Chagas disease, which is caused by the parasite Trypanosoma cruzi, is an important cause of heart failure. We investigated modifications in the cellular electrophysiological and calcium-handling characteristics of an infected mouse heart during the chronic phase of the disease. The patch-clamp technique was used to record action potentials (APs) and L-type Ca2+ and transient outward K+ currents. [Ca2+]i changes were determined using confocal microscopy. Infected ventricular cells showed prolonged APs, reduced transient outward K+ and L-type Ca2+ currents and reduced Ca2+ release from the sarcoplasmic reticulum. Thus, the chronic phase of Chagas disease is characterised by cardiomyocyte dysfunction, which could lead to heart failure. 相似文献

Termination of cardiac Ca(2+) sparks: an investigative mathematical model of calcium-induced calcium release

A Ca(2+) spark arises when a cluster of sarcoplasmic reticulum (SR) channels (ryanodine receptors or RyRs) opens to release calcium in a locally regenerative manner. Normally triggered by Ca(2+) influx across the sarcolemmal or transverse tubule membrane neighboring the cluster, the Ca(2+) spark has been shown to be the elementary Ca(2+) signaling event of excitation-contraction coupling in heart muscle. However, the question of how the Ca(2+) spark terminates remains a central, unresolved issue. Here we present a new model, "sticky cluster," of SR Ca(2+) release that simulates Ca(2+) spark behavior and enables robust Ca(2+) spark termination. Two newly documented features of RyR behavior have been incorporated in this otherwise simple model: "coupled gating" and an opening rate that depends on SR lumenal [Ca(2+)]. Using a Monte Carlo method, local Ca(2+)-induced Ca(2+) release from clusters containing between 10 and 100 RyRs is modeled. After release is triggered, Ca(2+) flux from RyRs diffuses into the cytosol and binds to intracellular buffers and the fluorescent Ca(2+) indicator fluo-3 to produce the model Ca(2+) spark. Ca(2+) sparks generated by the sticky cluster model resemble those observed experimentally, and Ca(2+) spark duration and amplitude are largely insensitive to the number of RyRs in a cluster. As expected from heart cell investigation, the spontaneous Ca(2+) spark rate in the model increases with elevated cytosolic or SR lumenal [Ca(2+)]. Furthermore, reduction of RyR coupling leads to prolonged model Ca(2+) sparks just as treatment with FK506 lengthens Ca(2+) sparks in heart cells. This new model of Ca(2+) spark behavior provides a "proof of principle" test of a new hypothesis for Ca(2+) spark termination and reproduces critical features of Ca(2+) sparks observed experimentally. 相似文献

Maned wolves are large canids currently considered vulnerable to extinction due to habitat loss. They are still commonly found
within the urban mesh inside the Brazilian Federal District (Distrito Federal—DF), in nearby Protected Areas (PAs), and in
surrounding farms. We evaluated the genetic diversity of maned wolves in three PAs of the DF, using both invasive and noninvasive
techniques to obtain DNA that was later amplified for five microsatellite markers. We sampled 23 wolves: 10 with the noninvasive
method, three captured in traps, six road-killed, and four rescued in urban areas. In águas Emendadas Ecological Station (ESECAE)
we also used samples from six specimens captured between 1997 and 1998 for a temporal comparison. For maned wolves, non-invasive
techniques are affordable and easier to conduct in the field, while laboratory costs are much lower for invasive samples.
Hence, a sampling strategy combining both techniques may provide an interesting approach for molecular ecology studies requiring
comprehensive coverage of local individuals. On the basis of such integrated sampling scheme, our analyses indicated that
none of the investigated populations currently present deviations from Hardy–Weinberg expectations or indication of inbreeding.
Furthermore, in ESECAE there was no reduction in genetic diversity during the last 9 years. Overall, maned wolves did not
present evidence of genetic structuring among the three sampled PAs. These results thus indicate that individual exchange
among PAs is still occurring at sufficient rates to avoid differentiation, and/or that the recent fragmentation in the region
has not yet produced measurable effects in the genetic diversity of maned wolves. 相似文献

The natural transmission cycle of Yellow Fever (YF) involves tree hole breeding mosquitoes and a wide array of nonhuman primates (NHP), including monkeys and apes. Some Neotropical monkeys (howler monkeys, genus Alouatta) develop fatal YF virus (YFV) infections similar to those reported in humans, even with minimum exposure to the infection. Epizootics in wild primates may be indicating YFV circulation, and the surveillance of such outbreaks in wildlife is an important tool to help prevent human infection. In 2001, surveillance activities successfully identified YF-related death in a black-and-gold howler monkey (Alouatta caraya), Rio Grande do Sul State (RGS) in southern Brazil, and the YFV was isolated from a species of forest-dwelling mosquito (Haemagogus leucocelaenus). These findings led the State Secretariat of Health to initiate a monitoring program for YF and other 18 arboviral infections in Alouatta monkeys. The monitoring program included monkey captures, reporting of monkey casualties by municipalities, and subsequent investigations. If monkey carcasses were found in forests, samples were collected in a standardized manner and this practice resulted in increased reporting of outbreaks. In October 2008, a single howler monkey in a northwestern RGS municipality was confirmed to have died from YF. From October 2008 to June 2009, 2,013 monkey deaths were reported (830 A. caraya and 1,183 A. guariba clamitans). Viruses isolation in blood, viscera, and/or immunohistochemistry led to the detection of YF in 204 of 297 (69%) (154 A. g. clamitans and 50 A. caraya) dead Alouatta monkeys tested. The number of municipalities with confirmed YFV circulation in howlers increased from 2 to 67 and 21 confirmed human cases occurred. This surveillance system was successful in identifying the largest YF outbreak affecting wild NHP ever recorded. 相似文献

Flavonoids are naturally occurring polyphenolic compounds that are present in a variety of fruits, vegetables, cereals, tea,
and wine, and are the most abundant antioxidants in the human diet. Evidence suggests that these phytochemicals might have
an impact on brain pathology and aging; however, neither their mechanisms of action nor their cell targets are completely
known. In the mature mammalian brain, astroglia constitute nearly half of the total cells, providing structural, metabolic,
and trophic support for neurons. During the past few years, increasing knowledge of these cells has indicated that astrocytes
are pivotal characters in neurodegenerative diseases and brain injury. Most of the physiological benefits of flavonoids are
generally thought to be due to their antioxidant and free-radical scavenging effects; however, emerging evidence has supported
the hypothesis that their mechanism of action might go beyond these properties. In this review, we focus on astrocytes as
targets for flavonoids and their implications in brain development, neuroprotection, and glial tumor formation. Finally, we
will briefly discuss the emerging view of astrocytes as essential characters in neurodegenerative diseases, and how a better
understanding of the action of flavonoids might open new avenues to develop therapeutic approaches to these pathologies. 相似文献

Tick species parasitizing yellow armadillo, Euphractus sexcinctus, were recorded from October 2006 to October 2007 in Pantanal. A total of 89 ticks were collected from 23 yellow armadillos. Most ticks (n=50) were identified as Amblyomma cajennense (Fabricius) (32 males and 18 females). The only other species found was Amblyomma parvum (Arag?o) (three females and two males). The remaining ticks were immatures of Amblyomma (29 nymphs and five larvae). The prevalence of A. cajennense was 55%, the mean intensity was 2.7±0.5 (SE) (n=17) and the mean abundance was 1.5±0.4 (n=31). 相似文献

The small G protein Ras-mediated signaling pathway has been implicated in the development of hypertrophy and diastolic dysfunction in the heart. Earlier cellular studies have suggested that the Ras pathway is responsible for reduced L-type calcium channel current and sarcoplasmic reticulum (SR) calcium uptake associated with sarcomere disorganization in neonatal cardiomyocytes. In the present study, we investigated the in vivo effects of Ras activation on cellular calcium handling and sarcomere organization in adult ventricular myocytes using a newly established transgenic mouse model with targeted expression of the H-Ras-v12 mutant. The transgenic hearts expressing activated Ras developed significant hypertrophy and postnatal lethal heart failure. In adult ventricular myocytes isolated from the transgenic hearts, the calcium transient was significantly depressed but membrane L-type calcium current was unchanged compared with control littermates. The expressions of sarco(endo)plasmic reticulum Ca(2+)-ATPase (SERCA)2a and phospholamban (PLB) were significantly reduced at mRNA levels. The amount of SERCA2a protein was also modestly reduced. However, the expression of PLB protein and gross sarcomere organization remained unchanged in the hypertrophic Ras hearts, whereas Ser(16) phosphorylation of PLB was dramatically inhibited in the Ras transgenic hearts compared with controls. Hypophosphorylation of PLB was also associated with a significant induction of protein phosphatase 1 expression. Therefore, our results from this in vivo model system suggest that Ras-induced contractile defects do not involve decreased L-type calcium channel activities or disruption of sarcomere structure. Rather, suppressed SR calcium uptake due to reduced SERCA2a expression and hypophosphorylation of PLB due to changes in protein phosphatase expression may play important roles in the diastolic dysfunction of Ras-mediated hypertrophic cardiomyopathy. 相似文献
