Crossing experiments within the genus Allium have been carried out for many years. Usually, with the aim of widening genetic variation, edible Alliaceae such as bulb onion, Japanese bunching onion, leek, garlic and shallot have been crossed with each other or with distant vegetable Allium crops. Interspecific hybridization, especially with wild relatives is considered as the best way of introgression desirable traits to the crops. By using sexual or somatic hybridization, many important characters, such as: disease and pest resistance, important metabolites and cytoplasmic male sterility have been transferred to other Allium crops. This review summarises some aspects of the directions of interspecific hybridization in edible Alliaceae and the significance and perspectives of using these hybrids in breeding programmes.  相似文献

To clarify the effects of storage temperature on potato components and acrylamide in chips, tubers from five cultivars were stored at various temperatures (2, 6, 8, 10, and 18 degrees C) for 18 weeks, and the contents of sugars, free amino acids in tubers, and acrylamide in chips after frying were analyzed. At temperatures lower than 8 degrees C, the contents of reducing sugars increased markedly in all cultivars, with similar increases in the acrylamide level and dark brown chip color. Free amino acids showed little change at the storage temperatures tested and varied within certain ranges characteristic of each cultivar. The contents of reducing sugars correlated well with the acrylamide level when the fructose/asparagine molar ratio in the tubers was <2. When the fructose/asparagine ratio was >2 by low-temperature storage, the asparagine content, rather than the reducing sugar content, was found to be the limiting factor for acrylamide formation.  相似文献

Changes in the sugar and amino acid contents of potato tubers during short-term storage and the effect on the acrylamide level in chips after frying were investigated. The acrylamide content in chips began to increase after 3 days of storage at 2 degrees C in response to the increase of glucose and fructose contents in the tubers. There was strong correlation between the reducing sugar content and acrylamide level, R(2)=0.873 for fructose and R(2)=0.836 for glucose. The sucrose content had less correlation with the acrylamide content because of its decrease after 4 weeks of storage at 2 degrees C, while the reducing sugar in potato tubers and the acrylamide in chips continued to increase. The contents of the four amino acids, i.e., asparatic acid, asparagine, glutamic acid and glutamine, showed no significant correlation with the acrylamide level. These results suggest that the content of reducing sugars in potato tubers determined the degree of acrylamide formation in chips. The chip color, as evaluated by L* (lightness), was correlated well with the acrylamide content.  相似文献

Targeted delivery of therapeutics possesses the potential to localize therapeutic agents to a specific tissue as a mechanism to enhance treatment efficacy and abrogate side effects. Antibodies have been used clinically as therapeutic agents and are currently being explored for targeting drug-loaded nanoparticles. Peptides such as RGD peptides are also being developed as an inexpensive and stable alternative to antibodies. In this study, cyclo(1,12)PenITDGEATDSGC (cLABL) peptide was used to target nanoparticles to human umbilical cord vascular endothelial cell (HUVEC) monolayers that have upregulated intercellular cell-adhesion molecule-1 (ICAM-1) expression. The cLABL peptide has been previously demonstrated to possess high avidity for ICAM-1 receptors on the cell surface. Poly( dl-lactic-coglycolic acid) nanoparticles conjugated with polyethylene glycol and cLABL demonstrated rapid binding to HUVEC with upregulated ICAM-1, which was induced by treating cells with the proinflammatory cytokine, interferon-gamma. Binding of the nanoparticles could be efficiently blocked by preincubating cells with free peptide suggesting that the binding of the nanoparticles is specifically mediated by surface peptide binding to ICAM-1 on HUVEC. The targeted nanoparticles were rapidly endocytosed and trafficked to lysosomes to a greater extent than the untargeted PLGA-PEG nanoparticles. Verification of peptide-mediated nanoparticle targeting to ICAM-1 may ultimately lead to targeting therapeutic agents to inflammatory sites expressing upregulated ICAM-1.  相似文献

Intact ovarian follicles, obtained from untreated and human chorionic gonadotropin (HCG) treated Japanese yellowtail Seriola quinqueradiata during different maturational stages, were incubated with radioactive [3H]pregnenolone, [3H]17‐hydroxyprogesterone or [14C] androstenedione and steroid metabolites identified by thin layer chromatography (TLC) followed by recrystallization to constant specific activity. In untreated late vitellogenic (0 h) follicles, androstenedione was the major product with smaller amounts of testosterone and oestradiol‐17α. In post‐vitellogenic (12 h post‐injection) intact follicles, androstenedione predominated, and although testosterone and oestradiol‐17α were not produced, there were small amounts of 17, 20β‐dihydroxy‐4‐pregnen‐3‐one (17,20β‐P) and 17,21‐dihydroxy‐4‐pregnene‐3, 20‐dione (11‐deoxycortisol). In HCG‐treated fish, a steroidogenic shift resulted in the disappearance of testosterone and oestradiol‐17 coinciding with the appearance of 17, 20β‐P. During early and late final oocyte maturation FOM (24 and 36 h post‐injection), there was a five‐ to seven‐fold increase in the production of 17, 20β‐P, whereas production of 11‐deoxycortisol remained almost the same. During FOM, in addition to 17,20β‐P, its 5β‐reduced metabolite, 17,20β‐dihydroxy‐5β‐pregnan‐3‐one (5β‐17,20β‐P) was synthesized, suggesting a decrease in maturation‐inducing 17,20β‐P activity. 17, 20β,21‐Trihydroxy‐4‐pregnen‐3‐one (20β‐S) was not synthesized by ovarian fragments in Japanese yellowtail at any maturational stage. The metabolites identified on TLC during FOM were tested to evaluate their maturation‐inducing activity in an in vitro bioassay. Of the steroids tested, 17,20β‐P was the most effective inducer of germinal vesicle breakdown (GVBD), followed by 5β‐17,20β‐P. Timely synthesis of 17,20β‐P immediately prior to and during FOM as well as its great potency in inducing GVBD in vitro supports the evidence for a physiological role of 17,20β‐P as a maturation‐inducing hormone in Japanese yellowtail.  相似文献

The radical scavenging activity of Japanese edible seaweeds was screened by the DPPH (1-diphenyl-2-picrylhydrazyl) assay to evaluate the DPPH radical scavenging activity in organic extracts. The fresh brown alga Hijikia fusiformis showed the strongest DPPH radical scavenging activity, followed by Undaria pinnatifida and Sargassum fulvellum. The major active compound from Hijikia fusiformis in its acetone extract was identified as fucoxanthin by 13C-NMR spectroscopy.  相似文献

We synthesized cyclic disulfide-bonded (i, i+4) peptides with various net positive charges (+2-+5) from linear peptides derived from the alpha helical domain of Tenecin 1, an insect defensin, and investigated the effect of the intradisulfide bridge (i, i+4) on hydrophobicity, secondary structure, leakage activity and binding activity for large unilamellar vesicles, antimicrobial activity, and hemolytic activity. Intradisulfide bridge formation of the peptides resulted in the increase of amphiphilicity and hydrophobicity. Cyclic forms of the peptides did not deeply penetrate into PG/PC (1:1, mole ratio) large unilamellar vesicles and had a decreased lipid membrane perturbation activity for PG/PC LUVs. When the peptides interacted with PG/CL (2:1, mole ratio) LUVs, cyclic peptides with a high net positive charge (+4-+5) showed similar binding affinities and leakage activities for vesicles to those of linear forms, whereas cyclic peptides with a low net positive charge (+2-+3) exhibited lower leakage activity than their linear forms. CD spectra indicate that the intradisulfide bridge (i, i+4) provided little conformational constraint to linear peptides in buffer solution but resulted in the decrease of alpha helicity of the peptides in lipid membrane mimic conditions. The cyclic peptide with the highest net positive charge had a similar antibacterial activity to that of the linear peptide, whereas the cyclic peptides with a low net positive charge (+3-+4) exhibited lower antibacterial activity than their linear forms. The cyclic peptides of an appropriate net charge showed more potent activities against some bacteria than those of linear forms under high salt conditions.  相似文献

Bisphenol A was oxidized to monoquinone and bisquinone derivatives by Fremy's salt, a radical oxidant, though salcomine and alkali did not catalyze the oxidation by molecular oxygen. Bisphenol A, bisphenol B, and 3,4'-(1-methylethylidene)bisphenol were converted to their monoquinone derivatives in the presence of tyrosinase at 25 degrees C at pH 6.5, but not to the bisquinone derivatives under these conditions.  相似文献

Ozone appeared to inhibit growth and caused the death of gram negative and gram positive tested bacteria: Escherichia coli, Salmonella sp., Staphylococcus aureus and Bacillus subtilis. Bacterial cultures at 10(3), 10(4), 10(5), 10(6), and 10(7) cfu/ml dilution were exposed to 0.167/mg/min/L of ozone at different time intervals (0, 5, 10, 15, 30, 60, 90, 120, and 150 min). Cell viability was observed in all types of tested bacteria at 10(3), 10(4), 10(3) cfu/ml within 30 min after ozone exposure. However, cell inactivation was not significantly observed at concentrations of 10(6), 10(7) cfu/ml even after an exposure of 150 min. Ultrastructural changes of treated bacteria showed deformation, rough damage and surface destruction revealed by scanning electron microscopy. Some bacterial cells showed collapsed and shrunken patterns within 60 min and severe rupture and cellular lysis after 90 min of ozone treatment. This study supports the proposed mechanism of the bacteria inactivation by ozone that caused cell membrane destruction and finally lysis reaction. Thus, the precaution of using ozone as a biocide should be used to address appropriate concentrations of bacterial contamination in water.  相似文献

Acrylamide in potato chips made from tubers stored at 2 or 20 degrees C for two weeks after harvest was analyzed by GC-MS. The acrylamide level in the former chips was higher than ten times of that in the latter, which was highly correlated with both glucose and fructose levels in the tubers.  相似文献
