Molecular docking and pharmacophore model approaches were used to characterise the binding features of four different series of Rho kinase (ROCK) inhibitors. Docking simulation of 20 inhibitors with ROCK was performed. The binding conformations and binding affinities of these inhibitors were obtained using AutoDock 4.0 software. The predicted binding affinities correlate well with the activities of these inhibitors (R 2 = 0.904). 3D pharmacophore models were generated for ROCK based on highly active inhibitors implemented in Catalyst 4.11 program. The best pharmacophore model consists of one hydrogen bond acceptor feature and two hydrophobic features, and they all seemed to be essential for inhibitors in terms of their binding activities. It is anticipated that the findings reported in this paper may provide very useful information for designing new ROCK inhibitors.  相似文献

CryoEM continues to produce density maps of larger and more complex assemblies with multiple protein components of mixed symmetries. Resolution is not always uniform throughout a cryoEM map, and it can be useful to estimate the resolution in specific molecular components of a large assembly. In this study, we present procedures to 1) estimate the resolution in subcomponents by gold-standard Fourier shell correlation (FSC); 2) validate modeling procedures, particularly at medium resolutions, which can include loop modeling and flexible fitting; and 3) build probabilistic models that combine high-accuracy priors (such as crystallographic structures) with medium-resolution cryoEM densities. As an example, we apply these methods to new cryoEM maps of the mature bacteriophage P22, reconstructed without imposing icosahedral symmetry. Resolution estimates based on gold-standard FSC show the highest resolution in the coat region (7.6 Å), whereas other components are at slightly lower resolutions: portal (9.2 Å), hub (8.5 Å), tailspike (10.9 Å), and needle (10.5 Å). These differences are indicative of inherent structural heterogeneity and/or reconstruction accuracy in different subcomponents of the map. Probabilistic models for these subcomponents provide new insights, to our knowledge, and structural information when taking into account uncertainty given the limitations of the observed density.  相似文献

目的:研究16层螺旋电子计算机断层扫描(CT)三维重建与数字化摄影(DR)平片检查对外伤性肋骨骨折的诊断价值,为临床诊治提供参考。方法:将2017年6月至2018年6月期间于本院接受诊治的82例外伤性肋骨骨折患者作为研究对象,所有患者均接受16层螺旋CT三维重建与DR平片检查,观察并记录患者的骨折发生部位,并比较两种诊断方法对外伤性肋骨骨折诊断的准确率、灵敏度、特异性、阳性预测值、阴性预测值以及漏诊情况。结果:82例外伤性肋骨骨折患者经影像学与临床诊断明确发生骨折179处,多发性骨折发生率为62.20%,单发性骨折发生率为37.80%,骨折肋骨段位中4-10段骨折发生率最高为69.83%,骨折肋骨水平阶段中腋肋骨折发生率最高为59.78%。相较于DR平片,16层螺旋CT三维重建诊断外伤性肋骨骨折的准确率、灵敏度、特异性、阳性预测值、阴性预测值更高,漏诊率更低,差异有统计学意义(P0.05)。结论:相较于DR平片检查,应用16层螺旋CT三维重建检查外伤性肋骨骨折能明显提高临床诊断的准确率、灵敏度和特异性,减少漏诊,可为临床诊治提供更可靠的信息,值得临床推广。  相似文献

【目的】前期发现水稻条纹病毒(rice stripe virus, RSV)可与介体灰飞虱Laodelphax striatellus体内的HiPV病毒(Himetobi P virus, HiPV)互作。本研究旨在制备HiPV外壳蛋白VP1的多克隆抗体,并评估其在HiPV病毒检测中的可用性,以为深入研究HiPV-RSV和HiPV-灰飞虱的互作机制提供技术支持。【方法】以RT-PCR方法从灰飞虱成虫体内扩增HiPV主要外壳蛋白基因VP1,然后将VP1基因亚克隆至原核表达载体pET-32a中,构建表达载体pET-VP1。将重组质粒转化大肠杆菌Escherichia coli BL21 (DE3),经IPTG诱导、Ni2+-NTA亲和层析纯化,获得重组蛋白,免疫新西兰大白兔,制备抗体。【结果】从灰飞虱体内克隆到774 bp的HiPV外壳蛋白基因VP1,经原核表达、纯化,获得分子量约47.5 kD的融合蛋白,免疫新西兰大白兔后获得VP1多克隆抗体。该抗体间接ELISA效价达1∶819 200,与HiPV外壳蛋白VP1有特异性反应,而与灰飞虱蛋白无交叉反应。利用该多克隆抗体建立了检测单头灰飞虱成虫体内HiPV的Western blot和免疫捕获RT-PCR方法,检测结果显示HiPV在携带和不携带RSV的灰飞虱高亲和性群体内均广泛存在。【结论】利用制备的HiPV的VP1多克隆抗体可特异性检测灰飞虱体内HiPV。本研究为HiPV病毒的快速检测以及HiPV-RSV互作、HiPV-灰飞虱互作研究提供了技术支持。  相似文献

为探讨食管癌手术前后患者外周血细胞的凋亡情况。利用流式细胞仪对70例食管癌手术前及其手术后10天~14天患者外周血细胞的DNA和CD4,CD8和CD16/56含量进行了分析。结果显示:手术前后患者外周血细胞的凋亡有差异(P<0.05),但血细胞的DNA合成期细胞比率(S-phage fraction,SPE)、细胞增长指数无明显的差异(P>0.05)。两组的CD4/CD8比值或CD16/56比例有明显差异(P<0.001)。实验数据表明:手术前患者的细胞凋亡增加可能有利于平衡肿瘤细胞的恶性增殖;而外周血细胞的CD8、NK(自然杀伤细胞,natural killer cells)增高,可能有助于这种平衡作用。但手术后患者肿瘤负荷减轻,免疫功能恢复常态,细胞凋亡率相应降低。  相似文献

Increasing terrestrial vegetation activity in China, 1982—1999   总被引:16,自引:0,他引:16  
Variations in vegetation activity during the past 18 years in China were investigated using the normalized difference vegetation index (NDVI) derived from the 3rd generation time series dataset of NOAA-AVHRR from 1982 to 1999. In order to eliminate the effects of non-vegetation factors, we characterized areas with NDVI < 0.1 as “sparsely vegetated areas” and areas with NDVI ≥ 0.1 as “vegetated areas”. The results showed that increasing NDVI trends were evident, to varying extents, in almost all regions in China in the 18 years, indicating that vegetation activity has been rising in recent years in these regions. Compared to the early 1980s, the vegetated area increased by 3.5% by the late 1990s, while the sparsely vegetated area declined by 18.1% in the same period. The national total mean annual NDVI increased by 7.4% during the study period. Extended growing seasons and increased plant growth rates accounted for the bulk of these increases, while increases in temperature and summer rainfall, and strengthening agricultural activity were also likely important factors. NDVI changes in China exhibited relatively large spatial heterogeneity; the eastern coastal regions experienced declining or indiscernibly rising trends, while agricultural regions and western China experienced marked increases. Such a pattern was due primarily to urbanization, agricultural activity, regional climate characteristics, and different vegetation responses to regional climate changes.  相似文献

目的探讨鼻眶脑毛霉菌病(ROCM)的临床与病理特征,提高对该病的认识和病理诊断水平。方法回顾分析首都医科大学附属北京同仁医院1998~2008年16例ROCM患者的病历资料。用HE、PAS和GMS染色显示组织病变特点及真菌的形态特征,对1例石蜡组织行透射电镜观察。结果 14例(87.5%)有基础疾病,2例(12.5%)无特殊病史。死亡5例(31.3%),3例死于ROCM,2例死于原发病。CT示鼻腔鼻窦软组织密度影16例,MRI示眶内(15例)和颅内(5例)异常信号影。真菌培养7例阳性(43.8%)。组织病理:16例均见组织凝固性坏死、真菌性血管炎及肉芽肿,骨质破坏9例,外周神经纤维坏死7例。特殊染色菌丝形态均符合毛霉目真菌;透射电镜见菌丝形态不规则,菌壁电子密度高。结论 ROCM主要见于糖尿病和血液系统恶性肿瘤者,亦见于无基础疾病者。常见致病菌为根霉和毛霉。在病变组织中找到符合毛霉目真菌形态特点的菌丝可确诊。对临床症状、影像学、真菌培养及病理学表现等多方面进行综合分析可明确诊断。  相似文献

采用分离群体分组分析法(BSA),用100个随机引物对满天星的正常苗和玻璃化苗进行RAPD分析的结果表明,7个随机引物扩增出多态性差异条带。再用上述7个引物分别对试管苗及其玻璃化苗个体进行DNA的PCR扩增的结果显示,引物J20在2种苗中出现差异条带。  相似文献
