In this work, we describe a novel pulse mode shear horizontal-surface acoustic wave (SH-SAW) polymer coated biosensor that monitors rapid changes in both amplitude and phase. The SH-SAW sensors were fabricated on 36 degrees rotated Y-cut X propagating lithium tantalate (36 YX.LT). The sensitivity of the device to both mass loading and visco-elastic effects may be increased by using a thin guiding layer of cross-linked polymer. Two acoustic modes are excited by the electrodes in this crystalline direction. Metallisation of the propagation path of the 36 YX.LT devices allows the two modes to be discriminated. Successive polymer coatings resulted in the observation of resonant conditions in both modes as the layer thickness was increased. Using the 36 YX.LT devices, we have investigated the application of a novel pulse mode system by sensing a sequence of deposition and removal of a biological layer consisting of vesicles of the phospholipid POPC. A continuous wave system was used to verify the accuracy of the pulse mode system by sensing a series of poly(ethylene glycol) (PEG) solutions. The data clearly demonstrates the ability of the 36 YX.LT pulse mode system to provide rapid measurements of both amplitude and phase for biosensing applications.  相似文献

1. Exposure to a combined forced swimming–confinement stress resulted in a decrease in insulin-like growth factor II (IGF-II) mRNA levels in the whole brain (without the cerebellum) and in the isolated brain areas of the cerebral cortex, the hippocampus, and the cerebellum.2. In an effort to elucidate the neurotransmitter systems involved in this stress-induced decrease, animals were injected prior to exposure to the stress, with either propranolol, diazepam, or MK-801.3. Administration of diazepam or MK-801 did not affect the stress-induced decrease in IGF-II mRNA in any of the three brain areas examined.4. Administration of propranolol prior to the exposure to the stress inhibited the stress-induced decrease in IGF-II mRNA in the cerebellum. Propranolol had no such effect in the cerebral cortex or the hippocampus.5. Our results suggest that in the cerebellum, the stress-induced decrease in IGF-II mRNA is mediated by 2-adrenergic receptors.  相似文献

The fungus Cunninghamella bainieri effects the oxidative N-demethylation of aminopyrine, O-demethylation of 4-nitroanisole and anisole, the aryl hydroxylation of anisole, aniline, and naphthalene, and the reduction of nitro and azo groups. The hydroxylation of 4-[2H]-anisole and 2-[2H]-anisole proceeds with migration and retention of isotopic hydrogen (NIH shift). The above reactions and the formation of the trans-dihydrodiol of naphthalene and the incorporation of oxygen-18 from 18O2 into the trans-dihydrodiol and hydroxylated anisole are characteristic of reactions catalyzed by the cytochrome P450 monooxygenases of hepatic microsomes. The product ratios in these hydroxylations are very similar to those obtained using liver microsomes providing further evidence that the C. bainieri monooxygenase enzymes are similar to the liver monooxygenases. Furthermore, an epoxide hydrase enzyme similar to that present in hepatic microsomes must also be present in C. bainieri.  相似文献

We measured the intrinsic viscosity of very small synthetic DNA molecules, of 20-395 base pairs, and incorporated them in a nearly complete picture for the whole span of molecular weights reported in the literature to date. A major transition is observed at M approximately 2 × 10(6) . It is found that in the range of approximately 7 × 10(3) ≤ M ≤ 2 × 10(6) , the intrinsic viscosity scales as [η] approximately M(1.05) , suggesting that short DNA chains are not as rigid as generally thought. The corresponding scaling for the range of 2 × 10(6) ≤ M ≤ 8 × 10(10) is [η] approximately M(0.69) . A comparison of our results with existing equations, for much narrower data distributions, is made, and the agreement is very satisfactory considering the huge range of data analyzed here. Experimental concerns such as the effect of ionic strength, polydispersity, temperature, and shear rate are discussed in detail. Some issues concerning the Huggins coefficient, polymer chain stiffness, and the relationship between the Mark-Houwink constants K, α are also presented; it is found that log K = 1.156 - 6.19α.  相似文献

Glutamic acid decarboxylase (GAD) enzyme activity was measured in synaptosomes prepared from the hypothalamus, the hippocampus, the striatum and the cerebral cortex of control, adrenalectomized and rat exposed to a thermal stress. Adrenalectomy caused a statistically significant decrease in the enzyme activity in the striatum, while it had no effect in the other three brain areas. On the other hand, exposure to the thermal stress resulted in a dramatic increase of GAD specific activity in all brain areas examined. We have studied the formation of histone Hv1-DNA complexes using an acoustic biosensor and AFM imaging. Our results show that DNA and histone molecules aggregate into amorphous accumulations which form a compact rigid layer on the sensor’s surface. By measuring changes in the acoustic wave amplitude, it was possible to titrate surface bound DNA with Hv1 and discriminate between DNA molecules of different size and shape. From the kinetic analysis of real time data, Keq was found equal to 3 × 105 M−1.  相似文献

TGF-beta activated kinase 1 (TAK1) is a MAP kinase kinase kinase (MAPKKK) that has been shown to function downstream of BMPs and TGF-beta (J. Biol. Chem. 275 (2000) 17647; EMBO J. 17 (1998) 1019; Science 270 (1995) 2008), as well as in the interleukin-1 (IL-1) signaling pathway (J. Biol. Chem. 276 (2001) 3508; Nature 398 (1999) 252). Using immunohistochemistry (IHC), we demonstrate that TAK1 is expressed ubiquitously during early development. At mid-gestation, TAK1 expression becomes more restricted, with high levels seen specifically during development of diverse organs and tissues including the nervous system, testis, kidney, liver and gut. Additionally, TAK1 expression is seen in the developing lung and pancreas. Our results suggest that TAK1 may play multiple roles in mouse development.  相似文献

MOTIVATION: The application of microarray chip technology has led to an explosion of data concerning the expression levels of the genes in an organism under a plethora of conditions. One of the major challenges of systems biology today is to devise generally applicable methods of interpreting this data in a way that will shed light on the complex relationships between multiple genes and their products. The importance of such information is clear, not only as an aid to areas of research like drug design, but also as a contribution to our understanding of the mechanisms behind an organism's ability to react to its environment. RESULTS: We detail one computational approach for using gene expression data to identify response networks in an organism. The method is based on the construction of biological networks given different sets of interaction information and the reduction of the said networks to important response sub-networks via the integration of the gene expression data. As an application, the expression data of known stress responders and DNA repair genes in Mycobacterium tuberculosis is used to construct a generic stress response sub-network. This is compared to similar networks constructed from data obtained from subjecting M.tuberculosis to various drugs; we are thus able to distinguish between generic stress response and specific drug response. We anticipate that this approach will be able to accelerate target identification and drug development for tuberculosis in the future.
