Substitution of Milk for Serum in the Production of Human Leukocyte Interferon

The presence of serum in suspensions of Sendai-induced human leukocytes is necessary for the synthesis of significant amounts of interferon. Very little interferon is obtained from serum-free suspensions. Cow's milk or milk casein can substitute for serum in the production of high yields of human leukocyte interferon.     

Toxicity of interferon.

The effects of partially purified human leucocyte interferon (PIF) and of a preparation purified by passage twice through a monoclonal antibody affinity chromatography column (NK21F) were compared with those of a control solution in healhty volunteers. After intramuscular injections both interferon preparations caused rises in pulse rate and body temperature, changes in circulating white cell counts, and various unpleasant symptoms, the most common of which were headache, malaise, and fever. Slightly lower doses of NK21F were given, and this was reflected in lower peak serum concentrations. Mean symptom scores, however, were not lower after NK21F than after PIF. Local inflammatory reactions eight hours after intradermal inoculations of these interferons were similar. Purification of interferon using a monoclonal antibody does not reduce the facets of its activity considered in this study. They are therefore inherent in the leucocyte interferon type selected by the antibody.     

Purified interferon as protection against rhinovirus infection.

In a double-blind placebo-controlled study a preparation of human leucocyte interferon purified by affinity chromatography using a monoclonal antibody and applied by repeated nasal sprays reduced the incidence and severity of colds in volunteers challenged with human rhinovirus 9. Although interferon itself caused some symptoms, these were minor compared with the clinical colds. Interferon activity was still detectable in nasal washings as long as 26 hours after the last dose in about half the volunteers on active treatment.     

Regulatory effect of interferon on T cells in vitro.

Purified human lymphocyte interferon (PIF) was added to mixed lymphocyte cultures; DNA synthesis was measured and killer-cell generation was determined. At certain concentrations, PIF ingibited proliferation, but at the same time increased the killer efficiency of the resultant culture cells. It is suggested that lymphokines, apart from their normal mediator function, may play a role as regulators of the generating immune response.     

Praealbonemertes whangateaunienses n. gen. and n. sp., a new heteronemertean from New Zealand

Abstract

A new genus and species of heteronemertean, Praealbonemertes whangateaunienses n. gen. and n. sp., is described and illustrated. The species is characterised by inter alia a cephalic lacuna with strands of longitudinal muscle fibres, a proboscis with three muscle layers, and a well-developed muscle plate dorsal to the foregut and anterior intestine. The material was collected in New Zealand.     

Interferon crosses blood-cerebrospinal fluid barrier in monkeys.

Interferon was detected in the cerebrospinal fluid (CSF) of monkeys injected iv or im with 30 million units of human leukocyte interferon. The im injection maintained a long-lasting plateau at about 1/30th of the corresponding level of interferon in the serum. Interferon injected into the serum. Interferon injected into the cerebrospinal canal was cleared from CSF at a similar rate as it disappeared from blood after iv administration of a high dose. A relatively stable serum level was maintained for 12-24 hr after the injection of interferon into the CSF space.
