线粒体是为细胞提供能量(ATP)的细胞器,携带着自己的DNA——mtDNA,已有多种灵芝属菌株的线粒体基因组相继报道,但对于种内的线粒体基因组的分析较少,对核相同、线粒体不同的菌株间差异的研究也鲜有报道。本研究对两株灵芝线粒体基因组进行组装注释,根据差异片段构建分子标记进行种间分类。结果显示:两株灵芝线粒体基因组大小分别为49 233bp、61 563bp的闭环结构,含有15个常见蛋白编码基因,rRNA大小亚基基因及26个携带氨基酸的tRNA基因,其差异区段主要为内含子序列、大亚基及基因间区。根据cobcox2基因序列能够进行灵芝种间区分,用于灵芝种间鉴定。本研究还根据灵芝119、灵芝无孢的单核菌株构建同核异质体(TY-119、TY-W),分析线粒体对菌落形态、菌丝生长速度及多糖、三萜成分的影响,结果显示:同核异质体TY-W与TY-119菌落形态上有一定差异,同核异质体TY-W菌丝生长速度为4.77mm/d,是TY-119菌丝生长速度4.50mm/d的1.06倍,同核异质体TY-119菌丝、子实体阶段多糖含量分别为4.45mg/g、12.14mg/g是TY-W菌丝体多糖含量(3.23mg/g)的1.38倍、子实体多糖含量(10.24mg/g)的1.19倍;同核异质体TY-W菌丝、子实体阶段的三萜含量分别为6.82mg/g、11.45mg/g是同核异质体TY-119菌丝体三萜含量(9.26mg/g)的0.74倍,子实体三萜含量(9.10mg/g)的1.26倍。利用高效液相色谱分析同核异质体中灵芝酸含量显示同核异质体TY-W灵芝酸A、灵芝酸E、灵芝酸F含量分别为3.77μg/mL、14.29μg/mL、12.91μg/mL;是TY-119灵芝酸A含量(2.59μg/mL)的1.46倍、灵芝酸E含量(13.65μg/mL)的1.17倍、灵芝酸F含量(12.72μg/mL)的1.06倍。对同核异质体菌丝、子实体阶段多糖、三萜合成通路关键基因(pgmugpglshmgshmgrmvdfpssqs)表达量进行检测,显示两菌株间多数基因具有显著性差异。结果表明线粒体的不同会影响灵芝菌落形态、菌丝生长速度及多糖、三萜的含量,有助于我们进一步研究线粒体基因组。  相似文献

目的:探讨中国近15年腹茧症的流行病学特征和诊疗经验.方法:总结中国1994年1月-2009年1月问的腹茧症文献资料.结果:中国近15年共报道903例腹茧症,男女比例为1:1.35,平均年龄33.14岁,51.7%分布在华东地区,90.5%以不同表现形式的肠梗阻为主要表现,70.5%属于弥漫型腹茧症,40.2%患者无大网膜,手术以包膜切除为主.结论:腹茧症主要分布在华东地区,术前诊断困难,切除包膜和松解粘连是治疗本病的有效方法.  相似文献

Hyperglycemia has a profound effect on gastric motility. However, little is known about the site and mechanism that sense alteration in blood glucose level. The identification of glucose-sensing neurons in the nodose ganglia led us to hypothesize that hyperglycemia acts through vagal afferent pathways to inhibit gastric motility. With the use of a glucose-clamp rat model, we showed that glucose decreased intragastric pressure in a dose-dependent manner. In contrast to intravenous infusion of glucose, intracisternal injection of glucose at 250 and 500 mg/dl had little effect on intragastric pressure. Pretreatment with hexamethonium, as well as truncal vagotomy, abolished the gastric motor responses to hyperglycemia (250 mg/dl), and perivagal and gastroduodenal applications of capsaicin significantly reduced the gastric responses to hyperglycemia. In contrast, hyperglycemia had no effect on the gastric contraction induced by electrical field stimulation or carbachol (10(-5) M). To rule out involvement of serotonergic pathways, we showed that neither granisetron (5-HT(3) antagonist, 0.5 g/kg) nor pharmacological depletion of 5-HT using p-chlorophenylalanine (5-HT synthesis inhibitor) affected gastric relaxation induced by hyperglycemia. Lastly, N(G)-nitro-L-arginine methyl ester (L-NAME) and a VIP antagonist each partially reduced gastric relaxation induced by hyperglycemia and, in combination, completely abolished gastric responses. In conclusion, hyperglycemia inhibits gastric motility through a capsaicin-sensitive vagal afferent pathway originating from the gastroduodenal mucosa. Hyperglycemia stimulates vagal afferents, which, in turn, activate vagal efferent cholinergic pathways synapsing with intragastric nitric oxide- and VIP-containing neurons to mediate gastric relaxation.  相似文献

白色念珠菌的粘附与其表面甘露糖结构有关,为探讨阻断粘附的方法,我们采用体外法测定其对口腔上皮细胞的粘附数量,显示:白色念珠菌粘附感染上皮细胞后,甘露糖可以在一定程度上减少粘附,绿慕安及刀豆素A可以有效地减少粘附。提示甘露糖及绿慕安可能有助于治疗白色念珠菌感染。  相似文献

通过合成的^3H-PPPA2′P5′A2′P5′A(2′-5′P3A3)与人淋巴细胞进行结合反应,结果表明:人淋巴细胞质膜存在着2′-5′P3A3受体。又通过用ATP、UTP与^3H-2′-5′P3A3竞争抑制实验表明,^3H-2′-5′P3A3与淋巴细胞膜受体的结合为特异性结构,结合率受^3H-2′-5′P3A3浓度、pH等因素影响。  相似文献

Sequence analysis is the basis of bioinformatics, while sequence alignment is a fundamental task for sequence analysis. The widely used alignment algorithm, Dynamic Programming, though generating optimal alignment, takes too much time due to its high computation complexity O(N(2)). In order to reduce computation complexity without sacrificing too much accuracy, we have developed a new approach to align two homologous sequences. The new approach presented here, adopting our novel algorithm which combines the methods of probabilistic and combinatorial analysis, reduces the computation complexity to as low as O(N). The computation speed by our program is at least 15 times faster than traditional pairwise alignment algorithms without a loss of much accuracy. We hence named the algorithm Super Pairwise Alignment (SPA). The pairwise alignment execution program based on SPA and the detailed results of the aligned sequences discussed in this article are available upon request.  相似文献

The distribution and characteristics of plastids and mitochondria in the generative and sperm cells of Lilium regale Wils. and L. davidii Duch. were described. In L. regale there were few plastids and abundant mitochondria in the newly formed generative cell. When the generative cell became free in the vegetative cytoplasm, the plastids degenerated completely within the generative cell. It was further proved by DAPI fluorescent technique that there was no organell DNA in the generative cell within the mature pollen grain or the pollen tube. However, distribution of the plastids was strictly polarizable during the division of the micmspore in L. davidii, resulting the lack of plastids in the newly formed generative cell. Data of RFLP analysis comparable between L. davidii, L. longifiorum and their interspecific hybrid have also proved the plastid inheritance in L. davidii to be of uniparental maternal transmission. Although the mitoehondria were observed both in the generative and sperm cells of L. regale and L. davidii but their DNA was decomposed in the male gametophyte stage. Therefore the mitochondda in the sperm cell could not be transmitted into the offspring. The results provided the detail, cytological evidence that organelles in the microgametophyte are incapable of genetic transmission in the two species of Lilium.  相似文献

Nucleotides and nucleosides represent an important and ubiquitous class of molecules that interact with specific receptors, regulate a variety of activities within the liver, and play a role in the pathogenesis of hepatic fibrosis. Ecto-nucleotide pyrophosphatase/phosphodiesterases (E-NPPs) are ecto-enzymes that are located on the cell surface. NPP1, NPP2, and NPP3 (abbreviated as NPP1–3 hereafter) have been implicated in the hydrolysis of nucleotides; together with other ecto-nucleotidases, they control the events induced by extracellular nucleotides. We have identified and compared the expression of E-NPP family members in two different phenotypes of the mouse hepatic stellate cell line (GRX). In quiescent-like hepatic stellate cells (HSCs), E-NPP activity was significantly higher, NPP2 mRNA expression decreased and NPP3 mRNA increased. The differential NPP activity and expression in two phenotypes of GRX cells suggests that they are involved in the regulation of extracellular nucleotide metabolism in HSCs. However, the role of E-NPPs in the liver remains to be clarified.  相似文献

 银杉(Cathaya argyrophylla)是一种极其珍稀的濒危松科植物。该研究建立了一种有效的银杉人工菌根诱导技术。通过将银杉种子播种于琼脂培养基7 d后,切除1/5~1/3的 种皮以及胚乳,从而使银杉种子的发芽率达到60%以上;同时筛选出既适合外生菌根菌土生空团菌(Cenococcum geophilum)平铺扩展生长的要求,又能满足银杉幼苗生长的培养基 RM, 使银杉的侧根能最大程度地与菌丝接触,确保了菌根的诱导率;通过紫外线照射加70%酒精 或0.05%NaOCl的表面消毒的双重处理,使不能高压灭菌的容器达到表面消毒效果,同时验证了用该灭菌法既能达到完全的灭菌效果又不会对植物的生长产生不良影响,而且银杉小苗的菌根形成率达到87.5%。该方法对于银杉的菌根学研究以及其它基础研究具有实际应用价值  相似文献
