Cytospora spp. and associated teleomorphic species (Ascomycota, Diaporthales, Valsaceae) are among the most common and widespread canker- and dieback-causing fungi on trees, shrubs and herbaceous plants worldwide. From specimens collected all over Iran a total of 114 isolates were morphologically identified, representing 20 Cytospora, one Leucostoma and five Valsa species from 38 plant species. Nine of the identified taxa were new records for Iran, and many new hosts were identified. The phylogenetic relationships of the Iranian strains, along with sequences of 13 reference strains from GenBank, were inferred from ITS1-5.8S-ITS2 nuclear rDNA sequences. Parsimony analysis established five distinct major clades and 12 subclades, which represented accepted species and genera. Some of these subclades corresponded to morphologically based taxonomic concepts of single Cytospora species, while others contained more than one morphospecies. Teleomorphic states were present in six subclades, and most clustered with the corresponding anamorphs. This suggests that morphological and phylogenetic species concepts overlap and that in most cases they are meaningful for correct species identification.  相似文献

This study was carried out using 155 monoconidial isolates collected from different areas of two major rice growing provinces in northern Iran, including 94 isolates from Guilan and 59 isolates from Mazandaran. Among 94 isolates from Guilan, 92 and two isolates recovered from rice and crabgrass (Digitaria sp.), respectively. All 61 rested isolates from Mazandaran were recovered from rice. All isolates were evaluated for in vitro sexual fertility and mating type status by pairing with Mat 1-1 and Mat 1-2 fertile standard hermaphrodite isolates including Br48 and Th12 (Mat 1-1) and KA9 and TH16 (Mat 1-2). Of 155 isolates, 98 (63.2%) were fertile and 57 (36.8%) were infertile and produced no perithecium when mated with standard isolates. Among 98 fertile isolates, 96 isolates were identified as Mat 1-1 and two isolates as Mat 1-2. All Mat 1-1 isolates were obtained from rice and two Mat 1-2 isolates obtained from crab grass. No Mat 1-2 isolate was identified from rice in this study. Both mating types were found in Guilan but all isolates recovered from Mazandaran were identified as Mat 1-1. Male fertility predominated in fertile Mat 1-1 and Mat 1-2 isolates from all sampling sites in northern Iran, and no female fertility was detected. This is the first report of existence of Mat 1-2 allele in Magnaporthe grisea population in Iran.  相似文献

Flexible biaxial goniometers are extensively used for measuring wrist positions and movements. However, they display an inherent crosstalk error. The aim was to evaluate the effect, of this error, on summary measures used for characterizing manual work. A goniometer and a torsiometer were combined into one device. An algorithm that effectively compensated for crosstalk was developed. Recordings from 25 women, performing five worktasks, were analyzed, both with and without compensation for crosstalk. The errors in the 10th, 50th and 90th percentiles of the flexion/extension distributions were small, on average <1 degrees. The ulnar/radial deviation distributions were weakly dependent on forearm position. The flexion/extension velocity measures were, for the 50th and 90th percentiles, as well as the mean velocity, consistently underestimated by, on average, 3.9%. For ulnar/radial deviation, the velocity errors were less consistent. Mean power frequency, which is a measure of repetitiveness, was insensitive (error <1%) to crosstalk. The forearm supination/pronation angular distributions were wider, and the velocities higher, than for the wrists. Considering wrist/hand exposure in epidemiologic studies, as well as for establishing and surveillance of exposure limits for prevention of work-related upper extremity musculoskeletal disorders, the crosstalk error can, when considering other errors and sources to variation, be disregarded.  相似文献

Though surface electromyography (EMG) has been widely used in studies of occupational exposure, its precision in terms of the variance between-days and between-subjects has seldom been evaluated. This study aimed at such an evaluation. Six women performed three different work tasks: 'materials picking', 'light assembly', and 'heavy assembly', repeated on 3 different days. EMG was recorded from m. trapezius, m. infraspinatus and the forearm extensors. Normalisation was made to a maximal (MVE), and a submaximal (RVE), reference contraction. Variance components between days (within subjects) and between subjects were derived for the 10th, 50th and 90th percentiles, as well as for muscular rest parameters. For the task 'heavy assembly', the coefficient of variation between days (CV(BD)) was 8% for m. trapezius (right side, 50th percentile, MVE normalised values). Larger variabilities were found for m. infraspinatus (CV(BD) 15%), and the forearm extensors (CV(BD) 33%). Between-subjects variability (CV(BS)) was greater, 16% for m. trapezius and 57% for m. infraspinatus, 29% for the forearm extensors. RVE normalisation resulted in larger CV(BD), while reducing CV(BS). The between-days and between-subjects variability may be used to optimise sampling strategy, and to assess the bias in epidemiological studies. The bias caused by measurement procedures per se is acceptable.  相似文献

During 2000--03, different areas in Zanjan, Golestan and Khorasan provinces were surveyed for the presence of olive dieback. Olive branches, leaves and roots showing typical symptoms and soil around the roots were collected for further study. Samples were surface-sterilized with sodium hypochlorite or ethanol and then cultured on PDA and Czapek media. Soil samples were diluted in ethanol-agar for fungal isolation and purification. Morphological characteristics of the fungal mycelium particularly phialide and spores identified the causal agent to be the soil-borne pathogen, Verticillium dahliae. The disease was present in all olive growing regions but it was severe in temperate and relatively humid regions such as Gorgan. Infection index of the disease varied between 5 to 30% with an average of 11.89+/-1.12 among various orchards in this area. The newly established orchards showed more infection than the older ones. A significant difference in disease incidence and severity were observed among olive cultivars of Michen, Roughani, Zard and Koronakei. The latter cultivar had the least amount of infection. Strains of V. dahliae isolated from olive trees had different morphological and pathogenicity characteristics. These strains had different growth rates in response to the optimum temperature of 20 or 25 degrees C. The number of fungal propagules per gram of air-dried soil ranged from 2 to 32 with an average number of 13.42+/-0.50. Regarding the number of propagules of V. dahliae in the soil and susceptibility of cultivars in the newly established orchards, it seems necessary to take serious control measures to prevent disease spread.  相似文献

The presence of Yersinia ruckeri in a French fish farm was investigated. Y. ruckeri was isolated mainly from algae and sediment samples rather than from water. Twenty-two Y. ruckeri isolates were obtained, and three strains were distinguished by enterobacterial repetitive intergenic consensus PCR amplification. These strains were able to adhere to solid supports. This characteristic was correlated with flagellum-mediated motility. Killing experiments showed that sessile cells were more resistant to oxolinic acid than their planktonic counterparts. Our results demonstrate that surface colonization of fish farm tanks by Y. ruckeri biofilms is a potential source of recurrent infection for extended periods of time.  相似文献

Saccharomyces cerevisiae and Candida shehatae were co-immobilized in an agar sheet which was introduced in an original two-chambered bioreactor asymmetrically fed in a batch mode with a mixture of glucose and xylose in a ratio of 35:15. The two sugars were consumed simultaneously. All glucose was fermented but only 20% of xylose. After incubation, yeast cells recovered from different areas of the agar sheet (close to, called Hi, and distant from, called Ho, the substrate chamber) were cultured as suspended cells in fresh culture medium provided with xylose or the sugar mixture. Xylose utilization by gel released Hi yeasts was significantly delayed compared to the Ho culture. Ethanol consumption by Hi yeasts in the two-substrate medium occurred after glucose exhaustion despite the presence of xylose. The waste medium resulting from incubation of the immobilized-cell structure inhibited xylose utilization by C. shehatae. Our results suggested that batch fermentation most probably favoured this incomplete xylose fermentation.  相似文献

Sclerotinia sclerotiorum is an important pathogen on canola. Due to the public concern over pesticide use, alternative methods of disease control, such as biological control, should be considered. Several bacterial strains were isolated from canola and soja plants. Inhibition of S. sclerotiorum by bacterial strains in vitro was assayed on PDA medium in dual culture test. Eight Pseudomonas sp. strains (PB-3, PB-4, PB-5, PB-6, PB-7, PB-8, PB-10 and PB-11) caused inhibition zone against 5. sclerotiorum hyphal growth. The biocontrol potential of the bacteria was tested in a plant assay. Disease suppression was investigated using a petal inoculation technique. Canola petals were pretreated with bacteria, and then inoculated with 5. sclerotiorum ascospores 24 h later. Greenhouse experiment showed that application of Pseudomonas sp. strains (1 x 10(8) cfu ml(-1)) effectively suppressed S. sclerotiorum (1 x 10(5) ascospores ml(-1)) on petals and all of them achieved significant (P<0.01) disease suppression. Fourteen days after inoculation, strain PB-3 had 88/7% disease control and strain PB-4 had 69/9% disease control. Result from all studies indicates PB-3 to be effective biocontrol against S. sclerotiorum of canola. PB-3, PB-4, PB-7, PB-8, PB-10 and PB-11 were identified as Pseudomonas fluorescens biovar III. PB-5 and PB-6 was identified as Pseudomonas fluorescens biovar II. Strains PB-3, PB-4, PB-6, PB-10 and PB-11 produced protease and HCN. Strain PB-5 produce protease; no HCN.  相似文献

Ascochyta blight, incited by Ascochyta rabiei (Pass.) Lab., is one of the most important fungal diseases that has been shown to cause significant yield losses on chickpeas in Kermanshah province, Iran. One of the most effective and reliable methods of controlling the disease has been the use of resistant cultivars worldwide. This requires the identification of the physiological races of the fungus in the region in advance. In this regard, pathogenicity of 30 isolates of A. rabiei was determined using 10 differential chickpea varieties (mean of 1-9 degree of disease in three replications) under greenhouse conditions. In this respect, isolates fell into four groups, whereas based on infection type three groups were identified using cluster analysis. Disease resistance of four chickpea cultivars, Hashem, Jam, ILC-482 and Bivanij, were evaluated against isolates from each of three groups separately and in combination in the greenhouse and field conditions (with 4 replications). The experiment was repeated twice with three replications in greenhouse. Chickpea cultivars were sprayed with fungal spore suspension (2x10(5) spores/ml). Results showed that Jam and Bivanij cultivars were susceptible under greenhouse as well as field conditions. Hashem was tolerant to group I and susceptible to groups II and III and in the field. ILC-482 was tolerant to groups II and I and susceptible to group III and the combination of groups under greenhouse condition.  相似文献
