Lactobacillus plantarum 24, isolated from marula fruit grows at pH 4.0 and tolerates acid levels and bile concentrations normally present in the human gastro-intestinal tract. Wistar rats that have been administered L. plantarum 24 showed no signs of discomfort or abnormal behavior. Tissue samples from the liver, spleen and intestine appeared normal. Furthermore, strain 24 harbors the genes encoding plantaricins A, F, and NC8α, a gene encoding immunity to plantaricin, and an ABC transporter similar in sequence to that reported for plantaricin G. At least one antimicrobial peptide within the size range of plantaricins A, F, and NC8α has been detected on a tricine-SDS–PAGE gel. Little is known about the microbial population in marula. This is the first report of a L. plantarum strain from marula fruit with bacteriocin genes and probiotic properties.  相似文献

Fourteen N-acetylated and non-acetylated 3,4,5-tri- or 2,5-dimethoxypyrazoline analogs of combretastatin-A4 (1) were synthesized. A non-acetylated derivative (5a) with the same substituents as CA-4 (1) was the most active compound in the series, with IC(50) values of 2.1 and 0.5 microM in B16 and L1210 cell lines, respectively. In contrast, a similar compound with an acetyl group at N1 of the pyrazoline ring (6g) showed poor activity in the cell lines studied. A cell-based assay indicated that compound 5a caused extensive microtubule depolymerization with an EC(50) value of 7.1 microM in A-10 cells while no activity was seen with the acetylated compound. Molecular modeling studies showed that these compounds possess a twisted conformation similar to CA-4 (1).  相似文献

Lactobacillus reuteri Lr1, isolated from healthy horses, remained viable after 2 h at pH 2.0 and in the presence of 1.5 % (w/v) bile. Strain Lr1 survived passage through the equine gastro-intestinal tract (GIT). However, no viable cells of L. reuteri Lr1 were detected on the third day after administration, suggesting that the strain did not colonise the GIT for longer than two days. Strain Lr1 adhered to non-viable, but not to viable, buccal epithelial cells in vitro. Adherence of strain Lr1 to buccal epithelial cells increased 25 % after treatment of the bacterial cells with pepsin. Treatment with pronase prevented the adhesion to epithelial cells. This suggested that specific proteins on the cell surface of L. reuteri Lr1 are involved in adhesion to epithelial cells. Strain Lr1 aggregated with Clostridium difficile C6, isolated from the GIT of a horse that died from severe colic. Adherence of C. difficile C6 to epithelial cells declined from 60 % to 3 % when challenged with L. reuteri Lr1 and the number of viable clostridia decreased tenfold during dosage. Red blood cell, haemoglobin and haemocrit levels were significantly (P ≤ 0.05) lower after dosage with L. reuteri Lr1. Cholesterol and glucose levels were mildly elevated for one day during dosage, but decreased significantly thereafter to levels similar than before dosage. Genes encoding adhesion to collagen, production of aggregation substances, cytolysin and β hemolysin III, resistance to vancomycin A, B and C, and gelatinase activity were not detected, suggesting that L. reuteri Lr1 is a potential probiotic that may be used to control C. difficile cell numbers in the GIT.  相似文献

A recently described avian neuropeptide, gonadotropin inhibitory hormone (GnIH), has been shown to have seasonal regulatory effects on the hypothalamic-pituitary-gonadotropin axis (HPG) in several avian species. In the bird, GnIH expression is increased during the photorefractory period and has inhibitory effects on the HPG. A recently described mammalian neuropeptide, RF-amide-related peptide-3 (RFRP-3), may be genetically related and functionally similar to this avian neuropeptide. The purposes of this study were to first see if rat RFRP-3 is expressed in the male rat brain and second to determine if ICV injections of RFRP-3 will have effects on feeding and sex behaviors, as well as hormone release from the anterior pituitary. Results confirm other studies in that immunoreactive cell bodies and fibers are observable in areas of the male rat brain known to control the HPG and feeding and sex behaviors. RFRP-3 fibers are also observed in close proximity to GnRH immunoreactive cell bodies. Behavioral tests indicate that high but not low ICV RFRP-3 (500 vs. 100 ng, respectively) significantly (p<0.05) suppressed all facets of male sex behavior while not having any observable effects on their ability to ambulate. Sex behavior was later exhibited when those same male rats received the ICV vehicle. While suppressing sex behavior, ICV RFRP-3 significantly (p<0.05) increased food intake compared to controls. ICV RFRP-3 also significantly reduced plasma levels of luteinizing hormone but increased growth hormone regardless of the time of day; however, at no time did RFRP-3 alter plasma levels of FSH, thyroid hormone, or cortisol. These results indicate that although RFRP-3 has similar effects on LH as observed with GnIH in avian species, in the rat RFRP-3 has additional roles in regulating feeding and growth.  相似文献

Lactobacillus equigenerosi strain Le1, a natural inhabitant of the equine gastrointestinal tract, survived pH 3.0 and incubation in the presence of 1.5% (wt/vol) bile salts for at least 2 h. Strain Le1 showed 8% cell surface hydrophobicity, 60% auto-aggregation, and 47% coaggregation with Clostridium difficile C6. Only 1% of the cells adhered to viable buccal epithelial cells and invaded the cells within 20 min after contact. Preincubation of strain Le1 in a buffer containing pronase prevented adhesion to viable epithelial cells. Preincubation in a pepsin buffer delayed invasion from 20 min to 1 h. Strain Le1 did not adhere to nonviable epithelial cells. Administration of L. equigenerosi Le1 (1 × 10(9) CFU per 50 kg body weight) to healthy horses did not increase white blood cell numbers. Differential white blood cell counts and aspartate aminotransferase levels remained constant. Glucose, lactate, cholesterol, and urea levels remained constant during administration with L. equigenerosi Le1 but decreased during the week after administration.  相似文献

Natural plankton communities from Masnou, a locality 20 km northof Barcelona (NW Mediterranean coast), were enclosed in 30 lmicrocosms to test the effect of different availability of nitrogen(N) and phosphorus (P) on the biomass of the main microplanktongroups, and the biochemical composition (DNA, protein and chlorophyllconcentration) of microbial communities. Immediately after enclosurein microcosms, three different nutrient enrichments were performed:N-deficient, P-deficient and nutrient-balanced. N and P deficienciesaffected the structure and the biochemical composition of themicrobial communities. Phytoplankton assemblages showed similartemporal patterns under the three nutrient treatments, althoughthe relative contribution of the different groups was notablyaffected. The lowest DNA concentration was measured in the P-deficienttreatment, suggesting that P availability imposes the limitson the DNA levels in the ecosystem. The availability of N inthe P-deficient microcosms allowed relatively high synthesisof chlorophyll and protein until the end of the experiment.Significantly high chlorophyll: DNA and protein: DNA ratioscharacterized the P-deficient treatment (where N was available)compared to the N-deficient microcosms. From the results obtained,we suggest that the protein: DNA ratios may constitute a biochemicalindicator of the P versus N availability in natural ecosystems.  相似文献
