Summary The origin of mitotic cells was investigated histologically in chorionic tissue, metaphase plates of which were used for direct cytogenetic study. Mitotic figures were often observed in the Langhans' cell layers, but absolutely none were seen in the syncytium and stromal cells.  相似文献

The dissociation of wheat glutenin into subunits was observed by treatment with a small amount of mercuric chloride under moderate conditions, suggesting that the cleavage of inter-polypeptide chain disulfide bonds in the glutenin might occur. The dissociation into the subunits was examined by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis. The electrophoretic patterns of the glutenin treated with mercuric chloride were essentially similar to those of the glutenin treated with 2-mercaptoethanol. Silver nitrate also had the same effects as mercuric chloride, and p-chloromercuribenzoate and N-ethylmaleimide showed no effect on the dissociation of the glutenin. Complete dissociation was achieved when the glutenin solution containing 0.5% SDS and 0.01 m phosphate buffer (pH 7.0) was incubated with 10?3 m mercuric chloride (about four moles per mole of disulfide groups) at 30°C for 20 hr. Partial dissociation was also observed after 30 min incubation. Increasing temperature and SDS concentration promoted the rate of the dissociation of the glutenin by mercuric chloride.  相似文献

A dietary carcinogen, 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indole (Trp-P-1) at 20 microM activates caspase-3-like proteases as an apoptotic marker in rat splenocytes. The present study demonstrated 100 microM Trp-P-1 induced necrosis with activation of caspase-3-like proteases. The activation in necrosis and apoptosis resulted from the activation of caspase-9 and caspase-8, respectively. Thus, Trp-P-1 induces apoptosis and necrosis with the activation of different caspases.  相似文献

The 569,750 base pair sequence corresponding to the 28.0–40.1min region on the genetic map of Escherichia coli K-12 (W3110)was determined. This region includes the replication terminusregion and contained at least 549 potential open reading frames.Among them, 160 (29%) were previously reported, 174 (32%) werehomologous to other known genes, 102 (18%) were identical orsimilar to hypothetical genes registered in databases, and theremaining 113 (21%) did not show a significant similarity toany other gene. Of interest was the finding of a large numberof genes and gene clusters in andnear the replication terminationregion which had been thought to be genetically silent. Thoseincludeda cluster of genes for fatty acid ß-oxidation,the third copy of the pot (spermidine/putrescine transport system)gene cluster, the second dpp (dipeptide transport system) operon,the second dsm (anaerobic dimethyl sulfoxide reductase) operon,a cluster of fim (fimbrial) genes anda DNA helicase-like genewith a high molecular weight. (+)-Cyanofenphos was at least more than 25-fold as toxic to rice stem borer larvae as the (?)-isomer. In addition, (+)-cyanofenphos-oxon was 36-fold more potent inhibitor than the (?)-oxon toward rice stem borer larva acetylcholinesterase. The (+)-, (?)- and racemic forms of cyanofenphos were metabolized in rice stem borer larvae at almost equal rates. However, (+)-cyanofenphos produced 4- to 9-fold larger amounts of cyanofenphos-oxon in the insect body than the (?)-isomer. On the other hand, (?)-cyanofenphos-oxon was metabolized to 4-cyanophenol and its conjugate at a faster rate compared to the (+)-oxon. The difference in toxicity to rice stem borer larvae exhibited by the optical isomers of cyanofenphos is attributable to the difference in the amount and persistence of cyanofenphos-oxon isomers formed in the insect body as well as in anti-acetylcholinesterase activity of the oxon isomers.  相似文献

3-Amino-1,4-dimethyl-5H-pyrido[4,3-b]indole (Trp-P-1), which is a tryptophan pyrolysate formed during cooking, induces apoptosis in rat splenocytes, thymocytes, and hepatocytes. In this study, we investigated whether Trp-P-1 is transported into these cells and causes apoptosis. Trp-P-1 was immediately incorporated into rat splenocytes, thymocytes, and hepatocytes in a dose- and time-dependent manner. Dopamine and serotonin significantly competed with the uptake of Trp-P-1 into these cells, and nomifensine and indatraline, which are inhibitors of dopamine- and serotonin-transporters, respectively, markedly suppressed the uptake of Trp-P-1. On the other hand, amino acids including tryptophan did not compete with Trp-P-1. Inhibition of monoamine transporters using nomifensine and indatraline partially suppressed Trp-P-1-induced cell death in these cells. In hepatocytes, the inhibition of transporters prevented Trp-P-1-induced morphological changes and activation of caspase-3. These results demonstrated that Trp-P-1 is incorporated into the cells through monoamine transporters and induces apoptosis.  相似文献

Cells of Bacillus coagulans, strain HN-68 grown on the medium containing d-glucose, did not show any measurable d-glucose-isomerizing activity. However, when d-glucose-grown cells were shaked for a few hours in an induction medium containing d-xylose, induced formation of d-glucose-isomerizing enzyme occurred in the cells. Cell weight and number of survival cells showed only an increase of 30 and 10%, respectively during 6 hr induction.

The induced formation of d-glucose-isomerizing enzyme required organic nitrogen such as polypeptone in addition to d-xylose. Development of the maximum activity was observed when the concentration of d-xylose and polypeptone were 2 and 3%, respectively. Initial velocity of induced formation of d-glucose-isomerizing enzyme increased in proportion to the decrease of initial pH values of the induction medium, i.e., at 2 hr induction, activity at pH 4.5 was 5-fold increase than that at pH 8.0.

Induced formation of d-glucose-isomerizing enzyme was inhibited strongly by addition of chloramphenicol, tetracycline, streptomycin, cyanide or azide, and was promoted by threonine plus glycine.  相似文献
