Helminths express various carbohydrate-containing glycoconjugates on their surface, and they release glycan-rich excretion/secretion products that can be very important in their life cycles, infection and pathology. Recent evidence suggests that parasite glycoconjugates could play a role in the evasion of the immune response, leading to a modified Th2-polarized immune response that favors parasite survival in the host. Nevertheless, there is limited information about the nature or function of glycans produced by the trematode Fasciola hepatica, the causative agent of fasciolosis. In this paper, we investigate whether glycosylated molecules from F. hepatica participate in the modulation of host immunity. We also focus on dendritic cells, since they are an important target of immune-modulation by helminths, affecting their activity or function. Our results indicate that glycans from F. hepatica promote the production of IL-4 and IL-10, suppressing IFNγ production. During infection, this parasite is able to induce a semi-mature phenotype of DCs expressing low levels of MHCII and secrete IL-10. Furthermore, we show that parasite glycoconjugates mediate the modulation of LPS-induced maturation of DCs since their oxidation restores the capacity of LPS-treated DCs to secrete high levels of the pro-inflammatory cytokines IL-6 and IL-12/23p40 and low levels of the anti-inflammatory cytokine IL-10. Inhibition assays using carbohydrates suggest that the immune-modulation is mediated, at least in part, by the recognition of a mannose specific-CLR that signals by recruiting the phosphatase Php2. The results presented here contribute to the understanding of the role of parasite glycosylated molecules in the modulation of the host immunity and might be useful in the design of vaccines against fasciolosis.  相似文献

Abstract— Lucilia is a South American genus with 23 species restricted to disjunct areas in southeastern Brazil and along the Andes. Lucilia is a monophyletic group defined by the co-occurrence of six characters: herbaceous, alternate-leaved, pappus with scabrid bristles fused at the base into a ring, style-branches with sweeping hairs far down, capitula sessile, and aseptate-flagellate hairs. A dadogram is presented using 41 morphological and anatomical characters arranged into 26 transformation series. The polarity of character states was determined by outgroup comparison with the genus Berroa. The cladistic analysis showed extensive parallel evolution in a number of the more conspicuous characters and produced four unresolved trichotomies. However, basing the hierarchy of Lucilia on the branching pattern produced by cladistic analysis results in a more natural and predicitive classification. Lucilia is divided into three sections, Lucilia, Intermedieae (sect, nov), and Lucilioides [divided into two subsections, Subspicata (subsect. nov.) and Lucilioides]. The latter subsection is subdivided into two series, Lucilioides (ser. nov.) and Paralucilia. The Brazilian species of section Lucilia (acutijolia, linearifolia, ferruginea, tmentosa, recurva, nitens, and flagelliformis) form the most primitive group within the genus. The more derived species of the genus, section lucilioides (plicatifolia, catamarcensis, burkartii, subspkata, lopezmirandae, alpina, pickeringii, piptolepis, santamca, chilensis, schultzii, longifolia, radians, lehmanni, pusilla) are found in the Andes L. eriophora (section Intermedieae) from central Chile bridges these two groups. An explanation for the distribution of the genus is given, based on the ecology of the species in relation to theories of the geologic and climatic history of South America. The present pattern has been determined by the age, geographical range, and vicissitudes of the habitat in which each group occurs. In the Brazilian species group, the habitat is old, and has remained relatively stable since well before the Pleistocene. In the Andean species group, the habitat is young and has undergone numerous rapid alterations since its inception at the end of the Pliocene.  相似文献

The interactions of the targeting sequence of the mitochondrial enzyme ornithine transcarbamylase with phospholipid bilayers of different molecular compositions have been studied by high-sensitivity heating and cooling differential scanning calorimetry, high-sensitivity isothermal titration calorimetry, fluorescence spectroscopy, and electron microscopy. These studies indicate that the leader peptide interacts strongly with dipalmitoylphosphatidylcholine (DPPC) bilayer membranes containing small mole percents of the anionic phospholipids dipalmitoylphosphatidylglycerol (DPPG) or brain phosphatidylserine (brain PS) but not with pure phosphatidylcholines. For the first time, the energetics of the leader peptide-membrane interaction have been measured directly by using calorimetric techniques. At 20 degrees C, the association of the peptide with the membrane is exothermic and characterized by an association constant of 2.3 X 10(6) M-1 in the case of phosphatidylglycerol-containing and 0.35 X 10(6) M-1 in the case of phosphatidylserine-containing phospholipid bilayers. In both cases, the enthalpy of association is -60 kcal/mol of peptide. Additional experiments using fluorescence techniques suggest that the peptide does not penetrate deeply into the hydrophobic core of the membrane. The addition of the leader peptide to DPPC/DPPG (5:1) or DPPC/brain PS (5:1) small sonicated vesicles results in vesicle fusion. The fusion process is dependent on peptide concentration and is maximal at the phase transition temperature of the vesicles and minimal at temperatures below the phase transition.  相似文献

The tumour-inducing T-DNA gene 4 (T-cyt gene) of the nopaline Ti plasmid pTiC58 was cloned and introduced into tobacco cells by leaf disc transformation using Agrobacterium plasmid vectors. Tobacco shoots exposed to elevated cytokinin levels were unable to develop roots and lacked apical dominance. Using exogenously applied phytohormone manipulations we were able to regenerate morphologically normal transgenic tobacco plants which differed in endogenous cytokinin levels from normal untransformed plants. Although T-cyt gene mRNA levels, as revealed by dot-blot hybridization data, in these rooting plants were only about half those in primary transformed shoots the total amount of cytokinins was much lower than in crown gall tissue or cytokinin-type transformed shoots as reported by others. Nevertheless the cytokinin content in T-cyt plants was about 3 times greater than in control tobacco plants.Elevated cytokinin levels have been shown to change the expression of several plant genes, including some nuclear genes encoding chloroplast proteins. Our results show that the mRNA levels of chloroplast rbcL gene increase in cytokinin-type transgenic tobacco plants as compared with untransformed plants. Data obtained suggest that T-cyt transgenic plants are a good model for studying plant gene activity in different parts of the plant under endogenous cytokinin stress.  相似文献

Thymocytes, bone marrow lymphocytes, as well as lymphocytes from spleen, lymphoid nodes and peripheral blood were obtained from BALB/c mice. Subpopulations of BALB/c bone marrow T-lymphocyte precursors and immature (small) and mature (large) thymocytes, as established by the percentage of terminal deoxynucleotidyl transferase (TdT) and peanut agglutinin (PNA) positive cells, were obtained by centrifugation on discontinuous density gradients. The activities of N-acetyl-beta-glucosaminidase (NAG), beta-glucuronidase (BG), acid alpha-naphthyl acetate esterase (ANAE) and alpha-naphthyl acetate esterase (NAE) were determined by enzymatic assays of cell extracts of the diverse T-lymphocyte subpopulations, in order to follow their evolution with the maturation of the T-lymphocytes in the thymus. These activities were compared with that determined in lymphocytes from spleen, lymphoid nodes and peripheral blood. The glucidases BG and NAG and the esterases ANAE and NAE present a high decrease in their activities from bone marrow T-lymphocyte progenitors to immature thymocytes. BG, NAG and ANAE activities undergo an about 3-fold increase with the evolution of the thymocytes from small to large cells. Whereas the level of the NAE activity decreases (2-fold) with that evolution of the thymocytes. Lymphocytes from spleen and lymphoid nodes exhibit activities of the glucidases and, specially, the esterases marked by higher than those of thymocyte populations. Peripheral blood lymphocytes also present NAG, ANAE and NAE activities higher than in thymocytes, but their BG activity is lower.  相似文献

Summary Reliable and efficientRhizobium germplasm banks are essential for the development of research and for practical application. The second edition of the World Catalogue ofRhizobium Collections lists 64 institutions in 38 countries holding some 3000 effective, tested strains. MIRCENs Culture Collections are playing an important role in this way and in the dissemination of valuable strains for legume inoculation. Constraints for development are: adequate facilities and equipment, trained personnel, co-ordination of effort. Recommendations are: preparation of an inventory on technical aspects, new directives for training and for next edition of the Catalogue, establishment of specialized germplasm banks.
Resumen Los bancos de germoplasma deRhizobium son esenciales tanto para el desarrollo de la investigación como para el de las aplicaciones prácticas. La segunda edición del catálogo mundial de las colecciones deRhizobium cuenta con 64 instituciones repartidas en 38 países con un total de más de 3000 cepas de probada eficacia. Las colecciones de cultivos de los MIRCEN tienen un papel importante tanto en su valor intrínseco como en la distribución de cepas eficaces para la inoculación de leguminosas. Los principales problemas con los que se enfrenta su desarrollo son: facilidades adecuadas y equipamientos; personal cualificado y coordinación de esfuerzos. Las recomendaciones son: preparación de inventarios sobre aspectos técnicos; nuevas directrices para la formación de personal y para la edición de catálogos; establecimiento de bancos de germoplasma especializados.

Résumé Des collections deRhizobium fiables et efficaces sont essentielles à la fois pour la recherche et pour les applications agricoles. La deuxième édition du catalogue mondial des collections deRhizobium comporte 64 institutions, situées dans 38 pays, et environ 3.000 souches testées. Les collections de cultures des MIRCEn jouent un rôle important sur le plan scientifique et en ce qui concerne la distribution de souches utilisées pour l'inoculation des légumineuses. Leur développement est soumis à des difficultés concernant les installations et l'équipement, le personnel qualifié, et la coordination des efforts. Il est recommandé de faire l'inventaire des aspects techniques et de formuler de nouvelles directives pour la formation technique, pour la prépartion d'une nouvelle édition du catalogue, et pour l'établissement d'une banque de gènes spécialisée.

The effects on some pharmacological and enzymatic properties were determined following methylation of histidine at the enzymatic active site of the basic relatively toxic Najanigricollis and the acidic relatively non-toxic Najanajaatra phospholipases A2. Following methylation a very low residual enzymatic activity (0.4 -- 1% of control) was accompanied by a parallel loss in intraventricular lethality, anticoagulant potency, direct hemolytic action and ability to block directly and indirectly evoked contractions of the mouse phrenic nerve-diaphragm preparation. Since methylation does not impair the enzyme's ability to bind monomeric of micellar substrates or Ca2+, the results suggest that the pharmacologicallly active region of the molecule is different from the micellular substrate binding site but strongly influenced by the invariant histidine-48 located at the enzymatic active site.  相似文献
