Genetic studies indicate that aging is modulated by a great number of genetic pathways. We have used Drosophila longevity and stress assays to test a multipath intervention strategy. To carry out this strategy, we supplemented the flies with herbal extracts (SC100) that are predicted to modulate the expression of many genes involved in aging and stress resistance, such as mTOR, NOS, NF-KappaB, and VEGF. When flies were housed in large cages with SC100 added, daily mortality rates of both male and female flies were greatly diminished in mid to late life. Surprisingly, SC100 also stabilized midlife mortality rate increases so as to extend the maximum life span substantially beyond the limits previously reported for D. melanogaster. Under these conditions, SC100 also promoted robust resistance to partial starvation stress and to heat stress. Fertility was the same initially in both treated and control flies, but it became significantly higher in treated flies at older ages as the fertility of control flies declined. Mean and maximum life spans of flies in vials at the same test site were also extended by SC100, but the life spans were short in absolute terms. In contrast, at an independent test site where stress was minimized, the flies exhibited much longer mean life spans, but the survival curves became highly rectangular and the effects of SC100 on both mean and maximum life spans declined greatly or were abolished. The data indicate that SC100 is a novel herbal mix with striking effects on enhancing Drosophila stress resistance and life span in some environments, while minimizing mid to late life mortality rates. They also show that the environment and other factors can have transformative effects on both the length and distribution of survivorship, and on the ability of SC100 to extend the life span.  相似文献

Food is one of the ultimate complex mixtures to which man is exposed and which cannot easily be dispensed with. Apart from certain well studied microcomponents for example, food pyrolysates, Sugimura 1990 human exposure to genotoxic agents arising from macrocomponents has been relatively little studied from the standpoint of DNA damage. The results of epidemiological studies into the relationship between diet and cancer have left many researchers with the impression that it is an intrinsically intractable problem which is perhaps best left well alone. However, given the popular conception that the normal human diet is safe and that such risks as there may be are due to contamination by pesticide and other chemical residues, there is clearly a need to evaluate the possible avenues open to investigators and which are likely to yield meaningful results which would enable scientifically based advice to be given to the public as to the best dietary habits. This overview of the current state of methodology for measurement of DNA damage in relation to diet as well as a summary of current MAFF supported work and future prospects in this area arose out of a workshop entitled DNA modification in carcinogen risk assessment held in London on November 18, 1996 . The object of this report is to summarize the results presented at the workshop and also to indicate the significance of the MAFF funded programme within the broader context of recently published studies from the international scientific community. Hence, a comprehensive review of all aspects of diet related DNA damage is beyond the scope of this article. The workshop was organized as part of the MAFF Risk Assessment Research Programme and contributes to an interdepartmental initiative, the Government Research Councils Initiative on Risk Assessment and Toxicology RATSC which aims to bring together regulators and toxicologists to discuss their respective perspectives on current problems in the risk assessment of chemicals. Further aims of RATSC are to identify subjects for future detailed workshops on specific issues and to identify priorities for research into toxic chemical risk assessment. The membership of RATSC is drawn from a wide range of Government Departments and Research Councils and is chaired by Dr David Shannon MAFF Chief Scientist.  相似文献

The mountain systems of the Hindu Kush Himalaya (HKH) are changing rapidly due to climatic change, but an overlooked component is the subnival ecosystem (between the treeline and snow line), characterized by short‐stature plants and seasonal snow. Basic information about subnival vegetation distribution and rates of ecosystem change are not known, yet such information is needed to understand relationships between subnival ecology and water/carbon cycles. We show that HKH subnival ecosystems cover five to 15 times the area of permanent glaciers and snow, highlighting their eco‐hydrological importance. Using satellite data from the Landsat 5, 7 and 8 missions, we measured change in the spatial extent of subnival vegetation from 1993 to 2018. The Landsat surface reflectance‐derived Normalized Difference Vegetation Index product was thresholded at 0.1 to indicate the presence/absence of vegetation. Using this product, the strength and direction of time‐series trends in the green pixel fraction were measured within three regions of interest. We controlled for cloud cover, snow cover and evaluated the impact of sensor radiometric differences between Landsat 7 and Landsat 8. Using Google Earth Engine to expedite data processing tasks, we show that there has been a weakly positive increase in the extent of subnival vegetation since 1993. Strongest and most significant trends were found in the height region of 5,000–5,500 m a.s.l. across the HKH extent: R2 = .302, Kendall's τ = 0.424, p < .05, but this varied regionally, with height, and according to the sensors included in the time series. Positive trends at lower elevations occurred on steeper slopes whilst at higher elevations, flatter areas exhibited stronger trends. We validated our findings using online photographs. Subnival ecological changes have likely impacted HKH carbon and water cycles with impacts on millions of people living downstream, but the strength and direction of impacts of vegetation expansion remain unknown.  相似文献

The pinyon jay (Gymnorhinus cyanocephalus) is a primary seed disperser of pinyon pines (Pinus edulis and P. monophylla). Both the pinyon jay and the pinyon pines are experiencing significant decline. While the pinyon jay is a species of management value and conservation concern, little is known about its fecundity, among-flock dispersal, and population differentiation. We initiated genetic studies in pinyon jays using a hybridization enrichment technique to isolate seven polymorphic microsatellite repeats (AAAG and GATA) from the pinyon jay genome. A locus from the Mexican jay (Aphelocoma ultramarina) that amplifies robustly in pinyon jays is also reported. These eight loci revealed moderate to high diversity in an Arizona population of pinyon jays (4–36 alleles and H O 0.42–0.90). As in other species, tetranucleotide repeats produced easily resolved amplification products.  相似文献

In 1966, William D. Hamilton published a landmark paper in evolutionary biology: "The Moulding of Senescence by Natural Selection." It is now apparent that this article is as important as his better-known 1964 articles on kin selection. Not only did the 1966 article explain aging, it also supplied the basic scaling forces for natural selection over the entire life history. Like the Lorentz transformations of relativistic physics, Hamilton's Forces of Natural Selection provide an overarching framework for understanding the power of natural selection at early ages, the existence of aging, the timing of aging, the cessation of aging, and the timing of the cessation of aging. His twin Forces show that natural selection shapes survival and fecundity in different ways, so their evolution can be somewhat distinct. Hamilton's Forces also define the context in which genetic variation is shaped. The Forces of Natural Selection are readily manipulable using experimental evolution, allowing the deceleration or acceleration of aging, and the shifting of the transition ages between development, aging, and late life. For these reasons, evolutionary research on the demographic features of life history should be referred to as "Hamiltonian."  相似文献

Fisher's theory of the sex ratio may be extended to the social Hymenoptera; this extension must consider the unusual genetic structure of the Hymenoptera. Queens, workers, and laying workers generally have different equilibrial sex ratios of offspring and different equilibrial ratios of investment in offspring of the two sexes; these differences are the consequence of asymmetries in the degrees of relatedness between the queen, a worker, and a laying worker to male and female offspring. The equilibrial ratios of investment for the queen, a worker, and a laying worker are derived by finding the relative expected reproductive successes of genes in males and in reproductive females.  相似文献
