Tobamoviruses represent a well-characterized system used to examine viral infection, whereas Arabidopsis is a choice plant for most genetic experiments. It would be useful to combine both approaches into one experimental system for virus–plant interaction. Most tobamoviruses, however, are not pathogenic in Arabidopsis . Here, we describe infection of Arabidopsis by a recently discovered crucifer-infecting turnip vein clearing tobamovirus (TVCV). Using this system, we determined patterns and kinetics of viral local and systemic movement within Arabidopsis plants. Localization studies showed that the virus infects both vegetative and reproductive plant tissues. However, there may be a transport barrier between the seed coat and the embryo which virions cannot cross, preventing seed transmission of TVCV. The ability to move both locally and systemically in Arabidopsis , causing mild and fast-developing symptoms but allowing survival and fertility of the infected plants, distinguish TVCV infection of Arabidopsis as a model system to study virus–plant interaction. 相似文献

Hydrolysis of purin-6-yl 2-deoxy-1-thio-β-d-arabino-hexopyranoside (2) to 6-mercaptopurine and 2-deoxy-d-glucose is catalyzed by hydronium ion and almond β-d-glucosidase. The dependence of rate on acidity in water and deuterium oxide indicates that 2 and its conjugate acid undergo hydrolysis via a mechanism that involves a partially rate-limiting proton transfer. Although 2 is ≈103 more reactive than 6-purinyl β-d-glucothiopyranoside (1) in dilute aqueous acid, 1 is a better substrate for almond β-d-glucosidase. 相似文献

Stimulation of plant productivity caused by Agaricus fairy rings has been reported, but little is known about the effects of these fungi on soil aggregation and the microbial community structure, particularly the communities that can bind soil particles. We studied three concentric zones of Agaricus lilaceps fairy rings in Eastern Montana that stimulate western wheatgrass (Pascopyrum smithii): outside the ring (OUT), inside the ring (IN), and stimulated zone adjacent to the fungal fruiting bodies (SZ) to determine (1) soil aggregate proportion and stability, (2) the microbial community composition and the N-acetyl-β-d-glucosaminidase activity associated with bulk soil at 0–15 cm depth, (3) the predominant culturable bacterial communities that can bind to soil adhering to wheatgrass roots, and (4) the stimulation of wheatgrass production. In bulk soil, macroaggregates (4.75–2.00 and 2.00–0.25 mm) and aggregate stability increased in SZ compared to IN and OUT. The high ratio of fungal to bacteria (fatty acid methyl ester) and N-acetyl-β-d-glucosaminidase activity in SZ compared to IN and OUT suggest high fungal biomass. A soil sedimentation assay performed on the predominant isolates from root-adhering soil indicated more soil-binding bacteria in SZ than IN and OUT; Pseudomonas fluorescens and Stenotrophomonas maltophilia isolates predominated in SZ, whereas Bacillus spp. isolates predominated in IN and OUT. This study suggests that growth stimulation of wheatgrass in A. lilaceps fairy rings may be attributed to the activity of the fungus by enhancing soil aggregation of bulk soil at 0–15 cm depth and influencing the amount and functionality of specific predominant microbial communities in the wheatgrass root-adhering soil. 相似文献

Effect of various lead (Pb) concentrations on the systemic movement of RNA viruses was examined in tobacco plants. Prior to
inoculation, plants were grown hydroponically for 6 days in Hoagland’s solution supplemented with five concentrations of lead
nitrate [Pb(NO3)2]: 0.0 (control), 10, 15, 50, and 100 μM. Four different RNA viruses with different cell-to-cell movement mechanisms were
used. Two weeks after inoculation lower and upper leaves of each treatment were harvested and examined for the presence of
viral coat protein. In plants inoculated with Tobacco mosaic virus, Potato virus X, and Tobacco etch virus, TEM images and western blot assays confirmed the presence of viral coat proteins in the upper leaves of all lead treatments.
However, in plants inoculated with Turnip vein-clearing virus (TVCV), no signs of viral particles were detected in the upper leaves of plants treated with 10 μM or 15 μM lead nitrate.
In contrast, plants treated with high concentrations of lead nitrate (50 μM or 100 μM) showed viral particles in their upper
leaves. In plants treated with 10 μM or 15 μM lead nitrate, callose accumulation was the same as in control plants. This suggests
that non-toxic concentrations of lead nitrate may trigger the production of putative cellular factors in addition to callose
that interfere with the TVCV systemic movement. In contrast, plants treated with 100 μM lead nitrate showed less callose as
compared to control plants, facilitating the systemic movement of TVCV. 相似文献

Background
Buruli ulcer (BU) is a skin disease caused by Mycobacterium ulcerans. Its exact mode of transmission is not known. Previous studies have identified demographic, socio-economic, health and hygiene as well as environment related risk factors. We investigated whether the same factors pertain in Suhum-Kraboa-Coaltar (SKC) and Akuapem South (AS) Districts in Ghana which previously were not endemic for BU.Methods
We conducted a case control study. A case of BU was defined as any person aged 2 years or more who resided in study area (SKC or AS District) diagnosed according to the WHO clinical case definition for BU and matched with age- (+/−5 years), gender-, and community controls. A structured questionnaire on host, demographic, environmental, and behavioural factors was administered to participants.Results
A total of 113 cases and 113 community controls were interviewed. Multivariate conditional logistic regression analysis identified presence of wetland in the neighborhood (OR = 3.9, 95% CI = 1.9–8.2), insect bites in water/mud (OR = 5.7, 95% CI = 2.5–13.1), use of adhesive when injured (OR = 2.7, 95% CI = 1.1–6.8), and washing in the Densu river (OR = 2.3, 95% CI = 1.1–4.96) as risk factors associated with BU. Rubbing an injured area with alcohol (OR = 0.21, 95% CI = 0.008–0.57) and wearing long sleeves for farming (OR = 0.29, 95% CI = 0.14–0.62) showed protection against BU.Conclusion
This study identified the presence of wetland, insect bites in water, use of adhesive when injured, and washing in the river as risk factors for BU; and covering limbs during farming as well as use of alcohol after insect bites as protective factors against BU in Ghana. Until paths of transmission are unraveled, control strategies in BU endemic areas should focus on these known risk factors. 相似文献6.
The preparation of 2-deoxy-2-amino-N-(5-dimethylamino-1-naphthalene sulfonyl)-glucose (III) designed as a fluorescent competitive inhibitor of hexokinase was achieved after reacting 2-deoxy-2-aminoglucose and 1-dimethylamino-5-naphthalene sulfonyl chloride. (III) showed fluorescence excitation and emission maxima in water at 330 and 507 nm, respectively. (III) was found to competitively inhibit hexokinase and a value of Ki = 3.0 x 10(-3)M was obtained for the system hexokinase B + Mg.ATP + glucose at pH 8.4. 相似文献

The need for new therapies to treat systemic fungal infections continues to rise. Naturally occurring hexapeptide echinocandin B (1) has shown potent antifungal activity via its inhibition of the synthesis of beta-1,3 glucan, a key fungal cell wall component. Although this series of agents has been limited thus far based on their physicochemical characteristics, we have found that the synthesis of analogues bearing an aminoproline residue in the 'northwest' position imparts greatly improved water solubility (> 5 mg/mL). Heavy metals, such as cadmium, have a significant impact on plant physiology. However, their potential effect on plant–pathogen interaction, an important biological process, has not been examined. This study shows that exposure of tobacco plants to non-toxic concentrations of cadmium completely blocked viral disease caused by turnip vein clearing virus. Cadmium-mediated viral protection was due to inhibition of the systemic movement of the virus, i.e. its spread from the inoculated into uninoculated leaves. Exposure of plants to cadmium had no effect on viral replication, assembly and local movement within the inoculated leaf. Analysis of the viral presence in different tissues suggested that cadmium treatment inhibited virus exit from the vascular tissue into uninoculated leaves rather than its entry into the host plant vasculature. Higher, toxic levels of cadmium did not produce this inhibitory effect on viral movement, allowing the systemic spread of the virus and development of the viral disease. These observations suggest that cadmium-induced viral protection requires a relatively healthy, unpoisoned plant in which non-toxic levels of cadmium may trigger the production of cellular factors which interfere with the viral systemic movement. 相似文献

Population changes in Pseudocercosporella herpotrichoides with and without fungicide selection A conidium mixture of W (wheat-type) and R (rye-type) strains of Pseudocercosporella herpotrichoides (W:R = 1:1) was incubated on PD-agar without fungicide or amended with the imidazole prochloraz or the triazole cyproconazole. The W:R ratio was assessed over four generations using the different benomyl sensitivity of the strains as marker. In untreated mixtures, the R strain completely dominated the population already after two generations. Similarly, in cyproconazole-treated mixtures, the R strain increased to 100% after the second generation, whereas in prochloraz-treated mixtures the R strain either followed the same pattern as in untreated mixtures or fluctuated around the initial proportion of 50% over four generations. The W and R strains displayed different fitness properties. The average hyphal growth rate was 8.5 mm/10 d and 4.5 mm/10 d for the W and the R strains, respectively; the sporulation capacity was 80 and 400 conidia/colony, respectively, for the two strains. The different fitness resulted in a strong dominance of the R strains both in vitro and in field populations towards the end of the vegetation period. This shift towards R-dominance occurred about equally fast with or without fungicide selection. 相似文献
