mesic deciduous forests is unknown because paleoecological evidence is lacking. We tested the fire-driven succession hypothesis on the development of this major forest type. Based on charcoal 14C dates of two sites, 44 and 55 fires occurred since early Holocene, with a mean interval of 170 to 215 years. The vegetation of both sites followed comparable post-glacial trajectories consisting of three distinct periods. Conifers dominated the two first periods during 5200–6000 years and were replaced by hardwoods–conifers over the last 3500 years. The first period was represented by boreal conifers, whereas the second period, dominated by white pine (Pinus strobus) forests, persisted during 3000–4300 years. The third period marked the development of hardwood (sugar maple, Acer saccharum) forests. Fires occurred continuously on the sites since early Holocene likely under dry conditions during the conifer periods and cooler and moister conditions during the hardwood–conifer period. Recurrent fires appear with climate as key drivers of the long-term dynamics of several temperate forests in eastern North America. Similar studies on other temperate forests should be pursued to test the hypothesis of climate–fire interactions influencing tree composition change.
Low levels of high-density lipoprotein (HDL) cholesterol are associated with an increased risk of acute myocardial infarction possibly through impaired endothelial atheroprotection and decreased nitric oxide (NO) bioavailability. Asymmetric dimethylarginine (ADMA) mediates endothelial function by inhibiting nitric oxide synthase activity. In patients with acute myocardial infarction, we investigated the relationship between serum levels of HDL and ADMA.
Approach and Results
Blood samples from 612 consecutive patients hospitalized for acute MI <24 hours after symptom onset were taken on admission. Serum levels of ADMA, its stereoisomer, symmetric dimethylarginine (SDMA) and L-arginine were determined using high-performance liquid chromatography. Patients with low HDL (<40 mg/dL for men and <50 mg/dL for women) were compared with patients with higher HDL. Most patients (59%) had low HDL levels. Median ADMA levels were markedly higher in the low HDL group (0.69 vs. 0.50 µmole/L, p<0.001). In contrast, SDMA and L-arginine levels were similar for the two groups (p = 0.120 and p = 0.064). Notably, ADMA, but not SDMA or L-arginine, was inversely correlated with HDL (r = −0.311, p<0.001). In stratified analysis, this relationship was only found for low HDL levels (r = −0.265, p<0.001), but not when HDL levels were higher (r = −0.077, p = 0.225). By multivariate logistic regression analysis, ADMA level was strongly associated with low HDL levels (OR(95%CI):6.06(3.48–10.53), p<0.001), beyond traditional confounding factors.
Conclusions
Our large population-based study showed for the first time a strong inverse relationship between HDL and ADMA in myocardial infarction patients, suggesting a functional interaction between HDL and endothelium, beyond metabolic conditions associated with low HDL levels. 相似文献
A majority of plant species has roots that are colonized by both arbuscular mycorrhizal (AM) and non-mycorrhizal (NM) fungi. The latter group may include plant mutualists, commensals, parasites and pathogens. The co-occurrence of these two broad groups may translate into competition for root volume as well as for plant-derived carbon (C). Here we provide evidence that the relative availability of soil nitrogen (N) and phosphorus (P) (i.e., soil nutrient stoichiometry) controls the competitive balance between these two fungal guilds. A decrease in the soil available N:P ratio resulted in a lower abundance of AM fungi and a corresponding increase in NM fungi. However, when the same fertilization treatments were applied in a soil in which AM fungi were absent, lowering the soil available N:P ratio did not affect NM fungal abundance. Taken collectively, our results suggest that the increase in NM fungal abundance was not a direct response to soil nutrient stoichiometry, but rather a competitive release from AM fungi responding negatively to higher soil P.
