Nodulation is the first and quantitatively most important cellular defense reaction to bacterial infections in insects. Treating adults of the 17-year periodical cicadas, Magicicada septendecim and M. cassini, with eicosanoid biosynthesis inhibitors immediately prior to intrahemocoelic injections of the bacterium, Serratia marcescens, sharply reduced the nodulation response to bacterial challenges. Separate treatments with specific inhibitors of phospholipase A(2), cyclooxygenase, and lipoxygenase reduced nodulation, supporting our view that nodule formation is a multi-step process in which individual steps are separately mediated by lipoxygenase and cyclooxygenase products. The inhibitory influence of dexamethasone was apparent by 2 h after injection, and nodulation was significantly reduced, relative to control insects, over the following 14 h. The dexamethasone effects were reversed by treating bacteria-challenged insects with the eicosanoid-precursor polyunsaturated fatty acid, arachidonic acid. Low levels of arachidonic acid were detected in fat body phospholipids. These findings in adults of an exopterygote insect species with an unusual life history pattern broaden our hypothesis that eicosanoids mediate cellular immune reactions to bacterial infections in most, if not all, insects.  相似文献

We report on the presence of high proportions of arachidonic acid (20:4n-6) and eicosapentaenoic acid (20:5n-3) in the tissue lipids of adult fireflies, Photinus pyralis. Arachidonic acid typically occurs in very small proportions in phospholipids (PLs) of terrestrial insects, ranging from no more than traces to less than 1% of PL fatty acids, while 20:5n-3 is often missing entirely from insect lipids. Contrarily, 20:4n-6 made up approximately 21% of the PL fatty acids prepared from whole males and females, and from heads and thoraces prepared from males. Proportions of 20:4n-6 associated with PLs varied among tissues, including approximately 8% for male gut epithelia, 13% for testes, and approximately 25% for light organs and body fat from males. Substantial proportions of 20:5n-3 were also associated with PLs prepared from male firefly tissues, including 5% for body fat and 8% for light organs. Because 20:4n-6 and 20:5n-3 are precursors for biosynthesis of prostaglandins and other eicosanoids, we considered the possibility that firefly tissues might produce eicosanoids at exceptionally high rates. Preliminary experiments indicated otherwise. Hence, fireflies are peculiar among terrestrial insects with respect to maintaining high proportions of PL 20:4n-6 and 20:5n-3.  相似文献

1. Co‐occurrence of closely related predators in a prey‐limited habitat appears to contradict the principle of competitive exclusion, however it may be explained through indirect effects, niche shifts, and intraguild predation. 2. The interactions between sympatric tiger beetle Cicindela species were examined. Cicindela circumpicta is the largest of three species (C. circumpicta, C. togata, C. fulgida) found in saline habitats throughout central North America. The temporal occurrence of these species overlaps, as does their spatial occurrence on exposed salt flats of saline marshes. During field observations, exoskeletal remains of C. togata were found at the study site in Nebraska, U.S.A. 3. In laboratory trials, male C. circumpicta ate C. togata in 38% of trials and female C. circumpicta ate C. togata in 50% of trials (n = 24). 4. In the field, potential prey, consisting mainly of small flies, was found mostly in shaded conditions but tiger beetles differed significantly in shade use, with C. circumpicta spending 70% of the time in the shade compared with ≈ 20% for C. togata. Differential habitat use was not explained by maximum temperature tolerances, which did not differ between the species. 5. Laboratory trials established that both tiger beetle species consumed small prey (apterous Drosophila) but C. togata was more efficient at capturing winged Drosophila. 6. Foraging efficiency, as measured by the time taken for a C. togata to capture three prey items, decreased significantly in the presence of other tiger beetles, especially C. circumpicta. 7. These results are an indication that intraguild predation and induced changes in foraging behaviour operate in the ecology of adult tiger beetles.  相似文献

Carrion insect succession studies have historically used repeated sampling of single or a few carcasses to produce data, either weighing the carcasses, removing a qualitative subsample of the fauna present, or both, on every visit over the course of decomposition and succession. This study, conducted in a set of related experimental hypotheses with two trials in a single season, investigated the effect that repeated sampling has on insect succession, determined by the number of taxa collected on each visit and by community composition. Each trial lasted at least 21 days, with daily visits on the first 14 days. Rat carcasses used in this study were all placed in the field on the same day, but then either sampled qualitatively on every visit (similar to most succession studies) or ignored until a given day of succession, when they were sampled qualitatively (a subsample) and then destructively sampled in their entirety. Carcasses sampled on every visit were in two groups: those from which only a sample of the fauna was taken and those from which a sample of fauna was taken and the carcass was weighed for biomass determination. Of the carcasses visited only once, the number of taxa in subsamples was compared to the actual number of taxa present when the carcass was destructively sampled to determine if the subsamples adequately represented the total carcass fauna. Data from the qualitative subsamples of those carcasses visited only once were also compared to data collected from carcasses that were sampled on every visit to investigate the effect of the repeated sampling. A total of 39 taxa were collected from carcasses during the study and the component taxa are discussed individually in relation to their role in succession. Number of taxa differed on only one visit between the qualitative subsamples and the actual number of taxa present, primarily because the organisms missed by the qualitative sampling were cryptic (hidden deep within body cavities) or rare (only represented by very few specimens). There were no differences discovered between number of taxa in qualitative subsamples from carcasses sampled repeatedly (with or without biomass determinations) and those sampled only a single time. Community composition differed considerably in later stages of decomposition, with disparate communities due primarily to small numbers of rare taxa. These results indicate that the methods used historically for community composition determination in experimental forensic entomology are generally adequate.  相似文献

Many amphipod crustaceans exhibit precopulatory mate guarding. Field samples of the amphipod Gammarus pseudolimnaeus indicated that pairs were positively size assortative. Receptive individuals readily formed pairs in the laboratory and the latency to formation of precopulatory pairs was decreased under threat of predation. In addition, females and, under conditions of extreme danger, males that formed pairs were significantly smaller when under the threat of predation. Amphipods distinguished between chemical stimuli (aquarium water) from predatory and nonpredatory fishes and between chemical cues from fish predators (trout) that had recently eaten conspecific amphipods vs. those fed a control diet of pelleted commercial fish food. These data indicate that chemical stimuli associated with predators can influence reproductive behavior of amphipods. The results also suggest the hypotheses that: 1. search time may be costly in terms of probability of predation; and 2. small pairs may be safer from predation than larger pairs.  相似文献

The endangered American burying beetle, Nicrophorus americanus Olivier, was previously widespread throughout eastern North America. In the past century numbers of this beetle have drastically declined and currently remnant populations are known from only six states despite intensive surveying efforts conducted for the last nine years. Efforts aimed at discovering and managing remnant populations have been generally limited by a lack of knowledge concerning N. americanus biology. We used baited pitfall traps to define the range of the Gothenburg, Nebraska population of N. americanus. Using mark-recapture techniques, we estimate that the annual Gothenburg population consists of more than one thousand individuals, meeting the recovery plan criterion to become the third breeding population in the Midwest region. Beyond estimates of population size and range, we present novel data on seasonal and daily activity, sex ratio, age-grading and foraging distances. In 1995 and 1996, the Nebraska population was univoltine and female biased, with over-wintering mature beetles emerging in early June and teneral beetles emerging in August. Nocturnal activity was highest in the third and fourth hours following sunset but was not strongly correlated with temperature. During foraging, beetles travel up to six kilometers, but the majority of our recaptures occurred at distances of less than 0.5 km, suggesting that distances between traps be increased to ensure independence of sampling units. This information will allow future work on captive breeding, re-introduction and genetic studies.  相似文献

The larvae of the tiger beetle, Cicindela hirticollis Say, inhabit sandy shoreline areas that flood periodically. This species has declined over much of its range and at least one subspecies is near extinction, possibly as a result of human alteration of waterways. In addition to physiological tolerance for anoxia, the larvae have physical and behavioral adaptations to avoid drowning. We hypothesized that C. hirticollis larvae would exhibit behavioral responses to soil moisture change and flooding because, unlike most other tiger beetles, they frequently relocate their burrows. Our laboratory studies demonstrated that larvae select surface soil moisture levels of 7–50% saturation in which to dig new burrows. Within 96 h of immersion, most larvae abandon burrows and larvae do not form new burrows in darkness. Larvae may relocate when flooded, suggesting a previously undocumented mechanism for dispersal; however, dams often eliminate suitable habitat areas downstream, suggesting that this behavior may be detrimental in riverine populations. Because larvae move during daylight hours, they also are likely to suffer mortality from trampling due to human recreational activity.  相似文献

In this study, we compared survivorship, heat dissipation and biochemical features of anaerobiosis of two tiger beetle species (Coleoptera: Cicindelidae) exposed to anoxia. One species commonly experiences environmental immersion from rainfall and snowmelt (Cicindela togata), and the habitat of the other (Amblycheila cylindriformis) is not prone to flooding. The ancestral genus, A. cylindriformis, survives anoxia for only 2 days at 25 °C. In response to anoxia, these larvae immediately lose locomotory abilities, tissue concentrations of ATP fall precipitously within 12 h, and significant amounts of lactate are quickly produced. In contrast, C. togata larvae tolerate anoxia for 5 days. Heat dissipation is downregulated to a greater degree than that seen in A. cylindriformis (3.4% versus 14% of standard normoxic rate, respectively), the ability for locomotion is maintained and normoxic levels of ATP are defended for at least 24 h. Lactate is not accumulated until well into anoxic bout, and significant amounts of alanine are also produced. This study provides evidence that tiger beetles differ in physiological responses to anoxia, and that these differences are correlated with flooding risk and with species distribution. Insects exhibit a remarkable array of adaptations that allow them to handle more or less severe hypoxia associated with numerous aquatic and terrestrial habitats. We consider these habitats and then discuss physiological, behavioral and morphological mechanisms that facilitate insect life under oxygen deprivation. Actually or potentially hypoxic habitats include aquatic systems, flood-prone soils and burrows, intertidal zones, ice encasement and high altitudes. Some microhabitats, including dung, carrion, mammalian alimentary canals, grain and wood, also are subject to hypoxia. Adaptations to hypoxia include the ability to switch from aerobic to anaerobic metabolic pathways (with attendant generation of end products), the ability to drastically attenuate basal metabolic rates, altered behaviors and enlarged tracheal system volumes. Research into the biology of hypoxia seems to be progressing from early observations of the abilities of some insects to withstand exposure to hypoxia or anoxia through studies of organismal mechanisms operating in hypoxia to detailed investigations of cellular and intracellular signaling processes. Our hope is this essay will help crystallize the emergent picture of this area for those interested in contributing to future research.  相似文献

The Platte River caddisfly (Ironoquia plattensis Alexander and Whiles 2000) was recently described from a warm-water slough along the Platte River in central Nebraska and was considered abundant at the type locality. Surveys of 48 sites in 1999 and 2004 found eight additional sites with this species on the Platte River. The caddisfly was not found at the type locality in 2004 and one additional site in 2007, presumably because of drought conditions. Because of its apparent rarity and decline, the Platte River caddisfly is a Tier I species in Nebraska. For this project, surveys for the caddisfly were conducted at 113 new and original sites primarily along the Platte, Loup, and Elkhorn Rivers between 2009 and 2011. These surveys identified 30 new sites with the caddisfly. Larval densities were quantified at a subset of inhabited sites, and there was a large variation of densities observed. Seven sites on other Nebraska drainages were found to support morphologically similar caddisflies, presumably the Platte River caddisfly. Because of the discovery of populations outside the Platte River drainage, amplified fragment length polymorphism (AFLP) was used to determine the amount of genetic variability and breeding among sites on the Platte, Loup, and Elkhorn Rivers. Analysis of molecular variance (AMOVA) suggested moderate gene flow among the three river systems and that there was more genetic variation within populations than between populations. Differentiation, but not total divergence, was exhibited by the northernmost population from the Elkhorn River. Because it may be considered an indicator species and is vulnerable to ongoing habitat loss and degradation, all Platte River caddisfly populations should be conserved.  相似文献
