Biochemical characteristics and virulence factors were compared in 147 Aeromonas spp. isolated from patients with diarrhea and in 94 strains isolated from metropolitan water supplies in the same area during the same period. Fermentation of arabinose occurred with 58.5% of the environmental strains and 15% of the clinical isolates; 39.4% of the strains from water and 6.8% of the fecal isolates fermented salicin. The frequency of esculin hydrolysis was the same in both groups. Ninety-one percent of clinical isolates and 70.2% of environmental strains were enterotoxigenic and, except for four clinical isolates, all of these strains also produced hemolysins. Hemagglutination that was inhibited by fucose and mannose but not by galactose was found in 67% of the water isolates and 10.2% of the clinical strains. Although the distribution of several characteristics differs in clinical and environmental strains, many of the strains found in water have properties identical with those of the clinical isolates. We suggest that such strains may be potential enteric pathogens.  相似文献

Mass mortality events occur in natural and cultured communities of bivalve molluscs. The Pacific oyster, Crassostrea gigas, is a dominant species in many intertidal locations as well as an important aquacultured bivalve species, and for the last 50 years, adult oysters have suffered frequent and extreme mass mortality events during summer months. To investigate the molecular changes that precede these mortality events, we employed a novel nonlethal sampling approach to collect haemolymph samples from individual oysters during the period that preceded a mortality event. Microarray-based gene expression screening of the collected haemolymph was used to identify a mortality gene expression signature that distinguished oysters that survived the mortality event from those individuals that died during the event. The signature was cross-validated by comparing two separate episodes of mortality. Here, we report that near-mortality oysters can be distinguished from longer-lived oysters by the elevated expression of genes associated with cell death, lysosomal proteolysis, and cellular assembly and organization. These results show the potential utility of nonlethal sampling approaches for investigating the environmental causes of mortality in natural populations in the field, and for predicting when such events could occur and which individuals will be affected.  相似文献

Future phosphorus (P) scarcity and eutrophication risks demonstrate the need for systems‐wide P assessments. Despite the projected drastic increase in world‐wide fish production, P studies have yet to include the aquaculture and fisheries sectors, thus eliminating the possibility of assessing their relative importance and identifying opportunities for recycling. Using Norway as a case, this study presents the results of a current‐status integrated fisheries, aquaculture, and agriculture P flow analysis and identifies current sectoral linkages as well as potential cross‐sectoral synergies where P use can be optimized. A scenario was developed to shed light on how the projected 2050 fivefold Norwegian aquaculture growth will likely affect P demand and secondary P resources. The results indicate that, contrary to most other countries where agriculture dominates, in Norway, aquaculture and agriculture drive P consumption and losses at similar levels and secondary P recycling, both intra‐ and cross‐sectorally, is far from optimized. The scenario results suggest that the projected aquaculture growth will make the Norwegian aquaculture sector approximately 4 times as P intensive as compared to agriculture, in terms of both imported P and losses. This will create not only future environmental challenges, but also opportunities for cross‐sectoral P recycling that could help alleviate the mineral P demands of agriculture. Near‐term policy measures should focus on utilizing domestic fish scrap for animal husbandry and/or fish feed production. Long‐term efforts should focus on improving technology and environmental systems analysis methods to enable P recovery from aquaculture production and manure distribution in animal husbandry.  相似文献

Endemic helminthic infection is a major public-health problem and affects a large proportion of the world's population. In Australia, helminthic infection is endemic in Aboriginal communities living in tropical northern regions of the continent. Such infection is associated with nonspecific (polyclonal) stimulation of IgE synthesis and highly elevated total serum IgE levels. There is evidence that worm-infection variance (i.e., human capacity of resistance) and total serum IgE levels may be related to the presence of a major codominant gene. The beta chain of the high-affinity IgE receptor, Fc epsilon R1-beta, has been previously identified as a candidate for the close genetic linkage of the 11q13 region to IgE responses in several populations. We show a biallelic RsaI polymorphism in Fc epsilon R1-beta to be associated with total serum IgE levels (P = .0001) in a tropical population of endemically parasitized Australian Aborigines (n = 234 subjects). The polymorphism explained 12.4% of the total residual variation in serum total IgE and showed a significant (P = .0000) additive relationship with total serum IgE levels, across the three genotypes. These associations were independent of familial correlations, age, gender, racial admixture, or smoking status. Alleles of a microsatellite repeat in intron 5 of the same gene showed similar associations. The results suggest that variation in Fc epsilon R1-beta may regulate IgE-mediated immune responses in this population.  相似文献

Purpose

Expanding renewable energy production is widely accepted as a promising strategy in climate change mitigation. However, even renewable energy production has some environmental impacts, some of which are not (yet) covered in life cycle impact assessment (LCIA). We aim to identify the most important cause-effect pathways related to hydropower production on biodiversity, as one of the most common renewable energy sources, and to provide recommendations for future characterization factor (CF) development.

Methods

We start with a comprehensive review of cause-effect chains related to hydropower production for both aquatic and terrestrial biodiversity. Next, we explore contemporary coverage of impacts on biodiversity from hydropower production in LCA. Further, we select cause-effect pathways displaying some degree of consistency with existing LCA frameworks for method development recommendations. For this, we compare and contrast different hydrologic models and discuss how existing LCIA methodologies might be modified or combined to improve the assessment of biodiversity impacts from hydropower production.

Results and discussion

Hydropower impacts were categorized into three overarching impact pathways: (1) freshwater habitat alteration, (2) water quality degradation, and (3) land use change. Impacts included within these pathways are flow alteration, geomorphological alteration to habitats, changes in water quality, habitat fragmentation, and land use transformation. For the majority of these impacts, no operational methodology exists currently. Furthermore, the seasonal nature of river dynamics requires a level of temporal resolution currently beyond LCIA modeling capabilities. State-of-the-art LCIA methods covering biodiversity impacts exist for land use and impacts from consumptive water use that can potentially be adapted to cases involving hydropower production, while other impact pathways need novel development.

Conclusions

In the short term, coverage of biodiversity impacts from hydropower could be significantly improved by adding a time step representing seasonal ecological water demands to existing LCIA methods. In the long term, LCIA should focus on ecological response curves based on multiple hydrologic indices to capture the spatiotemporal aspects of river flow, by using models based on the “ecological limits to hydrologic alteration” (ELOHA) approach. This approach is based on hydrologic alteration-ecological response curves, including site-specific environmental impact data. Though data-intensive, ELOHA represents the potential to build a global impact assessment framework covering multiple ecological indicators from local impacts. Further, we recommend LCIA methods based on degree of regulation for geomorphologic alteration and a fragmentation index based on dam density for “freshwater habitat alteration,” which our review identified as significant unquantified threats to aquatic biodiversity.

The recovery of Aeromonas spp. from the unchlorinated water supply for a Western Australian city of 21,000 people was monitored at several sampling points during a period of 1 year. Membrane filtration techniques were used to count colonies of Aeromonas spp., coliforms, and Escherichia coli in water sampled before entry to service reservoirs, during storage in service reservoirs, and in distribution systems. Aeromonas spp. were identified by subculture on blood agar with ampicillin, oxidase tests, and the use of Kaper medium and then were tested for production of enterotoxins and hemolysins. During the same period, two-thirds of all fecal specimens sent for microbiological examination were cultured on ampicillin-blood agar for Aeromonas spp. Recovery of Aeromonas spp. from water supplies at distribution points correlated with fecal isolations and continued during autumn and winter. Coliforms and E. coli were found most commonly in late summer to autumn. This pattern differs from the summer peak of Aeromonas isolations both from water and from patients with Aeromonas spp.-associated gastroenteritis in Perth, Western Australia, a city with a chlorinated domestic water supply. Of the Aeromonas strains from water, 61% were enterotoxigenic, and 64% produced hemolysins.  相似文献
