Spatial structure and conformational properties of the Dippu-AST 8 (allatostatin III) neuropeptide have been investigated by the molecular mechanics method. The conformational energy and geometrical parameters corresponding to the low-energy states of the molecule are obtained. A single backbone conformation with a very restricted set of Ser 3, Phe 4 and Leu 9 amino acids positions is observed for Dippu-AST 8 neuropeptide.  相似文献

The relationship between nutrient consumption and chronic disease risk is the focus of a large number of epidemiological studies where food frequency questionnaires (FFQ) and food records are commonly used to assess dietary intake. However, these self-assessment tools are known to involve substantial random error for most nutrients, and probably important systematic error as well. Study subject selection in dietary intervention studies is sometimes conducted in two stages. At the first stage, FFQ-measured dietary intakes are observed and at the second stage another instrument, such as a 4-day food record, is administered only to participants who have fulfilled a prespecified criterion that is based on the baseline FFQ-measured dietary intake (e.g., only those reporting percent energy intake from fat above a prespecified quantity). Performing analysis without adjusting for this truncated sample design and for the measurement error in the nutrient consumption assessments will usually provide biased estimates for the population parameters. In this work we provide a general statistical analysis technique for such data with the classical additive measurement error that corrects for the two sources of bias. The proposed technique is based on multiple imputation for longitudinal data. Results of a simulation study along with a sensitivity analysis are presented, showing the performance of the proposed method under a simple linear regression model.  相似文献

Cell spreading is regulated by signaling from the integrin receptors that activate intracellular signaling pathways to control actin filament regulatory proteins. We developed a hybrid model of whole-cell spreading in which we modeled the integrin signaling network as ordinary differential equations in multiple compartments, and cell spreading as a three-dimensional stochastic model. The computed activity of the signaling network, represented as time-dependent activity levels of the actin filament regulatory proteins, is used to drive the filament dynamics. We analyzed the hybrid model to understand the role of signaling during the isotropic phase of fibroblasts spreading on fibronectin-coated surfaces. Simulations showed that the isotropic phase of spreading depends on integrin signaling to initiate spreading but not to maintain the spreading dynamics. Simulations predicted that signal flow in the absence of Cdc42 or WASP would reduce the spreading rate but would not affect the shape evolution of the spreading cell. These predictions were verified experimentally. Computational analyses showed that the rate of spreading and the evolution of cell shape are largely controlled by the membrane surface load and membrane bending rigidity, and changing information flow through the integrin signaling network has little effect. Overall, the plasma membrane acts as a damper such that only ∼5% of the actin dynamics capability is needed for isotropic spreading. Thus, the biophysical properties of the plasma membrane can condense varying levels of signaling network activities into a single cohesive macroscopic cellular behavior.  相似文献

We discuss recent developments in the modeling of negative autoregulated genetic networks. In particular, we consider the temporal evolution of the population of mRNA and proteins in simple networks using rate equations. In the limit of low copy numbers, fluctuation effects become significant and more adequate modeling is then achieved using the master equation formalism. The analogy between regulatory gene networks and chemical reaction networks on dust grains in the interstellar medium is discussed. The analysis and simulation of complex reaction networks are also considered.  相似文献

Caveolae are invaginated plasma membrane domains of 50–100 nm in diameter involved in many important physiological functions in eukaryotic cells. They are composed of different proteins, including the membrane-embedded caveolins and the peripheric cavins. Caveolin-1 has already been expressed in various expression systems (E. coli, insect cells, Toxoplasma gondii, cell-free system), generating intracellular caveolin-enriched vesicles in E. coli, insect cells and T. gondii. These systems helped to understand the protein insertion within the membrane and its oligomerization. There is still need for fundamental insights into the formation of specific domains on membrane, the deformation of a biological membrane driven by caveolin-1, the organization of a caveolar coat, and the requirement of specific lipids and proteins during the process. The aim of this study was to test whether the heterologously expressed caveolin-1β was able to induce the formation of intracellular vesicles within a Gram+ bacterium, Lactococcus lactis, since it displays a specific lipid composition different from E. coli and appears to emerge as a good alternative to E. coli for efficient overexpression of various membrane proteins. Recombinant bacteria transformed with the plasmid pNZ-HTC coding for the canine isoform of caveolin-1β were shown to produce caveolin-1β, in its functional oligomeric form, at a high expression level unexpected for an eukaryotic membrane protein. Electron microscopy revealed several intracellular vesicles from 30 to 60 nm, a size comparable to E. coli h-caveolae, beneath the plasma membrane of the overexpressing bacteria, showing that caveolin-1β is sufficient to induce membrane vesiculation. Immunolabelling studies showed antibodies on such neo-formed intracellular vesicles, but none on plasma membrane. Density gradient fractionation allowed the correlation between detection of oligomers on Western blot and appearance of vesicles measurable by DLS, showing the requirement of caveolin-1β oligomerization for vesicle formation. Lactococcus lactis cells can heterologously overexpress caveolin-1β, generating caveolin-1β enriched intracellular neo-formed vesicles. These vesicles might be useful for potential co-expression of membrane proteins of pharmaceutical interest for their simplified functional characterization.  相似文献

Evaluation of rice husk (RH) as bulking agent in bioremediation of automobile gas oil (AGO) hydrocarbon polluted agricultural soil using renewal by enhanced natural attenuation (RENA) as control was the subject of the present investigation. The effect of different parameters such as total petroleum hydrocarbon (TPH), dehydrogenase activity (DHA), optical density and pH on bioremediation performance were evaluated. The studied parameters such as microbial dynamics, percentage degradation and DHA were found to be higher in RH-amended system and differed significantly with control at P < 0.05. RH resulted in high removal efficiency of 97.85 ± 0.93% under a two-month incubation period, while RENA had lesser removal efficiency of 53.15 ± 3.81%. Overall hydrocarbon biodegradation proceeded very slowly in the RENA particularly from week 0 to 4. Experimental data perfectly fitted into the first-order kinetic and generated high r2 values (0.945), first-order degradation constant (0.47 day?1), and shorter degradation half-life (1.50 d)—t1/2 = Ln2/K and Ln2 numerically equals to 0.693 and hence written as 0.693/K. Micrococcus luteus and Rhizopus arrhizus were isolated in the present study, which displayed extreme AGO hydrocarbon biodegradative abilities. The use of RH in hydrocarbon-polluted soil significantly increased biodegradation rate and resulted in effective AGO cleanup within 2 months period. Therefore, RH provides an alternative source of bioremediation material in field application for abundant petroleum hydrocarbon soil pollution.  相似文献
