Diatoms, an important group of phytoplankton, bloom annually in the Southern Ocean, covering thousands of square kilometers and dominating the region''s phytoplankton communities. In their role as the major food source to marine grazers, diatoms supply carbon, nutrients and energy to the Southern Ocean food web. Prevailing environmental conditions influence diatom phenotypic traits (for example, photophysiology, macromolecular composition and morphology), which in turn affect the transfer of energy, carbon and nutrients to grazers and higher trophic levels, as well as oceanic biogeochemical cycles. The paucity of phenotypic data on Southern Ocean phytoplankton limits our understanding of the ecosystem and how it may respond to future environmental change. Here we used a novel approach to create a ‘snapshot'' of cell phenotype. Using mass spectrometry, we measured nitrogen (a proxy for protein), total carbon and carbon-13 enrichment (carbon productivity), then used this data to build spectroscopy-based predictive models. The models were used to provide phenotypic data for samples from a third sample set. Importantly, this approach enabled the first ever rate determination of carbon productivity from a single time point, circumventing the need for time-series measurements. This study showed that Chaetoceros simplex was less productive and had lower protein and carbon content during short-term periods of high salinity. Applying this new phenomics approach to natural phytoplankton samples could provide valuable insight into understanding phytoplankton productivity and function in the marine system.  相似文献

Transgender women are particularly at risk of HIV infection, but little evidence exists on effective HIV prevention strategies with this population. We evaluated whether Sisters, a peer-led program for transgender women, could reduce HIV risks in Pattaya, Thailand. The study used time-location sampling to recruit 308 transgender women in Pattaya into a behavioral survey in 2011. Coarsened exact matching was used to create statistically equivalent groups of program participants and non-participants, based on factors influencing likelihood of program participation. Using multivariable logistic regression, we estimated effects of any program participation and participation by delivery channel on: condom use at last sex; consistent condom and condom/water-based lubricant use in the past 3 months with commercial, casual, and regular partners; and receipt of HIV testing in the past 6 months. Program coverage reached 75% of the population. In a matched sub-sample (n = 238), participation in outreach was associated with consistent condom/water-based lubricant use with commercial partners (AOR 3.22, 95% CI 1.64–6.31). Attendance at the Sisters drop-in center was associated with receiving an HIV test (AOR 2.58, 95% CI 1.47–4.52). Dedicated transgender-friendly programs are effective at reducing HIV risks and require expansion to better serve this key population and improve HIV prevention strategies.  相似文献

The ultrastructure of saprophytic and parasitic spores of the AscomyceteClaviceps purpurea (Fr.) Tul. was studied. Considerable differences were found to exist between the saprophytic and parasitic spores as to morphology and fine structure. The reason for the different ultrastructural morphology is probably connected with the intensity of cell metabolism. Whereas the parasitic spores obtained from the honeydew possess the character of a resting cell with a thick electron-dense cytoplasm, abundant lipid bodies, few mitochondria, an underdeveloped and hence little active endoplasmic reticulum and with a homogenous thick cell wall, the saprophytic spores appear as cells with higher metabolic rate, containing more numerous mitochondria, a thinner cytoplasm, a highly developed endoplasmic reticulum, fewer lipid bodies and abundant large vacuoles as well as frequently a new wall layer.  相似文献

Comparative CD and Fourier transform ir (FTIR) spectroscopic data on N-Boc protected linear peptides with or without the (Pro-Gly) β-turn motif (e.g., Boc-Tyr-Pro-Gly-Phe-Leu-OH and Boc-Tyr-Gly-Pro-Phe-Leu-OH) are reported herein. The CD spectra, reflecting both backbone and aromatic contributions, were not found to be characteristic of the presence of β-turns. In the amide I region of the FTIR spectra, analyzed by self-deconvolution and curve-fitting methods, the β-turn band shewed up between 1639 and 1633 cm?1 in trifluoroethanol (TFE) but only for models containing the (Pro-Gly) core. This band war-also present in the spectra in chloroform but absent in dimethylsulfoxide. These findings, in agreement with recent ir data on cyclic models and 310-helical polypeptides and protein in D2O [see S. J. Prestrelski, D. M. Byler, and M. P. Thompson (1991), International Journal of Peptide and Protein Research, Vol. 37, pp. 508–512; H. H. Mantsch, A. Perczel. M. Hollósi, and G. D. Fasman (1992), FASEB Journal, Vol. 6, p. A341; H. H. Mantsch. A. Perczel, M. Hollósi, and G. Fasman (1992), Biopolymers. Vol. 33, pp. 201–207; S. M. Miick, G. V. Martinez, W. R. Fiori, A. P. Tedd, and G. L. Millhauser (1992). Nature, Vol. 359, pp. 653–655], suggest that the amide I band, with a major contribution from the acceptor C ? O of the 1 ← 4 intramolecular H bond of β-turns, appears near or below 1640 cm?1, rather than above 1660 cm?1. In TFE, bands between 1670 and 1660 cm?1 are mainly due to “free” carbonyls, that is, C ? Summary The composition of the ant community was assessed along standardized 100 m transects in annually flooded Varzea forest and in terra firme forests on sandy soil (Flanco forest) and on claytopped mesas (Planalto forest). Standardized samples were taken by unit-time hand collecting (day and night times), sweeping, beating, baiting and by Winkler sacks. A total of 156 species, representing 49 genera were found, of which 98, 88 and 55 were respectively found in the Planalto, Flanco and Varzea forests. Species lists are presented and the ant community composition and species richness are compared between the three forests. By considering the nesting and foraging habits of the various species, the differences in overall community composition are related to the forest type and susceptibility to inundation of the three forests which were surveyed.The data confirm the view that tropical rain forests support an extremely diverse ant fauna and comparisons with other forested areas suggest that ant species richness declines in subtropical and temperate rain forests. Although alpha diversity is high, species turnover between forests is lower than expected, suggesting that ant species richness in this forested region is not as great as is implied in some published estimates of global arthropod diversity.  相似文献

A study of spider (Araneae) communities was conducted in rehabilitated bauxite mines at the Jarrahdale mine site of Alcoa of Australia Ltd. and in the nearby native jarrah (Eucalyptus marginata) forest in southwest Western Australia. The study was conducted from March to August 1993 in five rehabilitated sites of different age and method of rehabilitation and in two forest sites. A variety of collection methods was used, including pitfall trapping, litter sampling, sweep netting, tree beating, and visual searching. These methods were the same as those carried out in a previous study of some of these areas in 1983. We collected 151 spider species belonging to 102 genera and 34 families. We examined the relationship between various habitat features, including the age and method of rehabilitation, of the spider communities present. It was found that leaf litter depth and cover and vegetation density had a significant positive influence on recolonization by the various spider guilds. The age and method of rehabilitation were found to influence different vegetational and habitat features; these, in turn, influenced the spider communities. Thus, the older a rehabilitated site the greater the species richness of both plants and spiders. We compared these results with those of the 1983 study to determine the spider succession of the aging rehabilitation. The spider communities and guild composition were found to change as the vegetation matured, from a dominance of pioneer species to a community of species requiring less harsh conditions. By comparison with the pre-1983 rehabilitation, the latest method of rehabilitation increased the rate of recolonization by both plants and spiders.  相似文献

Rhizopuspepsin and other fungal aspartic proteinases are distinct from the mammalian enzymes in that they are able to cleave substrates with lysine in the P1 position. Sequence and structural comparisons suggest that two aspartic acid residues, Asp 30 and Asp 77 (pig pepsin numbering), may be responsible for generating this unique specificity. Asp 30 and Asp 77 were changed to the corresponding residues in porcine pepsin, Ile 30 and Thr 77, to create single and double mutants. The zymogen forms of the wild-type and mutant enzymes were overexpressed in Escherichia coli as inclusion bodies. Following solubilization, denaturation, refolding, activation, and purification to homogeneity, structural and kinetic comparisons were made. The mutant enzymes exhibited a high degree of structural similarity to the wild-type recombinant protein and a native isozyme. The catalytic activities of the recombinant proteins were analyzed with chromogenic substrates containing lysine in the P1, P2, or P3 positions. Mutation of Asp 77 resulted in a loss of 7 kcal mol-1 of transition-state stabilization energy in the hydrolysis of the substrate containing lysine in P1. An inhibitor containing the positively charged P1-lysine side chain inhibited only the enzymes containing Asp 77. Inhibition of the Asp 77 mutants of rhizopuspepsin and several mammalian enzymes was restored upon acetylation of the lysine side chain. These results suggest that an exploitation of the specific electrostatic interaction of Asp 77 in the active site of fungal enzymes may lead to the design of compounds that preferentially inhibit a variety of related Candida proteinases in immunocompromised patients.  相似文献

Transmission and scanning electron microscopy of the lateral ventricles of the toad brain revealed the presence of supraependymal cells that have the features of macrophages. Based solely on their surface morphology three different cell forms could be identified. The most frequently observed cells are flat and multipolar, and have a smooth or ruffled surface. The second type is spherical with a ruffled surface and occurs either singly, in which case it lacks processes, or in clusters from which processes radiate. The third type has surface blebs and numerous thin, smooth processes. However, when specimens that had been examined in the scanning electron microscope are viewed in the transmission electron microscope, all cells appear to belong to a single cell type. All cells viewed closely resemble macrophages in that they contain nuclei with clumped chromaffin, single cisternae of rough endoplasmic reticulum, numerous dense bodies, and many Golgi complexes. In addition, when horseradish peroxidase (HRP) was perfused into the ventricles, reaction product was found a short time thereafter within cytoplasmic vacuoles, and after a longer period within dense bodies. Because of their ultrastructural resemblance to macrophages and their capacity to ingest HRP, we suggest that these cells function as phagocytes and, as such, act to remove foreign materials from the cerbrospinal fluid.  相似文献
