By using the isotope pairs (22)Na-(24)Na and (42)K-(86)Rb, the uptake and retention of Na and K was studied in the salt-tolerant Debaryomyces hansenii and in the less tolerant Saccharomyces cerevisiae at NaCl levels of 4 mm and 0.68, 1.35, and 2.7 m in the medium. The ratio of K to Na is much higher in the cells than in the media, and higher in D. hansenii than in S. cerevisiae under comparable conditions. The difference between the two species is due to a better Na extrusion and a better uptake of K in D. hansenii. The kinetics of ion transport show that at about the time when extrusion of Na could be demonstrated in D. hansenii, K-Rb previously lost to an easily washable compartment of the cells was reabsorbed in both organisms. More H(+) was given off from S. cerevisiae than from D. hansenii in the course of these events. The findings fit the working hypothesis tested, which regards salt tolerance as partly dependent on the ability to mobilize energy to extrude Na from the cells and to take up K. The volume changes in S. cerevisiae are greater and are more slowly overcome than those in D. hansenii. The total salt level of the cells is not sufficient to counteract the osmotic potential of the medium, so that additional osmoregulatory mechanisms must be involved in determining halotolerance.  相似文献

Homogenates from the salt-excreting leaves of the mangrove Avicennia nitida were subjected to differential centrifugation and investigated for adenosine triphosphatase activities. At pH 6.75 a salt stimulation with peaks at three different sodium to potassium ratios could be demonstrated above the activity due to Mg2+ ions. The stimulation by sodium and potassium depends on the ionic strength of the test medium, higher salt concentrations being inhibitory. The plant system seems thus more complicated than the animal activities. Technically, this means that a search for (Na+ + K+)-activated ATPases in plants should be performed with a close spacing of Na:K ratios at several constant levels of salt. Literature data on the transport of Na+ and K+ indicate that the physiological situation is rather complex in plants.  相似文献

Treatment of Nicotiana glutinosa L. cell suspension cultures with chitosan results in the co-induction of phenylalanine ammonia lyase, 4-eoumarate:CoA ligase, tyrosine decarboxylase and tyramine hydroxycinnamoyltransferase, all involved in the biosynthesis of hydroxycinnamoyltyramines. The highest enzyme activities were observed around 12 h after addition of 0.8 to 1 mg chitosan per g fresh weight of cells. No hydroxycinnamoyltyramines could be detected by TLC or HPLC of extracts made from non-treated or elicited cells. [14C]-Tyramine was incorporated into insoluble polymeric material at a higher rate in elicitor-treated than in non-treated cells of N. glutinosa. Tyramine hydroxycinnamoyltransferase could be induced in suspension cultured cells of Eschscholtzia califortnca Cham, but not in cells of Phaseolus vulgaris L. or Catharanthus roseus (L.) G. Don by addition of a yeast elicitor to the growth medium.  相似文献

The effects of NaCl and replacement of K+ by Na+ on the lipid composition of the two sugar beet inbred lines FIA and ADA were studied (a) with increasing additions of NaCl to the basal medium, and (b) with increasing replacement of K+ by Na+ at the same total concentration as in the basal medium. Direct relations were noted between NaCl concentration of the nutrient solution and the phospholipid concentration in the roots of FIA, the genotype characterized by a low K+/Na+ ratio, as well as between NaCl in the medium and the phospholipid concentration in the shoots of ADA, the genotype with a high K +/Na + ratio. The sulfolipid level in the roots of FIA was maintained at higher NaCl concentrations, while it was decreased in ADA. The glycolipid concentration in the shoots of ADA and the degree of unsaturation of the fatty acids of the total lipid fraction were decreased by salinity, indicating reduced biosynthesis of chloroplast glycolipids and/or accelerated oxidation of these lipids in the presence of NaCl.
In the Na+ for K+ replacement experiment a low content of K+ in the medium resulted in decreased levels of total lipids, phospholipids and sulfolipid in the roots of both genotypes, which did not relate to root growth. K+-leakage from the roots at low K+-level in the medium may be reduced by the increase in saturation of the lipids. In the shoots of ADA increased levels of total lipids, phospholipids and Sulfolipid were noted at a low K+-concentration of the nutrient solution.  相似文献

Mg2+- and Ca2+-uptake was measured in dark-grown oat seedlings ( Avena sativa L. cv. Brighton) cultivated at two levels of mineral nutrition. In addition the stimulation of the ATPase activity of the microsomal fraction of the roots by Mg2+ was measured. Ca2+-uptake by the roots was mainly passive. Mg2+-uptake mainly active; the passive component of Mg2+-uptake was accompanied by Ca2+-efflux up to 60% of the Ca2+ present in the roots.
In general Mg2+ -uptake of oat roots was biphasic. The affinity of the second phase correspond well with that of the Mg2+-stimulation of the ATPase activity, in low-salt roots as well as in high-salt roots and in roots of plants switched to the other nutritional condition. Linear relationships were observed when [phase 2] Mg2+-uptake was plotted against Mg2+-stimulation of the ATPase activity of the microsomal fraction of the roots. In 5 days old high-salt plants 1 ATP (hydrolysed in the presence of Mg2+ J corresponded with active uptake of a single Mg2+ ion, but in older high-salt roots and in low-salt roots more ATP was hydrolysed per net uptake of a Mg2+ ion. The results are discussed against the background of regulation of the Mg2+-level of the cytoplasm of root cells by transport of Mg2+ by a Mg2+-ATPase to the vacuole, to the xylem vessels, and possibly outwards.  相似文献

The cells used in the present investigation had a phosphate content of about 20 per cent as compared with the status in normal cultures. The uptake of phosphate during a period of 4 hours was determined at a pH of 6,5, kept constant with the aid of a citrate buffer. In the absence of CO2, light increased the uptake of phosphate with saturation around 14,000 erg/cm2s. With 5 per cent CO2 in the air the relationship was more complicated, and the uptake of phosphate must he related to more than one process during active photosynthesis. The inhibiting effect of CO2 in air was noticeable already at low concentrations both in light and in darkness. With the system used, this supports earlier indications for internal recycling of orthophosphate, CO2 was inhibiting also in nitrogen in the light. Selenate in a concentration of 2 mM gave a slight and rather irregular inhibition.—Anaerobiosis had no effect in the light but gave a large decrease in the dark.—DNP (0.1 mM) was somewhat more active in the dark than in the light. The lower concentrations tested had no effect in either case.—Menadione (0.1 mM) inhibited strongly, and more in illuminated than in non-illuminated cells.  相似文献
