NADPH2 and ATP competitively inhibit sweet potato glucose 6-phosphatedehydrogenase with NADP and glucose 6-phosphate (G6P), respectively.At pH 8.0, a Lineweaver-Burk plot of the reciprocal rate againstreciprocal G6P concentration was concave downwards in the presenceand absence of ATP, whereas a double reciprocal plot followedthe Michaelis-Menten relationship at pH 7.0, irrespective ofthe presence of ATP. Many of the other metabolic intermediatestested had no effects on the enzyme reaction.

Intact tubers of potato (Solanum tuberosum L. cv. Irish Cobblerand an interspecific hybrid between S. tuberosum and S. demissumcv. Rishiri) contain a very low activity of 3-hydroxy-3-methylglutaryl(HMG)-CoA reductase. The activity increased first in responseto slicing, and again in response to additional treatments suchas inoculation with an incompatible race of Phytophthora infestans,application of a hyphal wall component of the fungus or HgCl2solution, and then decreased. Both the first and the secondincreases in activity in response to slicing and additionaltreatment with a hyphal wall component to elicit phytoalexinproduction were inhibited by blasticidin S. Properties of HMG-CoAreductase induced by slicing and by additional treatment withHgCl2 or fungal inoculation were investigated.

In roots of sweet potato (Ipomoea batatas Lam. cv. Kokei 14),the metabolic response to wounding was remarkable only in theproximal side. We assumed that the polarity resulted from apolar movement of indole-3-acetic acid (IAA) produced in thecut surface (8). As the metabolic response was slight in thedistal side, the effect of IAA and the other plant hormoneson the development of various enzyme activities was examinedin this side. Increases in activities of L-phenylalanine ammonia-lyase,acid invertase, NADPHa2 : cytochrome c oxidoreductase, peroxidase,cytochrome c : O2 oxidoreductase and o-diphenol oxidase, whichdeveloped in response to wounding, were stimulated by the treatmentwith IAA. Gibberellic acid had a stimulative effect on the developmentof only acid invertase activity. Abscisic acid and kinetin hadlittle effect. Spore germination of Ceratocystis fimbriata was studied in termsof host-parasite specificity. The sweet potato, coffee and cacaostrains of Ceratocystis fimbriata germinated well in a fractionof sweet potato root water extract which had been passed througha column of cation exchange resin. The results showed that germinationof these strains was independent of exogenous cations. On theother hand, the prune, oak, taro and almond strains requiredfor germination both the absorbed and unabsorbed fractions ofsweet potato root water extract which were separated from eachother with a cation exchange resin column. Divalent cationssuch as Ca2+ Mg2+, Mn2+ and Zn2+ were identified as the activeprinciples in the absorbed fraction and Ca2+ showed the highestinductive activity for spore germination in the presence ofthe unabsorbed fraction. The active principle(s) in the unabsorbedfraction has not yet been identified. There was no relationshipbetween the Ca2+ and Mg2+ contents of the spores and the requirementof exogenous Ca2+ for germination. Ca2+ appeared to functionas a trigger of spore germination, not as a normal nutrient.These results suggest that the divalent cations such as Ca2+and Mg2+ in sweet potato contribute to the establishment ofhost-parasite specificity of this system. Germinated spores of various strains of Ceratocystis fimbriatawere agglutinated differentially by concanavalin A. -Methyl-D-mannosideand -methyl-D-glucoside, specific hapten sugars of concanavalinA, inhibited agglutination of the fungus by concanavalin A.These results support the assumption that differences existamong various strains of C. fimbriata in carbohydrate constituentsor locations on the cell surfaces. The difference in the type of codon-anticodon base pairing at the ribosomal P-site is one of the determinants of the translational rate 总被引:3,自引:0,他引:3
By utilizing an enzymatically reconstructed tRNA variant containing an altered anticodon sequence, we have examined the different biochemical behavior of translation between the Watson-Crick type and the wobble type base pair interactions at the first anticodon position. We have found that the Watson-Crick type base pair has an advantage in translation in contrast to the wobble type base pair by comparing the efficiency of transpeptidation of native tRNA(Phe) (anticodon; GmAA) with its variant tRNA (anticodon; AAA) in the poly(U)-programmed ribosome system. Thomas et al. [Proc. Natl. Acad. Sci. U.S. (1988) 85, 4242-4246] showed that the wobble codon at the ribosomal A-site accepted its cognate tRNA less efficiently than the Watson-Crick base pairing codon. We report here that the wobble interaction at the ribosomal P-site also affected the rate of translation. This variable translational rate may be a mechanism of gene regulation through preferential codon usage. 相似文献

The bar-headed goose, a specialized high-altitude species, has a capacity for high oxygen uptake from a hypoxic environment. It thus has a higher oxygen affinity than other bird species of lower-altitude environments. Oxygen affinity is determined by molecular structures and genetic mutations of hemoglobin (Hb), which can also influence the coordinating structures and dynamics of oxygen-Hb. To explore the structural differences in Hbs as between high and low altitude species, photolysis dynamic parameters, including quantum yield, enthalpy, and conformational volume changes in carboxy-Hbs (HbCO) for the bar-headed goose and low altitude counterparts (the Chinese goose and chicken) were investigated by the laser pumping-probing technique and photoacoustic calorimetry. Comparing the photolysis results for HbCO of the three species, the enthalpy and conformational volume changes of the bar-headed goose were much smaller than those of the others, although the quantum yields of all three species are similar. To explain the possible mechanisms of these differences, modifications of salt bridges and key residue mutations at the α β subunit interfaces of the proteins are described and discussed briefly. 相似文献

It was shown in previous papers that in sweet potato infected with Ceratostomella fimbriata, there occurred metabolic activations such as respiratory increase, polyphenols production, protein synthesis and organic phosphate accumulation. Thienodolin, a new plant growth-regulating substance, was isolated from the fermentation broth of a streptomycete strain identified as Streptomyces albogriseolus.The active principle was extracted with ethyl acetate and purified by silica gel column chromatography and preparative HPLC.
