Plasmid analysis of 46 strains of Shigella species confirmed the view about heterogeneity and variability of plasmids composition in these microorganisms. On the other hand,the presence of low-molecular, multiplicated extrachromosomal replicons, which were stable, was shown. Using them, it was possible to divide the Shigella isolates into two major plasmidovars, characterize them according to repertoire of the most stable plasmids and determine epidemically significant variants during sporadic and outbreak incidence of shigellosis in towns Nizhny Novgorod and Kstovo (Nizhny Novgorod region).  相似文献

The frequency and spectrum of mutations in RAS/BRAF genes were studied in rectal adenomas, carcinomas in situ, and adenocarcinomas. It is shown that the frequency of KRAS mutations decreases from adenoma to adenocarcinoma; most adenomas and carcinomas in situ are heterogeneous and consist of several subclones. Possible models of colorectal cancer pathogenesis are discussed.  相似文献

The expression profile of the ZEB1, ZEB2, VIM, CDH1, SFRP2, FOXQ1, TNC, MACC1, PLS3, CFTR, FLNA, MUC2, TFF3, and RARRES3 genes, as well as the mutational status of the KRAS, NRAS, BRAF, and PIK3CA genes, were investigated in 40 patients with colorectal cancer and liver metastases. A comparative analysis of changes in gene expression in primary tumor cells and liver metastases was performed. Statistically significant differences were found between the expression levels of the ZEB2 (p = 0.004), VIM (p < 0.001), FLNA (p = 0.04), and MUC2 (p < 0.001) genes. It was demonstrated that the overall frequency of mutations of the KRAS gene was 18/40 (45%) and the PIK3CA gene was 9/40 (23%). Mutations in the NRAS and BRAF genes were not found. The concordance between the primary tumor and metastases in the liver by mutation status was 100%.  相似文献

Presence of plasmid-contained strains in probiotics prepared on the basis of Bacillus bacteria was assessed. Plasmid analysis was performed by the method based on neutralization of bacterial suspension after alkaline treatment with lithium chloride. Presence of 4-6 plasmids with molecular weight from 1 to 75 MDa was revealed in 4 commercially available probiotics. Since plasmids can facilitate transfer of genes of the antimicrobial resistance to pathogenic microorganisms it was recommended to perform thorough search of probiotic microorganisms without plasmids or ensure the elimination of R-plasmids in start cultures for the most promising biopreparations.  相似文献

The primary structure of the APC gene DNA was examined in 108 patients younger than 45 years old diagnosed with “familial adenomatous polyposis, classic form” using PCR, conformation-sensitive electrophoresis, and Sanger sequencing. Mutations in the APC gene were observed in 78 patients; de novo mutations were observed in 17 cases. In the majority of cases (n = 45), patients exhibited frameshift mutations, 28 patients had nonsense mutations, and other 5 patients showed splicing mutations. We also revealed recurring variants: p.Arg232X (2 cases), p.Asp849GlufsX11 (2), p.Ser1068GlyfsX57 (2), p.Arg216X (3), p.Gln1062X (5), p.Arg213X (5), and p.Glu1309AspfsX4 (16). It was shown that, compared with other pathogenic variants in the APC gene in Russian patients, mutation p.Glu1309AspfsX4 does not result in earlier development of colorectal cancer and polyps. Nineteen mutations were described for the first time. The identified mutations were located between codons 142 and 1492 of the APC gene. This indicates the importance of investigation of all the gene coding exons. Pathogenic variants were observed in 16 of 35 studied relatives of the mutation carriers. All 16 relatives were included in the “risk group” for lifelong clinical monitoring.  相似文献

The optimum and available methods for the isolation of DNA from bacteria, used as the basis of the industrial manufacture of eubiotics (colibacterin, lactobacterin, biobactone) have been developed. The proposed approach makes it possible to isolate DNA, suitable for genosystematic studies under laboratory conditions, directly from producer strains.  相似文献
