SYNOPSIS In a culture system of Paramecium bursaria , virus particles were found in large number. The particle was able to infect and multiply in certain cells of the zoochlorella, an intracellular symbiotic alga of P. bursaria. The infective particle, designated as zoochlorella cell virus (ZCV), was icosahedral and 120–180 nm in edge to edge diameter. The ZCV particle was found to differ from any of the already established viruses attacking the green and the blue-green algae. Within the system where P. bursaria cells were growing, ZCV particles were detected in the depression of the pellicle, among the cilia growing in the cytopharynx, and in the food vacuole of P. bursaria. ZCV particles were infective only for the zoochlorella cells which were recently released from the cytoplasm of P. bursaria. The multiplication process of ZCV comprised the adsorption of the particle to the cell wall of the zoochlorella, the penetration of nucleic acid into the host cell interior, the replication of viral constituents, the maturation of viral particles and their final release by the burst of the zoochlorella cell. ZCV particles appeared only in the cytoplasmic region of the zoochlorella cell in which many ribosomes were distributed. A possible ecosystem among the 3 members consisting of P. bursaria , zoochlorella and ZCV is discussed.  相似文献

1. After it enters streams, terrestrially derived organic matter (OM) rapidly absorbs water. Using field and laboratory experiments, we examined how this process affected the buoyancy, settling velocity, transport distance and retention locations of four types of organic matter typically found in Pacific coastal streams (‘flexible’ red alder leaves and three ‘stiff’ particle types – Douglas‐fir needles, red cedar fronds and Douglas‐fir branch pieces). 2. Immersion in water rapidly changed the physical characteristics of alder leaves, Douglas‐fir needles and red cedar fronds, which all reached constant still‐water settling velocities after only a few days of soaking. In contrast, the settling velocity of branch pieces continued to increase for 13 days, eventually reaching much higher values than any other OM type. Dried alder leaves became negatively buoyant after only two days of immersion, while other types took substantially longer (up to 24 days) before the specific gravity of all particles was >1. 3. We released saturated OM particles in an experimental channel and found that all particle types travelled further in a fast, shallow ‘riffle’ than a slow, deep ‘pool’. Comparisons with a passive settlement null model indicated that leaves were retained more rapidly than expected in the riffle (by large protruding stones), while the three stiff particle types travelled further than expected (probably due to turbulent suspension) and were retained when they settled in deeper water between larger stones. In pools, passive settlement appeared to dominate the retention of all OM types, with leaves travelling furthest. 4. These retention patterns corresponded well with those observed when saturated OM particles collected in the field were released in two pools and two riffles in a second‐order coastal stream. 5. When the experimental channel and in‐stream data were combined, the retention rates of the three stiff OM types were closely related to calculated Rouse numbers (Rouse number = particle settling velocity/shear velocity), whereas the retention rate of alder leaves was not. This suggests that different physical mechanisms are responsible for the retention of leaves and stiff OM types in shallow streams.  相似文献

We directly evaluated the role of extrafloral nectaries (EFN) in ant attraction and herbivore exclusion by experimental removal of EFN in the laboratory. When EFN of Vicia faba Linnaeus (Leguminosae) were artificially removed, the number of workers of Tetramorium tsushimae Emery (Hymenoptera: Formicidae) visiting the plant decreased, and the efficiency of herbivore exclusion by ants also decreased. Herbivore exclusion by ants was mostly ineffective on a plant when less than four workers visited the plant, but when more than four workers visited, the time a herbivore resided on the plant decreased rapidly with increasing numbers of visiting ants. Therefore, the efficiency of herbivore exclusion from a plant is determined by the number of ants visiting, and EFN play an important role in ant attraction.  相似文献

  1. Formyltetrahydrofolate synthetase (E. C. 6. 3. 4. 3) was foundto be widely distributed in higher plants and the high enzymeactivity was observed in green leaves of Brassica and Alliumspecies, spinach, and in pea seedlings. In pea seedlings, theenzyme activity changed during the course of germination, andmost of the enzyme activity was located in a soluble fractionof the cytoplasm.
  2. The enzyme was labile and lost the activityrapidly, even whenstored at 5 in the presence of 0.1 M mercaptoethanol.It was,however, found that ammonium sulfate was very effectivein stabilizingthe enzyme activity.
  3. The enzyme has been purifiedapproximately 500-fold from extractsof pea seedlings by treatmentswith ammonium sulfate, protaminesulfate, hydroxylapatite, calciumphosphate gel, and DEAE-cellulosecolumn chromatography.
  4. Thepurified enzyme was specific for formate, ATP and FAH4,andthe Michaelis constants for these reactants were 2.1 10–2M, 5.1 10–4 M, and 5.6 10–3 M, respectively.
  5. The optimum pH was found to be 8.0, and the optimal temperaturewas observed at 37. Both NH4$ and a divalent cation (MgSS orMnSS) were required for the optimal activity.

Summary 1. To examine spatial heterogeneity of trophic pathways on a small scale (<5 m diameter), we conducted dual stable isotope (δ13C and δ15N) analyses of invertebrate communities and their potential food sources in three patchy habitats [sphagnum lawn (SL), vascular‐plant carpet (VC) and sphagnum carpet] within a temperate bog (Mizorogaike Pond, Kyoto, Japan). 2. In total, 19 invertebrate taxa were collected from the three habitats, most of which were stenotopic, i.e. collected from a single habitat. Amongst the habitats, significant variation was observed in the isotopic signatures of dominant plant tissues and their detrital matter [benthic particulate organic matter (BPOM)], both of which were potential organic food sources for invertebrates. Site‐specific isotopic variation amongst detritivores was found in δ13C but not in δ15N, reflecting site‐specificity in the isotopic signatures of basal foods. The eurytopic hydrophilid beetle Helochares striatus was found in all habitats, but showed clear site variation in its isotopic signatures, suggesting that it strongly relies on foods within its own habitat. 3. The most promising potential foods for detritivores were the dead leaf stalks of a dominant plant in the VC and BPOM in the SL and carpet. An isotopic mixing model (IsoSource version 1.3.1) estimated that aquatic predators rely on unknown trophic sources with higher δ13C than detritus, whereas terrestrial predators forage on allochthonous as well as autochthonous prey, suggesting that the latter predators might play key roles in coupling between habitats. 4. Our stable isotope approach revealed that immobile detritivores are confined to their small patchy habitats but that heterogeneous trophic pathways can be coupled by mobile predators, stressing the importance of habitat heterogeneity and predator coupling in characterising food webs in bog ecosystems.  相似文献

Deceptive orchid species that offer no floral rewards to pollinators often experience pollinator limitation because they. The breeding system and fruit set were investigated in order to examine pollinator limitation for a population of a deceptive orchid, Pogonia japonica , on a floating peat mat in Mizorogaike Pond, western Japan. Fruit sets for outcross-pollinated, self-pollinated and pollinator-excluded flowers were 75%, 80%, and 0%, respectively. Thus, this species was self-compatible but neither autogamous nor apogamous. The fruit set for open-pollinated flowers was 22.9% and 17.4% in 1994 and 1995, suggesting that pollinator limitation did occur in the field. Larger flowers were more likely to develop into fruit than smaller ones in open pollination (36% vs 8%). Thus, flower size seems to affect fruiting of this species. No effective visits by major pollinators such as bees, which could carry pollinia, were observed during 16.5 h of observation during daytime. Instead, thrips were often found moving on gynostemium or pollinia and circumstantial evidence that suggests thrips may partly transfer pollen in pieces out of a granular pollinium of P. japonica was obtained. Pogonia japonica should come to use thrips as a pollinator to supplement low fruit set under limited flower visitation by major pollinators.  相似文献

We previously proposed that an enzymatic cooperation between Kupffer cells and hepatocytes may play an important role in cysteinyl leukotriene (LT) production in rat liver. Anin vitrotranscellular synthesis cysteinyl LTs by a Kupffer cell—hepatocyte coculture system was characterized here. Kupffer cells alone, with A23187 stimulation, did not generate cysteinyl LTs until supplemented either with isolated hepatocytes or with LTC4synthase and glutathione, indicating that Kupffer cells can synthesize LTA4but not convert it into LTC4. In contrast, hepatocytes converted the LTA4into cysteinyl LTs and further degraded the cysteinyl LTs. Cysteinyl LT production by the Kupffer cell—hapatocyte coculture system was optimized by addition of 1–3% serum albumin to the culture and by bringing the cell—cell distance closer to less than 3μ. Tumour necrosis factor also stimulated cysteinyl LT production by the coculture system. From these results, it is expected that the Kupffer cell—hepatocyte transcellular system for cysteinyl LT production actually functionsin vivo.  相似文献

Dihydrofolate reductase (E.C. 1.5.1.3 [EC] ) was found in pea seedlingsand was partially purified by treatments with ammonium sulfate,protamine sulfate and by DEAE-cellulose column chromatography.Some properties of the enzyme were investigated. Optimum pHfor the reaction was 6.5. In the enzyme reaction, FAH2 and NADPH2were specifically required. MICHAELIS constants for FAH2 andNADPH2 were 4.3x10–6 M and 4.0x10–5 M, respectively.Folate antagonists such as aminopterin, methotrexate and pyrimethaminewere potent inhibitors of this enzyme. Enzyme activity was almostcompletely inhibited at a concentration of 10–7 M of aminopterinand methotrexate and 10–6 M of pyrimethamine. Growth of germinating pea seeds was inhibited by aminopterin,methotrexate and pyrimethamine, and it recovered significantlywith a tetrahydro-derivative of folate, CF, but not with dihydrofolicor folic acid. These results suggest that growth inhibitionof pea seedlings by these antagonists is due to inhibition ofdihydrofolate reductase in seedlings. 1Studies on the enzymatic synthesis and metabolism of folatecoenzymes in plants IV. An acid α-glucosidase (EC 3.2.1.20) was purified to homogeneity from the culture medium of Tetrahymena thermophila CU 399. Its general molecular, catalytic and immunological properties were compared to those of the T. pyriformis W enzyme. The enzyme from T. thermophila was a 105-kD monomer and the N-terminus (25 amino acid residues) displayed some homology with that of T. pyriformis enzyme. The purified enzyme was most active at 56° C and showed resistance to thermal inactivation. The acid α-glucosidase appears to have α-1,6-glucosidase as well as α-1,4-glucosidase activity. The Km values determined with p-nitrophenyl-α-glucopyranoside, maltose, isomaltose and glycogen were 0.7 mM, 2.5 mM, 28.5 mM and 18.5 mg/ml, respectively. The enzyme was antigenically distinct from T. pyriformis acid α-glucosidase.  相似文献
