A detailed analysis of the composition and properties of hydrophobic nuclei and microclusters in pancreatic ribonuclease A (RNase A) has been carried out. Distance calculations for all noncovalently bonded atoms revealed that the average number of nonpolar contacts between a side chain of an amino acid and its neighbors is substantially larger if it involves hydrophobic residues rather than nonhydrophobic ones. However, the difference decreased when the number of contacts per nonpolar group and/or atom were calculated. Three main nuclei and five microclusters were identified, and their quantitative parameters were calculated. These nuclei include hydrophobic residues with a substantial number of nonpolar contacts with the environment (Phe 8, Phe 120, Phe 46, Tyr 25, Tyr 97, Ile 107, Leu 35, Ile 81, Val 54, Val 108, Met 29, Met 30). Hydrophobic nuclei of RNase A differ in shape and in composition, in the number of intranuclear contacts and of associated residues, as well as in their internal mobility. All eight cysteine residues are involved in nonpolar interactions with amino acid residues of hydrophobic nuclei. Active site amino acid residues of RNase A form a noncovalent contact network comprised of themselves, as well as of many conserved residues from hydrophobic nuclei. Sequence alignment with some other members of the RNase A family of proteins shows remarkable similarity in positions and in conservation of the main nonpolar residues, comprising cores of two (out of three) hydrophobic nuclei. A correlation was shown to exist between the average density of contacts for side-chain atoms and the number of amino acids to be found in the appropriate positions in the sequences of related mammalian ribonucleases. However, there are certain amino acid positions in the third, smaller nucleus, which are highly variable within the family. Taking into account that this nucleus is composed of residues belonging to different elements of the secondary structure, it is likely that the mutual orientation of these elements can be somehow different for these proteins.  相似文献

Patterns of medium-term (decades) postfire succession of a mire ecosystem were studied using multiproxy paleoecological analysis. The resistance of mire ecosystems featuring thick peat deposits to fires due to surface and patchy incineration has been noted. It was found that, in favorable climatic conditions (i.e., sufficient precipitation), the vegetation cover recovers and the vertical growth and development towards oligotrophication are resumed within a medium-term period.  相似文献

An experiment was proposed and accomplished that was based on the hypothesis of the dissociation of the luciferase-luciferin complex in photoexcitation. A pump-probe experiment was performed with the use of picosecond laser pulses and was based on the effect of quenching of enzyme tryptophan fluorescence caused by luciferin binding. A photoinduced increase of the tryptophan fluorescence intensity was detected. Experimental results were interpreted on the basis of the assumptions on photoinduced dissociation of the luciferin-luciferase complex and Forster energy transfer from tryptophan to luciferin. Under the assumption on the photoinduced dissociation and stationary quenching of tryptophan fluorescence the rate of propagation of the conformational changes in the protein caused by the complex dissociation was estimated to be >20 m/s.  相似文献

Appropriate management of contemporary environments requires knowledge of their long-term history. We use palaeoecological data to explore how contemporary forest-steppe environments have been shaped by climate change and human impacts through the Holocene using the western Mid-Russian Upland as a case-study. Our paper presents new reconstructions of Mid- and Late Holocene climate, vegetation dynamics and local environmental change based on pollen, plant macrofossil and testate amoeba records from a site at Selikhovo (Mid-Russian Upland, Russia). Eutrophic fen vegetation dominated by Phragmites australis developed around 6800 cal year BP and has been resilient to episodes of local burning and variable input of mineral material through the Holocene. New and previously-published data show that the boundary between broadleaf forest and steppe occupied a similar position to present during the period 7000–4800 cal year BP, despite a warmer and drier climate, but shifted to the south following climate cooling and an increase in precipitation from 4800–2500 cal year BP. A subsequent decline in woodland cover was caused by both climate change and human impacts, with human activity becoming increasingly significant over the last two millennia. Prior to major human disturbance (about 1700 cal year BP) the landscape was dominated by mixed broadleaf-pine forests with some spruce covering about 60 % of the study area. Our results emphasize the variability of steppe-forest habitats over long time periods and the need to consider human impacts and climate change when setting targets for habitat conservation.  相似文献

New pollen and plant macrofossil data, backed by radiocarbon dates, from the Kulikovo battlefield area in the forest-steppe region of the Upper Don River basin (central part of European Russia) indicate that the area was covered by mosaic vegetation in the second half of the Holocene. Steppe communities dominated during the mid—late Atlantic (7.2–5.7?cal.?kyr b.p.) and early Subatlantic (2.7–2.4?cal.?kyr b.p.), while forest-steppe dominated during the Subboreal (2.7–5.7?cal.?kyr b.p.), middle and late Subatlantic (2.4?cal.?kyr b.p. – present). Climatic reconstructions based on these data show that landscape dynamics in the region were most probably driven by changes in effective moisture: an excess of precipitation over evaporation. Even small reductions in annual precipitation, accompanied by a rise in summer temperatures by 1–3°C above present values, were sufficient to increase the proportion of steppe communities within this landscape complex, and also probably resulted in higher frequencies of wildfires. Signals of anthropogenic disturbance of vegetation are clearly pronounced in the pollen and plant macrofossil records since the middle Atlantic. However, human-induced changes in the vegetation remain subtle until the medieval period.  相似文献

Women with hypoestrogenemia either through secondary amenorrhea of unknown etiology, post-menopausal period or removed ovaries due to benign tumor were studied to determine if there is a relationship between the hormone state of women and their skeletal and periodontal tissues. Results indicated that women with hypoestrogenemia showed more pronounced changes in periodontium tissue as compared to normal women. These women also showed evidence of periodontium injury. These results are discussed along with implications for treatment and preventive measures.  相似文献

The mouse c-Ki-ras protooncogene promoter contains a homopurine-homopyrimidine domain that exhibits S1 nuclease sensitivity in vitro. We have studied the structure of this DNA region in a supercoiled state using a number of chemical probes for non-B DNA conformations including diethyl pyrocarbonate, osmium tetroxide, chloroacetaldehyde, and dimethyl sulfate. The results demonstrate that two types of unusual DNA structures formed under different environmental conditions. A 27-bp homopurine-homopyrimidine mirror repeat adopts a triple-helical H-DNA conformation under mildly acidic conditions. This H-DNA seems to account for the S1 hypersensitivity of the promoter in vitro, since the observed pattern of S1 hypersensitivity at a single base level fits well with the H-DNA formation. Under conditions of neutral pH we have detected Z-DNA created by a (CG)5-stretch, located adjacent to the homopurine-homopyrimidine mirror repeat. The ability of the promoter DNA segment to form non-B structures has implications for models of gene regulation.  相似文献
