Methanobacterium thermoautotrophicum, an archaebacterium, possesses the first and last enzymes of the diaminopimelic acid pathway for lysine biosynthesis, dihydrodipicolinate synthase, and diaminopimelate decarboxylase. It does not have saccharopine dehydrogenase, the last enzyme of the aminoadipate pathway for lysine biosynthesis. The dihydrodipicolinate synthase is inhibited but not repressed by lysine. We conclude that this microbe uses the diaminopimelate pathway for synthesis of lysine.Deceased.  相似文献

The in vitro activity of several new imidazoles, cloconazole, sulconazole, butoconazole, isoconazole and fenticonazole, were compared with those of amphothericin B, flucytosine, and three azoles: econazole, miconazole and ketoconazole against isolates of pathogenic Candida. A total of 186 clinical isolates of 10 species of the genus Candida and two culture collection strains were tested by an agar-dilution technique. Isoconazole was the most active azole, followed by butoconazole and sulconazole. Differences between some of the species in their susceptibility to the antifungal agents were noted. Sulconazole and cloconazole had the highest activity in vitro against 106 isolates of C. albicans. Butoconazole and isoconazole were also very active against isolates of C. albicans, and were the most active azole compounds against 80 isolates of Candida spp.  相似文献

Summary Met-enkephalin (ME) exerts a bimodal effect on functional activities of rat peritoneal macrophages (PM); in a range of low concentration (10-9-10-7 M) antibody dependent cellular cytotoxicity (ADCC)was markedly stimulated with a simultaneous decrease of Fc receptor (FcR) mediated phagocytosis while the opposite was observed at 10-6-10-5 M concentrations.Studying the possible underlying mechanism(s) the followings were recorded: (1) ME in all applied concentrations induced an early Na+ influx which was followed by a Ca2+ efflux in the range of low concentrations. In the range of high concentrations Na+ influx was accompanied by a Ca2+ influx. (2) ME at 10-8 M concentration induced a rise in cGMP level with a plateau in the 60–120th min of incubation. This effect was prevented by 10-5 M of naloxone. At 10-6 M concentration a transient rise of cAMP level was recorded which was not affected by naloxone. (3) Verapamil in 10-6 M abolished both the Ca2+ influx and the rise in cAMP level induced by 10-6-10-5 M ME but not the rise in cGMP level induced by lower ME concentrations. (4) cAMP elevation by high ME concentrations was abolished by enkephalinase inhibitory puromycin. (5) PM-enkephalinase as assessed by the cleavage of fluorogenic substrate L-alanine beta naphthylamide (ABNA), was inhibited by 10-6-10-5 M of ME. This inhibition was abolished by verapamil, but not affected by naloxone. In the range of low concentrations ME appears to act on specific delta opioid receptors and its action is positively coupled to guanylate cyclase. In relatively higher concentrations ME-action is not mediated by specific delta opioid receptors and it appears to involve Ca2+ influx, adenylate cyclase activation as well as the processing of hormone by PM-enkephalinase.  相似文献

Summary Small intensely fluorescent (SIF) cells appeared singly or, more frequently, in variably-sized clusters in the sacroccygeal 8th and 9th sympathetic ganglia of the bullfrog. Smaller clusters containing only two to nine SIF cells accounted for 61% of 1773 clusters examined. The largest cluster contained 283 cells. The number of cells in individual ganglia also varied from 21 to 3332. SIF cells, solitary as well as in smaller clusters, received no distinct form of the synaptic contact. In contrast, the cells in larger clusters were frequently innervated by nerve endings that were similar in vesicular constitution to the nerve endings on principal ganglion (PG) cells. No synaptic contact was found between SIF cells and PG cells. SIF cells were also characterized by their location in the vicinity of blood capillaries with a continuous endothelium. p]Our observation seems to suggest that larger clusters of SIF cells receiving nerve endings are linked to a paracrine and/or endocrine system. Chemical influence via the blood stream and intraganglionic milieu for non-innervated SIF cells in the solitary or smaller clusters is a subject for speculation. An interneuronal role of SIF cells to relay stimuli to PG cells seems unlikely. The possible functions here assigned to SIF cells could be variable in efficiency depending on their population and density.  相似文献

1. (14)C from [1-(14)C]glucose injected intraperitoneally into mice is incorporated into glutamate, aspartate and glutamine in the brain to a much greater extent than (14)C from [2-(14)C]glucose. This difference for [1-(14)C]glucose and [2-(14)C]glucose increases with time. The amount of (14)C in C-1 of glutamate increases steadily with time with both precursors. It is suggested that a large part of the glutamate and aspartate pools in brain are in close contact with intermediates of a fast-turning tricarboxylic acid cycle. 2. (14)C from [1-(14)C]acetate and [2-(14)C]acetate is incorporated to a much larger extent into glutamine than into glutamate. An examination of the time-course of (14)C incorporated into glutamine and glutamate reveals that glutamine is not formed from the glutamate pool, labelled extensively by glucose, but from a small glutamate pool. This small glutamate pool is not derived from an intermediate of a fast-turning tricarboxylic acid cycle. 3. It is proposed that two different tricarboxylic acid cycles exist in brain.  相似文献

A new intermediate was identified in the 2-tridecanone pathway of Pseudomonas multivorans, formerly designated pseudomonad 4G-9. This intermediate, undecyl acetate, was isolated directly from growing cultures of the organism; the structure of the intermediate was determined by infrared spectroscopy and by gas-liquid chromatographic identification of its hydrolytic products. An amended pathway is presented that accounts for the conversion of 2-tridecanone to provide carbon and energy for growth. It was shown that all early intermediates in the pathway arise biologically and sequentially from their precursors. Studies with P. aeruginosa showed that this organism also degrades 2-tridecanone by the pathway characteristic of P. multivorans. Biochemical mechanisms of the pathway are discussed. Discovery of undecyl acetate confirms our earlier contention that the primary attack on methyl ketones by bacteria can be by subterminal oxidation.  相似文献

A study of the microbial utilization of long-chain methyl ketones was under-taken. In general, enrichment culture experiments revealed that soil microorganisms capable of utilizing these compounds as growth substrates are ubiquitous. Gram-negative, rod-shaped bacteria were the prominent organisms exhibiting this capability. In particular, a strain of Pseudomonas isolated from soil degraded 2-tridecanone into several products that were recovered from cell-free culture fluid. These products were identified by gas-liquid chromatography as 2-tridecanol, 1-undecanol, 1-decanol, and undecanoic acid. A large amount of the substrate was converted to 1-undecanol. This compound was characterized further by classical methods of organic analysis. Unequivocal identification of 1-undecanol has established that some unique mechanism that involves subterminal oxidation must exist to degrade 2-tridecanone. No such mechanism has been reported for the biological degradation of long-chain, aliphatic, methyl ketones. A pathway for utilization of 2-tridecanone was proposed that is consistent with, but not confirmed by, the data presented.  相似文献

Corticotropin-releasing hormone (CRH) has been shown to be a central mediator for most, if not all, stress-induced responses. Since stressful stimuli may decrease hypothalamic tuberoinfundibular and tuberohypophysial dopaminergic neuronal activities, we aimed to determine whether CRH is involved. Using central administration of various doses of ovine CRH (oCRH; 1, 3 and 10 µg/rat) into the lateral cerebroventricle of either male or female rats, the neurochemical changes in various parts of the central nervous system, including the hypothalamus, were determined by high-performance liquid chromatography at various times after the injection (30, 60, 120 and 240 min). The concentrations of 3,4-dihydroxyphenylacetic acid (DOPAC) and 3-methoxy-4-hydroxy-phenylethyleneglycol (MHPG), two major metabolites of dopamine and norepinephrine, respectively, in discrete brain regions were used as indices for catecholaminergic neuron activity. Plasma corticosterone levels increased significantly after all doses of oCRH and at all time points studied. oCRH also exerted significant stimulatory effects on noradrenergic neuron terminals in the frontal cortex, and on dopaminergic neuron terminals in the nucleus accumbens, hypothalamic paraventricular and periventricular nuclei, and intermediate pituitary lobe. Dopaminergic neuron terminals in the median eminence and the neural lobe of the pituitary, however, were not affected. There was no major difference in the responses between male and female rats. We conclude that CRH has a differential effect on central catecholaminergic neurons.  相似文献

Mitogen- and isoproterenol-induced changes of [Ca2+]i in T cells attached to a glass substrate were examined. Murine (C57BL/6) splenic T cells were attached to coverslips or 35-mm dishes (MatTek) precoated with Cell Tak® (3.5 µg/cm2). The cells were then loaded with fluorescent dye (2 µg/ml of fura2-AM or fluo3-AM) and changes in [Ca2+]i in a population of cells (using a spectrofluorometer) or in single cells (using a confocal microscope) were measured during continuous superfusion. Population measurements of [Ca2+]i demonstrated that concanavalin A (Con A, 2 or 5 µg/ml) caused an increase in [Ca2+]i that rose to a peak and then declined to a steady state. The concentration-response relationship (0.05–5 µg/ml) had an EC50 of 0.3 µg/ml. Isoproterenol decreased the Con A-induced elevation of steady state [Ca2+]i. In single cell studies, the increase in [Ca2+]i in response to Con A typically occurred in about 50% of the cells in a microscope field, and the delay before activation varied among cells. Taken together, these data demonstrate that Cell Tak® can be used to attach T cells to glass coverslips and will be useful for the study of signaling mechanisms in T cells.  相似文献
