Three different rhabdovirus isolates, eggplant mottled dwarf virus (EMDV), tomato veinyellowing virus (TVYV) and a hitherto undefined isolate from tomato in Portugal (Tom-P) were compared by two different serological techniques, immunosorbent electron microscopy (ISEM) and electro-blot-immunoassay (EBIA). Antisera were prepared either against purified virus or against partially purified virus preparations extracted by a one-step procedure. Even the partially purified virus preparations yielded antisera that allowed unequivocal identification of two of the five structural virus proteins, G and N, in EBIA and were suitable for specific trapping of complete virus particles on electron microscope grids. With both serological techniques identical results were obtained indicating a close serological relationship between the three isolates tested. Cross-reactions between the G proteins could be deduced from heterologous trapping of complete virus particles in ISEM. The cross-reactions for the G proteins were substantiated by the EBIA-results which indicated in addition cross-reactions between the remaining three major structural virus proteins, N, M1, and M2, but also revealed significant differences in the molecular weights of the G and M1 proteins between TVYV and the other two isolates. The results obtained indicate that Tom-P is serologically closely related and in respect of protein molecular weights identical to EMDV, and that TVYV is not a separate virus but rather a strain of EMDV. 相似文献

A virus reported earlier to cause a green mosaic disease of eggplant in Nigeria was studied in more detail. Its filamentous particles with a normal length of 820 nm reacted in immunoelectron microscopical tests strongly with the homologous antiserum and less strongly with antisera to dioscorea green banding mosaic, groundnut eyespot, zucchini yellow mosaic viruses and to a tomato potyvirus isolate from Taiwan. No reactions were seen with antisera to 25 other potyviruses. Several new host plants were identified. Infected cells contained cylindrical inclusions with scrolls and short curved laminated aggregates and clusters of small vesicles with electron-dense content. Host range and serological reactivities differentiate the virus for which the name eggplant green mosaic virus is suggested from all potyviruses so far known. 相似文献

Isometric virus-like particles (VLPs) have been purified from cucumber leaf tissue. Three dsRNA segments with estimated molecular weights of 1.8, 1.1 and 1.0 × 106d have been isolated from VLPs occurring in CsCl density gradient fractions but were also readily detected in preparations from as little as 1 g of fresh leaf tissue. The VLPs resemble dsRNA containing cryptic viruses and have been named Cucumis sativus cryptic virus (CsCV). 相似文献

A number of virus isolates were obtained from ditches and drainage canals in the Palatinate grapevine-growing area (‘Rheinpfalz’) in West Germany. By means of the immunoelectron microscopical decoration and the agar gel double diffusion tests these isolates were identified as petunia asteroid mosaic virus, carnation ringspot virus and tobacco necrosis virus strain D, respectively. 相似文献

In groundnut rosette diseased groundnut plants collected near Zaria, Nigeria, a luteovirus was detected by ELISA and ISEM. In ELISA only beet western yellows virus antiserum reacted, while in ISEM luteovirus particles were trapped by antisera beet western yellows virus, potato leafroll virus, pea leafroll virus and barley yellow dwarf virus. The data are in agreement with the interpretation that the assistor of groundnut rosette virus is possibly a member of the luteovirus group. 相似文献

Samples of commercially grown Impatiens holstii from several sources in Minnesota contained a carlavirus which serologically was closely related, if not identical with Helenium virus S described earlier in Germany. The two viruses have similar experimental host ranges and caused similar cytopathogenic effects. After mechanical transmission the Impatiens isolate produced less severe symptoms in Impatiens than the Helenium isolate. Only the latter isolate was transmitted by Myzus persicae in the non-persistent manner. 相似文献

The Tomato spotted wilt virus (TSWV) encoded NSm movement protein facilitates cell-to-cell spread of the viral genome through structurally modified plasmodesmata. NSm has been utilized as bait in yeast two-hybrid interaction trap screenings. As a result, a protein of unknown function, called At-4/1, was isolated from an Arabidopsis thaliana GAL4 activation domain-tagged cDNA library. Using polyclonal antibodies against bacterially expressed At-4/1, Western blot analysis of protein extracts isolated from different plant species as well as genome database screenings showed that homologues of At-4/1 seemed to be encoded by many vascular plants. For subcellular localization studies, At-4/1 was fused to green fluorescent protein, and corresponding expression vectors were used in particle bombardment and agroinfiltration assays. Confocal laser scannings revealed that At-4/1 assembled in punctate spots at the cell periphery. The protein accumulated intracellularly in a polarized fashion, appearing in only one-half of a bombarded epidermal cell, and, moreover, moved from cell to cell, forming twin-structured bodies seemingly located at both orifices of the plasmodesmatal pore. In coexpression studies, At-4/1 colocalized with a plant virus movement protein TGBp3 known to reside in endoplasmic reticulum-derived membrane structures located in close vicinity to plasmodesmata. Thus, At-4/1 belongs to a new family of plant proteins capable of directed intra- and intercellular trafficking. 相似文献

Structural abnormalities include various types of translocations, inversions, deletions, duplications and isochromosomes. Structural abnormalities of the Y chromosome are estimated to affect less than 1% of the newborn male population and are particularly hazardous for male reproductive function. The objective of this study was to characterize a group of patients with structural abnormalities of the Y chromosome. All patients who visited our laboratory between 2007 and 2010 underwent cytogenetic investigations. Among these, we detected 26 patients with structural abnormalities of the Y chromosome. To confirm the structural Y chromosome alterations, we used special bandings, FISH and multiplex PCR to detect Y chromosome microdeletions. Of the 26 patients presented here, 11 had an isodicentric Y chromosome, 7 had an inversion, 3 had a translocation, 2 had a derivative, 2 had a Yqs and 1 had a deletion. Sixteen were diagnosed with azoospermia, 8 as normal fertile males and 1 as a man who was unable to donate semen due to mental retardation. One of the patients having 45,X/46,X,idic(Y) was reported to be phenotypically female with primary amenorrhea and without uterus. Deletions of the AZFbc region were correlated with the sperm concentration (p < 0.05), but no correlation with the levels of FSH, LH, testosterone, prolactin and estradiol were found. The present report shows that the precise identification of structural Y chromosome aberrations may be clinically important for genetic counseling and assisted reproductive technology treatment. 相似文献
