Abstract.  1. Theoretical models predict that ovipositional decisions of parasitoid females should lead to the selection of the most profitable host for parasitoid development. Most parasitoid species have evolved specific adaptations to exploit a single host stage. However, females of the aphid hyperparasitoid Syrphophagous aphidivorus (Mayr) (Hymenoptera: Encyrtidae) display a unique and atypical oviposition behaviour by attacking either primary parasitoid larvae in live aphids, or parasitoid pupae in dead, mummified aphids.
2. In the laboratory, the correlation between host suitability and host preference of S. aphidivorus on the host Aphidius nigripes Ashmead parasitising the aphid Macrosiphum euphorbiae (Thomas) was investigated.
3. The relative suitability of the two host stages was determined by measuring hyperparasitoid fitness parameters (survival, development time, fecundity, sex ratio, and adult size of progeny), and calculating the intrinsic rate of population increase ( r m). Host preference by S. aphidivorus females and the influence of aphid defence behaviour on host selection was also examined.
4. Hyperparasitoid offspring performance was highest when developing from hosts in aphid mummies and females consistently preferred this host to hosts in parasitised aphids. Although aphid defensive behaviour may influence host selection, it was not a determining factor. Ecological and evolutionary processes that might have led to dual oviposition behaviour in S. aphidivorus are discussed.  相似文献

U.v.-visible-absorption and e.p.r. spectroscopy were used to study the type 2 and type 3 copper centres in the mercury derivative of laccase. After treatment with peroxide the mercury derivative of laccase exhibits a fully developed absorption band at 330 nm (delta epsilon = 2900 +/- 100 M-1.cm-1, which is characteristic of type 3 copper in the oxidized state. In addition, there is a weak ligand-field absorption at 740 nm (epsilon = 380 +/- 30 M-1.cm-1), which can be assigned to the type 3 pair. Because the e.p.r. spectrum of the type 2 copper is well resolved in the case of the mercury derivative of laccase, for the first time we have been able to observe spectroscopic evidence for a pH-dependent structural transition that has been invoked to explain the kinetics of enzyme reduction [Andréasson & Reinhammar (1979) Biochim. Biophys. Acta 568, 145-156]. According to the e.p.r. data the pKa lies in the range 6-7, and comparisons with a model compound show that the spectral changes can plausibly be interpreted in terms of the deprotonation of a water molecule in the co-ordination sphere of the type 2 copper.  相似文献

1. The wasp Megascolia flavifrons stings larvae of the stag beetle Oryctes nasicornis on the ventral side of all segments, except the last three, which do not contain nerve ganglia. 2. Experiments indicate a central rather than a peripheral action of the venom. 3. From pharmacological analysis it is concluded that the venom does not contain cholinergic or serotonergic activity, but contains histamine- and bradykinin-like substances. 4. Pepino mosaic virus (PepMV) has become an important viral disease of greenhouse tomatoes worldwide. The ability of bumble‐bees (Bombus impatiens), used for pollination, to acquire and transmit PepMV was investigated, and the prevalence of PepMV in plants and bumble‐bees in commercial tomato greenhouses was determined. PepMV infection in plants was determined using enzyme‐linked immunosorbent assay, while in bumble‐bees direct real‐time PCR was used. In the first experiment, the bumble‐bees were exposed for 14 days to PepMV‐infected plants. After 14 days, almost all bumble‐bees were PepMV positive both in the hive (78.5 ± 17.5%) and in the flowers (96.3 ± 3.6%). In the second experiment, bumble‐bees were released into a greenhouse with both PepMV‐infected source plants and healthy non‐infected target plants for 14 days. At the end of the experiment, 61.0 ± 19.5% of the bees collected from the hive and 83.3 ± 16.7% of the bees sampled from the flowers were PepMV positive. Bumble‐bees transmitted PepMV from the infected to the healthy non‐infected tomato plants. Two weeks after bumble‐bee release, the virus was detected in leaf, fruit and flower samples of formerly healthy plants. After 6 weeks, the percentage of PepMV positive samples from the target plants increased to 52.8 ± 2.8% of the leaves and 80.6 ± 8.4% of the fruits. In the control greenhouse without bumble‐bees, the target plants did not become infected. Based on the infection levels in flowers, fruits and leaves, the PepMV infection occurred possibly first in the pollinated flowers, and then spread from the fruit that developed from the flowers to other parts of the plant. In commercial greenhouses where PepMV was present, 92–100% of the plants and 88–100% of the bumble‐bees were PepMV positive. No infected plant samples were found in the control commercial greenhouse, but a small number of bumble‐bees (10%) tested PepMV positive.  相似文献

The morphology (mainly prosthecae length), ultrastructure, and antenna pigment composition of the green sulfur bacterium Prosthecochloris aestuarii changed when grown under different light intensities. At light intensities of 0.5 and 5 micromol quanta m(-2) s(-1), the cells had a star-like morphology. Prosthecae, the characteristic appendages of the genus Prosthecochloris, were 232 nm and 194 nm long, respectively. In contrast, when grown at 100 micromol quanta m(-2) s(-1), these appendages were shorter (98 nm) and the cells appeared more rod-shaped. Transmission electron microscopy revealed a significant decrease in the cell perimeter to area ratio and in the number of chlorosomes per linear microm of membrane as light intensity increased. In addition to these morphological and ultrastructural responses, Prosthecochloris aestuarii exhibited changes in its pigment composition as a function of light regime. Lower specific pigment content and synthesis rates were found in cultures grown at light intensities above 5 micromol quanta m(-2) s(-1). A blue shift in the bacteriochlorophyll (BChl) c Q(y) absorption maximum of up to 17.5 nm was observed under saturating light conditions (100 micromol quanta m(-2) s(-1)). This displacement was accompanied by changes in the composition of BChl c homologs and by a very low carotenoid content. The morphological, ultrastructural and functional changes exhibited by Prosthecochloris aestuarii revealed the strong light-response capacity of this bacterium to both high and low photon-flux densities.  相似文献

To test the possibility of inorganic carbon limitation of the marine unicellular alga Emiliania huxleyi (Lohmann) Hay and Mohler, its carbon acquisition was measured as a function of the different chemical species of inorganic carbon present in the medium. Because these different species are interdependent and covary in any experiment in which the speciation is changed, a set of experiments was performed to produce a multidimensional carbon uptake scheme for photosynthesis and calcification. This scheme shows that CO2 that is used for photosynthesis comes from two sources. The CO2 in seawater supports a modest rate of photosynthesis. The HCO is the major substrate for photosynthesis by intracellular production of CO2 (HCO+ H+→ CO2+ H2O → CH2O + O2). This use of HCO is possible because of the simultaneous calcification using a second HCO, which provides the required proton (HCO+ Ca2+→ CaCO3+ H+). The HCO is the only substrate for calcification. By distinguishing the two sources of CO2 used in photosynthesis, it was shown that E. huxleyi has a K½ for external CO2 of “only” 1.9 ± 0.5 μM (and a Vmax of 2.4 ± 0.1 pmol·cell−1·d−1). Thus, in seawater that is in equilibrium with the atmosphere ([CO2]= 14 μM, [HCO]= 1920 μM, at fCO2= 360 μatm, pH = 8, T = 15° C), photosynthesis is 90% saturated with external CO2. Under the same conditions, the rate of photosynthesis is doubled by the calcification route of CO2 supply (from 2.1 to 4.5 pmol·cell−1·d−1). However, photosynthesis is not fully saturated, as calcification has a K½ for HCO of 3256 ± 1402 μM and a Vmax of 6.4 ± 1.8 pmol·cell−1·d−1. The H+ that is produced during calcification is used with an efficiency of 0.97 ± 0.08, leading to the conclusion that it is used intracellularly. A maximum efficiency of 0.88 can be expected, as NO uptake generates a H+ sink (OH source) for the cell. The success of E. huxleyi as a coccolithophorid may be related to the efficient coupling between H+ generation in calcification and CO2 fixation in photosynthesis.  相似文献

Marigold (cv. Lemon Gem), castor bean, ornamental pepper (cv. Black Pearl and Purple Flash), gerbera daisy (cv. Festival), feverfew, and sunflower (cv. Choco Sun) were evaluated for their suitability as banker plants (BP) for Orius insidiosus (Hemiptera: Anthocoridae) in commercial greenhouses. Oviposition, egg hatch, nymphal development to adulthood, and population increase were quantified in laboratory trials. Assessments of oviposition and egg hatch indicated that all plants tested were equally accepted by O. insidiosus. Nymphal development to adulthood and survival tests indicated that gerbera may be a suitable BP as survival was the highest (58.1 %), whereas marigold would not be an acceptable BP as only 10.7 % of nymphs survived to adulthood. Nymphal development time differed by only one day among all plants. In greenhouse cage experiments, Purple Flash pepper supported the greatest population growth over a ten week period. Based on the combined results from all tests, Purple Flash pepper appears to have the greatest potential as a BP species for O. insidiosus.  相似文献

The aggregation of amyloid-β (Aβ) is postulated to be the crucial event in Alzheimer’s disease (AD). In particular, small neurotoxic Aβ oligomers are considered to be responsible for the development and progression of AD. Therefore, elimination of thesis oligomers represents a potential causal therapy of AD. Starting from the well-characterized d-enantiomeric peptide D3, we identified D3 derivatives that bind monomeric Aβ. The underlying hypothesis is that ligands bind monomeric Aβ and stabilize these species within the various equilibria with Aβ assemblies, leading ultimately to the elimination of Aβ oligomers. One of the hereby identified d-peptides, DB3, and a head-to-tail tandem of DB3, DB3DB3, were studied in detail. Both peptides were found to: (i) inhibit the formation of Thioflavin T-positive fibrils; (ii) bind to Aβ monomers with micromolar affinities; (iii) eliminate Aβ oligomers; (iv) reduce Aβ-induced cytotoxicity; and (v) disassemble preformed Aβ aggregates. The beneficial effects of DB3 were improved by DB3DB3, which showed highly enhanced efficacy. Our approach yielded Aβ monomer-stabilizing ligands that can be investigated as a suitable therapeutic strategy against AD.  相似文献

Escape of 14CO2 from the reaction mixture into the gas phase may seriously affect the accuracy of in vitro measurement of vitamin K-dependent carboxylase activity (and probably that of other carboxylases as well). In this paper we describe the effect of (a) the volume of the test tubes in which the reaction is performed, (b) the addition of an excess of NaH12CO3 in parallel with standard amounts of NaH14CO3, and (c) the incubation temperature. In this way optimal conditions are defined and used for the carboxylation of various peptide and protein substrates. It is shown that both a prosequence and an internal recognition site contribute to the effective recognition of a substrate by carboxylase. The maximal efficiency of carboxylation was 1-2% with substrates lacking both signals and 20-50% if only one was present. This indicates the need for developing peptide substrates containing both recognition signals for vitamin K-dependent carboxylase.  相似文献
