The 'Opinions' will not necessarily represent the views of the Society for Applied Bacteriology or of the Editors. The Editors may invite or readers may submit 'Responses' to published 'Opinions' , provided that they are not merely polemics, and these will also be published at the discretion of the Editors. 'Responses' will be treated precisely like 'Opinions'. They should clearly indicate, in their first sentence, the 'Opinion' to which they are a response.     

Quality assurance of selective culture media for bacteria, moulds and yeasts: an attempt at standardization at the international level

To facilitate monitoring of culture media, a simple quantitative streaking technique, implying ever-decreasing numbers of colony-forming units per surface area, as in spiral plating, was developed. The procedure evaluates, in quantitative terms, the ability of media (1) to support the formation of colonies by organisms that it was designed to grow and (2) to resist colonization by organisms that it is expected to suppress. The procedure was therefore termed ecometric evaluation. The ecometric results appeared to agree well with observations made on productivity and selectivity of the media studied during routine examination of specimens. These encouraging results prompted further, rigorous standardization of ecometry. A template was developed to standardize inoculation and the depth of the agar was controlled to within +/- 10%. Finally the attributes of the inocula used were accurately defined. The standardized ecometric technique has been found useful for the following purposes: (1) to assess the practical significance of the inhibitory effect of gentamicin observed in some moulds and yeasts (this was solved by replacing poorer basal media by one particular richer modification, viz, yeast morphology agar); (2) the development of a blood-free selective enumeration medium for Campylobacter jejuni, i.e. sulphide iron motility agar plus the combination of antibiotics suggested by Skirrow (1977); and (3) verification of the absence of antimicrobial activity of enzyme preparations, e.g. catalase, used in culture media to remedy sublethal damage in certain groups of bacteria. Ecometric evaluation can now be recommended for (1) routine monitoring of consignments of dehydrated or ready-to-use, purchased media; and (2) in-house checking of the functioning of medium preparation departments. Only occasionally is it necessary to use conventional counting techniques to confirm the results.     

Cα-Methyl phenylglycine-based semi-synthetic ampicillin and cephalexin analogues

Summary Reaction of the N-carboxyanhydride from C^α-methyld-phenylglycine with either 6-aminopenicillanic acid or 7-amino-3-desacetoxy-cephalosporanic acid furnishes the corresponding ampicillin and cephalexin analogues. Neither the biological activity nor the chemical stability of these new semi-synthetic antibiotics are superior to those of their unmethylated counterparts.     

Severe acute respiratory syndrome coronavirus 3a protein is a viral structural protein

The present study showed the association of a severe acute respiratory syndrome coronavirus (SCoV) accessory protein, 3a, with plasma membrane and intracellular SCoV particles in infected cells. 3a protein appeared to undergo posttranslational modifications in infected cells and was incorporated into SCoV particles, establishing that 3a protein was a SCoV structural protein.     

Molecular Determinants of Mouse Neurovirulence and Mosquito Infection for Western Equine Encephalitis Virus

Western equine encephalitis virus (WEEV) is a naturally occurring recombinant virus derived from ancestral Sindbis and Eastern equine encephalitis viruses. We previously showed that infection by WEEV isolates McMillan (McM) and IMP-181 (IMP) results in high (∼90–100%) and low (0%) mortality, respectively, in outbred CD-1 mice when virus is delivered by either subcutaneous or aerosol routes. However, relatively little is known about specific virulence determinants of WEEV. We previously observed that IMP infected Culex tarsalis mosquitoes at a high rate (app. 80%) following ingestion of an infected bloodmeal but these mosquitoes were infected by McM at a much lower rate (10%). To understand the viral role in these phenotypic differences, we characterized the pathogenic phenotypes of McM/IMP chimeras. Chimeras encoding the E2 of McM on an IMP backbone (or the reciprocal) had the most significant effect on infection phenotypes in mice or mosquitoes. Furthermore, exchanging the arginine, present on IMP E2 glycoprotein at position 214, for the glutamine present at the same position on McM, ablated mouse mortality. Curiously, the reciprocal exchange did not confer mouse virulence to the IMP virus. Mosquito infectivity was also determined and significantly, one of the important loci was the same as the mouse virulence determinant identified above. Replacing either IMP E2 amino acid 181 or 214 with the corresponding McM amino acid lowered mosquito infection rates to McM-like levels. As with the mouse neurovirulence, reciprocal exchange of amino acids did not confer mosquito infectivity. The identification of WEEV E2 amino acid 214 as necessary for both IMP mosquito infectivity and McM mouse virulence indicates that they are mutually exclusive phenotypes and suggests an explanation for the lack of human or equine WEE cases even in the presence of active transmission.

Rapid Detection of Sublethally Impaired Cells of Enterobacteriaceae in Dried Foods

Samples of dried foods, feeds, and drugs were examined for Enterobacteriaceae by using buffered glucose-Brilliant Green-bile broth as the final enrichment medium and deep tubes of violet-red-bile-glucose-agar for confirmation. The pre-enrichment treatments used for the resuscitation of sublethally impaired cells were overnight incubation in lactose broth versus incubation for 1 to 6 hr at room temperature in shallow layers of tryptone soya peptone broth. The latter restoration treatment appeared to be significantly more productive.
