Background
DNA Microarrays have become the standard method for large scale analyses of gene expression and epigenomics. The increasing complexity and inherent noisiness of the generated data makes visual data exploration ever more important. Fast deployment of new methods as well as a combination of predefined, easy to apply methods with programmer's access to the data are important requirements for any analysis framework. Mayday is an open source platform with emphasis on visual data exploration and analysis. Many built-in methods for clustering, machine learning and classification are provided for dissecting complex datasets. Plugins can easily be written to extend Mayday's functionality in a large number of ways. As Java program, Mayday is platform-independent and can be used as Java WebStart application without any installation. Mayday can import data from several file formats, database connectivity is included for efficient data organization. Numerous interactive visualization tools, including box plots, profile plots, principal component plots and a heatmap are available, can be enhanced with metadata and exported as publication quality vector files. 相似文献4.

During the genomics era, the use of thermostable DNA polymerases increased greatly. Many were identified and described—mainly of the genera Thermus, Thermococcus and Pyrococcus. Each polymerase has different features, resulting from origin and genetic modification. However, the rational choice of the adequate polymerase depends on the application itself. We present a synthesis of the palaeolimnological and palaeoclimatic reconstructions of four sites in Colombia. The record from Lake El Caimito, the westernmost site on the Pacific Coast, dates from the Late Holocene and shows lacustrine sedimentation frequently interrupted by fluvial pulses. These pulses probably reflect periods of increased precipitation related to La Niña phases. East of El Caimito site is the Patía swamp, situated between the Western and Central Andean Cordilleras. The Patía records the dynamics of forest expansion/reduction and changes in water levels. Although the climatic signal of the Patia core is difficult to reconstruct, there is a clear increase in humidity in the Mid-Holocene. The Fúquene Lake record, on the Eastern Andean Cordillera, records dry and cold conditions during the Late Pleistocene, very humid conditions for the early Mid-Holocene, and dry conditions during the mid-Late Holocene. Las Margaritas site, on the eastern savannas, records dry conditions during the Early Holocene and overall humid conditions for the Mid- and Late Holocene. Climate conditions from the Fuquene and Las Margaritas sites seem to reflect the Holocene movements of the Intertropical Convergence Zone (ITCZ); the latter site being more affected by humidity coming from the Amazon region. 相似文献

Few fish species form single, panmictic populations throughout their geographic range, most form subpopulations or 'stocks' with differing levels of interconnectivity. Different patterns of interconnectivity between subpopulations will give rise to different responses to exploitation and management, but they will also have different capacities to generate the genetic and phenotypic differences often used to discriminate between stocks. Consequently, knowledge of ontogenetic and seasonal patterns in the distribution, movement and behaviour of individuals is crucial to identifying population substructure. The preparation of lactate dehydrogenase covalently attached to anion-exchange cellulose particles and sheets by use of a dichloro-sym-triazinyl dyestuff, Procion brilliant orange MGS, is described. 2. The stability and kinetic properties of these preparations were investigated. 3. An equation is derived to describe the change in concentration of a substrate when passed through a uniform bed of a substrate-inhibited enzyme. A number of theoretical curves are shown to illustrate the system. 4. A titrimetric assay for lactate dehydrogenase is described, and shown to be stoicheiometric over the range pH5.0-9.2. 5.
