The use of biomaterials or microorganisms in PAHs degradation had presented an eye-catching performance. Pleurotus eryngii is a white rot fungus, which is easily isolated from the decayed woods in the tropical rain forest, used to determine the capability to utilize naphthalene, a two-ring polycyclic aromatic hydrocarbon as source of carbon and energy. In the meantime, biotransformation of naphthalene to intermediates and other by-products during degradation was investigated in this study. Pleurotus eryngii had been incubated in liquid medium formulated with naphthalene for 14 days. The presence of metabolites of naphthalene suggests that Pleurotus eryngii begin the ring cleavage by dioxygenation on C1 and C4 position to give 1,4-naphthaquinone. 1,4-Naphthaquinone was further degraded to benzoic acid, where the proposed terepthalic acid is absent in the cultured extract. Further degradation of benzoic acid by Pleurotus eryngii shows the existence of catechol as a result of the combination of decarboxylation and hydroxylation process. Unfortunately, phthalic acid was not detected in this study. Several enzymes, including manganese peroxidase, lignin peroxidase, laccase, 1,2-dioxygenase and 2,3-dioxygenase are enzymes responsible for naphthalene degradation. Reduction of naphthalene and the presence of metabolites in liquid medium showed the ability of Pleurotus eryngii to utilize naphthalene as carbon source instead of a limited glucose amount.  相似文献

The hyaluronan (HA) synthase, PmHAS, and the chondroitin synthase, PmCS, from the Gram-negative bacterium Pasteurella multocida polymerize the glycosaminoglycan (GAG) sugar chains HA or chondroitin, respectively. The recombinant Escherichia coli-derived enzymes were shown previously to elongate exogenously supplied oligosaccharides of their cognate GAG (e.g. HA elongated by PmHAS). Here we show that oligosaccharides and polysaccharides of certain noncognate GAGs (including sulfated and iduronic acid-containing forms) are elongated by PmHAS (e.g. chondroitin elongated by PmHAS) or PmCS. Various acceptors were tested in assays where the synthase extended the molecule with either a single monosaccharide or a long chain (approximately 10(2-4) sugars). Certain GAGs were very poor acceptors in comparison to the cognate molecules, but elongated products were detected nonetheless. Overall, these findings suggest that for the interaction between the acceptor and the enzyme (a) the orientation of the hydroxyl at the C-4 position of the hexosamine is not critical, (b) the conformation of C-5 of the hexuronic acid (glucuronic versus iduronic) is not crucial, and (c) additional negative sulfate groups are well tolerated in certain cases, such as on C-6 of the hexosamine, but others, including C-4 sulfates, were not or were poorly tolerated. In vivo, the bacterial enzymes only process unsulfated polymers; thus it is not expected that the PmCS and PmHAS catalysts would exhibit such relative relaxed sugar specificity by acting on a variety of animal-derived sulfated or epimerized GAGs. However, this feature allows the chemoenzymatic synthesis of a variety of chimeric GAG polymers, including mimics of proteoglycan complexes.  相似文献

Compared to other melon types, Cantaloupe Charentais melons are highly aromatic with a major contribution to the aroma being made by aliphatic and branched esters. Using a transgenic line in which the synthesis of the plant hormone ethylene has been considerably lowered by antisense ACC oxidase mRNA (AS), the aliphatic ester pathway steps at which ethylene exerts its regulatory role were found. The data show that the production of aliphatic esters such as hexyl and butyl acetate was blocked in AS fruit and could be reversed by ethylene. Using fruit discs incubated in the presence of various precursors, the steps at which ester formation was inhibited in AS fruit was shown to be the reduction of fatty acids and aldehydes, the last step of acetyl transfer to alcohols being unaffected. However, treating AS fruit with the ethylene antagonist 1-methylcyclopropene resulted in about 50% inhibition of acetyl transfer activity, indicating that this portion of activity was ethylene-dependent and this was supported by the low residual ethylene concentration of AS fruit discs (around 2 microl l(-1)). In conclusion, the reduction of fatty acids and aldehydes appears essentially to be ethylene-dependent, whilst the last step of alcohol acetylation has ethylene-dependent and ethylene-independent components, probably corresponding to differentially regulated alcohol acetyltransferases.  相似文献

We have fabricated a bilayer heterojunction solar cell consisting of p-n active layers of naphthalenetetracarboxylic bis-benzimidazole, N-CON, and zinc phthalocyanine, ZnPc. Conjugated naphthalene derivative of naphthalene bis-benzimidazole was employed as an electron acceptor. Bilayer solar cells were produced by successive evaporation of zinc phthalocyanine, ZnPc, and naphthalene bis-benzimidazole, N-CON, on glass substrates coated with indium doped tin oxide, ITO/PEDOT:PSS/ZnPc/N-CON/Al. Photovoltaic characteristics of bilayer cell were measured under simulated AM 1.5 sun illumination (Voc = 0.50 V, Jsc = 2.11 mA/cm2, FF = 0.51, η = 0.54%). Efficiency of the cell was discussed in relation to energy gap and work functions of the thin layers.  相似文献

Conventional and microwave-assisted synthesis of ZnO nanorods have been performed with and without using PEG400. ZnO nanorods were synthesized with 50-250 nm of diameter which depends on the used surfactant and methods. Surfactant effects of PEG400 on the size and morphology of ZnO nanorods were investigated. The microwave method was compared to the conventional heating method. Morphologies were investigated by using scanning electron microscopy (SEM).  相似文献

Heparan sulfate carries a wide range of biological activities, regulating blood coagulation, cell differentiation, and inflammatory responses. The sulfation patterns of the polysaccharide are essential for the biological activities. In this study, we report an enzymatic method for the sulfation of multimilligram amounts of heparan sulfate with specific functions using immobilized sulfotransferases combined with a 3'-phosphoadenosine 5'-phosphosulfate regeneration system. By selecting appropriate enzymatic modification steps, an inactive precursor has been converted to the heparan sulfate having three distinct biological activities, associated with binding to antithrombin, fibroblast growth factor-2, and herpes simplex virus envelope glycoprotein D. Because the recombinant sulfotransferases are expressed in bacteria, and the method uses a low cost sulfo donor, it can be readily utilized to synthesize large quantities of anticoagulant heparin drug or other biologically active heparan sulfates.  相似文献

Norethisterone ethanate (200 mg every 84 days) and medroxyprogesterone acetate (150 mg every three months) were found to be completely effective in fertility control when started in the puerperium. Neither agent had any ill effect on the amount of milk or the duration of lactation. From the third month onward the three-hourly available milk and the infant weight gain per month were statistically higher in treated groups than in controls. Milk proteins showed a slight decrease in most groups, including the controls, owing to the low-protein diet.No important side effect was produced by these agents other than amenorrhoea.  相似文献

BackgroundIn Malaysia, dengue remains a top priority disease and usage of insecticides is the main method for dengue vector control. Limited baseline insecticide resistance data in dengue hotspots has prompted us to conduct this study. The present study reports the use of a map on the insecticide susceptibility status of Aedes aegypti and Aedes albopictus to provide a quick visualization and overview of the distribution of insecticide resistance.Method and resultsThe insecticide resistance status of Aedes populations collected from 24 dengue hotspot areas from the period of December 2018 until June 2019 was proactively monitored using the World Health Organization standard protocol for adult and larval susceptibility testing was conducted, together with elucidation of the mechanisms involved in observed resistance. For resistance monitoring, susceptibility to three adulticides (permethrin, deltamethrin, and malathion) was tested, as well as susceptibility to the larvicide, temephos. Data showed significant resistance to both deltamethrin and permethrin (pyrethroid insecticides), and to malathion (organophosphate insecticide) in all sampled Aedes aegypti populations, while variable resistance patterns were found in the sampled Aedes albopictus populations. Temephos resistance was observed when larvae were tested using the diagnostic dosage of 0.012mg/L but not at the operational dosage of 1mg/L for both species.ConclusionThe present study highlights evidence of a potential threat to the effectiveness of insecticides currently used in dengue vector control, and the urgent requirement for insecticide resistance management to be integrated into the National Dengue Control Program.  相似文献

Archaeobotanical samples from the Middle Bronze Age (MBA; c. 2000–1700 b.c.) city of Kanesh, excavated at the site of Kültepe in Kayseri Province, Turkey, preserve the charred shells of hazelnut (Corylus sp.). Hazelnut species do not naturally grow in the Kayseri area, being a native element of the broadleaf woodlands of Turkey’s Black Sea region, today home to a multi-million dollar international hazelnut export industry. The finds come from both the upper and lower city, being restricted to the Middle Bronze Age Karum level II, an occupation phase which saw the greatest development of the Assyrian trade network of which Kanesh was the administrative centre. This archaeobotanical discovery at Kültepe provides the earliest direct evidence for trade in hazelnuts in the region, probably imported on a small scale as luxury items facilitated by the Assyrian trade network. It also provides independent support for historical claims that hazelnut was traded at Kanesh based on the analysis of cuneiform tablets.  相似文献

The morphological and molecular studies provide greater taxonomic resolution for the scleractinian coral identification. The Euphylliidae corals are among the scleractinian family for which their corallite and polyp morphologies have been examined for species identification. However, knowledge on the molecular study for coral identification in Malaysia is very limited. Therefore, this study aimed to examine the morphological structures and phylogenetic analyses for six Euphylliidae coral species using the mitochondrial gene of cytochrome oxidase subunit I (COI). The results showed that the Euphylliidae corals are present under both “complex” and “robust” coral clades as supported by many researchers. The result also revealed that the species phylogeny of Euphylliidae corals is in concordances with its morphological structures of corallites. It can be concluded the combination between morphological structures and phylogenetic analyses provide more accurate identification than relying on morphological study alone. Hence, it provides a future direction for the scleractinian research progress in species identification.  相似文献
