A synthetic version of the cry1Ab gene from Bacillus thuringiensis (Bt) was introduced into white spruce (Picea glauca) by microprojectile bombardment. A plasmid carrying the cry1Ab gene, driven by a ubiquitin (maize) promoter, was co-transferred with a plasmid containing the gus–nptII fusion gene as a screenable selection marker. Molecular analysis of the transgenic lines showed a high level (more than 90%) of co-integration of the cry1Ab gene with the screenable marker. A wide range of expression levels of the cry1Ab gene and corresponding endotoxin was obtained. Accumulation of the Cry1Ab protein was evaluated in embryogenic tissue, the needles of somatic seedlings and in the needles of 5-year-old field-grown trees of individual lines. Laboratory and field insect feeding trials suggest that several spruce transgenic lines were lethal to spruce budworm larvae.  相似文献

We compared the effects of Manduca sexta allatotropin (Manse-AT) on the rate of in vitro juvenile hormone (JH) biosynthesis by the corpora allata (CA) of different-aged virgin females from migrant (Quebec) and non-migrant (Azores) populations of the armyworm, Pseudaletia unipuncta, as well as from early- and late-calling lines selected from the Quebec population. There was a significant age x strain interaction, with the observed rates of JH biosynthesis in early adult life closely reflecting strain-specific differences in the age at onset of calling. In considering data for all ages combined, treatment of CA with Manse-AT resulted in a significant increase in the rate of JH biosynthesis for all but the Late strain, although significant differences for this strain were detected at certain ages. The CA of females from the Azores strain showed the strongest stimulation, with those of 0- and 1-day-old individuals displaying a singularly high degree of sensitivity. Selection for early- and late-calling lines resulted in significant differences in the temporal patterns of JH biosynthesis but did not markedly affect the sensitivity of the CA to Manse-AT. These findings are discussed within the context of the age-related differences observed in the rates of in vitro JH biosynthesis and JH haemolymph titers previously reported in comparisons of the Quebec and Azorean strains of the true armyworm.  相似文献

Seventy transgenic tissue lines (translines) of three spruce species ( Picea mariana, P. glauca and P. abies) were characterized with respect to the integration pattern of the gus (beta-glucuronidase) gene, and the level of GUS activity was determined in 81 lines. The majority of the P. mariana translines (18 lines of 22) integrated multicopies of the transgene, whereas mostly single integrations were detected in the other two species. The activity levels of GUS varied widely among the individual translines of P. mariana, and there was a strong indication that the logarithm of GUS activity increased with the number of gus copies ( P=0.0003) in lines with one to five known insertions (uncensored). The average level of GUS activity, in lines that integrated one gene copy, was the highest in white spruce followed by black spruce and Norway spruce (22.7, 16.5 and 6.3 nmol 4-methylumbelliferone min(-1 )mg(-1 )protein, respectively).  相似文献

Maturation of five embryogenic lines of Pinus strobus L. was tested on media with various sugars and sources of organic nitrogen, and solidified with two gellan gum concentrations (0.6 and 1.0%). Mature somatic embryo production was more abundant at 1.0% gellan gum than at 0.6%. Complex combinations of amino acids had little effect on mature embryo production of most tested embryogenic lines. Increasing glutamine concentration of the maturation medium from 1.7 to 7.3 g l−1 was beneficial to one embryogenic line. Increasing sucrose concentration or substituting part of the sucrose with mannitol or sorbitol had variable effects on somatic embryo maturation depending on the embryogenic line. A medium with 88 mM sucrose plus 175 mM sorbitol solidified with 1.0% gellan gum produced high numbers of somatic embryos in four out of five embryogenic lines tested. Several factors affecting somatic embryogenesis (SE) in Pinus sylvestris from self- and cross-pollinated seed families were studied with the aim of producing large quantities of clonal plants. Somatic embryogenesis initiation from zygotic embryos was improved on a medium with lower than standard concentrations of 2,4-dichlorophenoxyacetic acid (2.2 vs. 9.5 μM) and 6-benzyladenine (2.2 vs. 4.5 μM). On this medium, initiation rates of four controlled crosses, including one self-cross, varied from 3% to 25%. Among the maturation factors tested, the concentration of abscisic acid (ABA 80, 120 μM) had no significant effect on the production of mature somatic embryos when the medium contained 0.1 M sucrose. When sucrose concentration was 0.2 M, however, 1.4 times more mature somatic embryos were produced on medium with 80 μM compared with 120 μM ABA. Under our best maturation conditions, mature somatic embryos accumulated amounts of storage proteins that were similar to the amounts in mature zygotic embryos. Activated charcoal exerted a beneficial effect on mature somatic embryo production of 24-week-old cultures; there was no evidence of such an effect in 8-week-old cultures. Thirty-seven embryogenic lines from a self-cross and an out-cross were chosen for clonal plant production. Highly embryogenic lines produced mature somatic embryos that were more likely to convert to plants than those from less embryogenic lines. After 4 months of growth in a shade house, plantlet survival rates exceeded 70% for 31 lines out of 35. This report describes an improved method for accelerated production of large quantities of Scots pine for clonal tests.  相似文献

In this study, several improvements and simplifications of SE protocols in Pinus pinaster (Ait.), a species of economic importance in the regions of Western Europe, are described. These improvements pertained to all stages of SE including high initiation frequencies in eight control pollinated seed families, relatively high somatic embryo maturation yield when cells were coated with particles of activated charcoal and a rapid production of plants directly in a shade house. The SE initiation frequency from isolated zygotic embryos was high (up to 100%) and plants were produced from 11 embryogenic lines representing all crosses. Based on these results, the estimated number of somatic embryos required to produce 1,000 plants varied from slightly more than the required number of plants to more than double this number depending on the line. Such an estimate is critical in developing plant production strategy when a number of embryogenic lines are considered for production of clonal plants.  相似文献

The effects of N, P, K and the vesicular-arbuscular mycorrhizal fungusGigaspora calospora (Nicol. and Gerd.) Gerdemann and Trappe on root colonization, spore production, dry biomass and shoot mineral content of onion (Allium cepa L. cv. Autumn spice) were investigated. All combinations of presence or absence of N, P, K and inoculum were tested. Nitrogen fertilization stimulates root colonization, and spore production is higher on plants fertilized with K. Phosphorus fertilization reduces root colonization and spore production. Biomass of endomycorrhizal plants is higher than that of non-mycorrhizal plants. Yield increases due to P are larger when plants are not fertilized with additional N. When plants are fertilized with either N, P or K they absorb more N, P or K, respectively. Some interactions between N, P, and K have an effect on tissue P and Ca content. Optimal ratio of available nutrients in the soil is essential for the host plant-VAM fungus system to be most efficient.  相似文献

Summary Calcium alginate beads containingLactococcus lactis cells were used for three batch fermentations of milk or a commercially available growth medium (Gold Complete, Nordica) with the aim of producing concentrated cultures. Repeated fermentations did not significantly increase bead CFU counts which were between 3.3–7.8×1010 CFU/g. During the second and third fermentations, which lasted 6 h each, the bead populations decreased if the incubation was extended over 2 h. There was cell release from the beads. Fermentation media and fermentation time all had an effect on free cell counts, but none of these factors statistically interacted. Free cell counts were higher at the end of fermentations 2 and 3 than in the first fermentation and approximately 50% of the population was in the free state. Free cell counts were higher when the beads were incubated in Gold complete than in milk. Although the total bacterial population of a standard free cell fermentation was always higher than those having immobilized cells, immobilized cell technology did enable the production of dense cultures.  相似文献

The effect of simultaneous modification of medium composition and growth conditions on the production of Lactococcus lactis subsp. cremoris biomass in calcium alginate beads was studied by the response surface method. Statistical methods of data analysis for unbalanced experiments are illustrated. The media tested were whey, whey supplemented with yeast extract and/or meat extract, milk, and the commercial medium Gold Complete (Nordica). Fermentations were performed at 23°C under pH control (5.6, 6.0, 6.4, or 6.8). In one complete series, 1% CaCO3 was added to the growth media. There were strong interactions between CaCO3 and media, CaCO3 and pH level, and CaCO3, media, and pH level. In media with CaCO3, all first-order interactions between media, pH, and sampling time were significant. The addition of CaCO3 increased cell counts in whey-meat extract medium, but no significant difference was found with the other media. Uncoupling between growth and acidification occurred between 16 and 22 h. Highest counts were obtained on milk and Gold Complete (6 × 1010/g). In CaCO3-containing media, pH influenced cell counts only in whey and in Gold Complete (pH 5.6 and 6.0 giving the best results); pH also influenced the bead mass obtained at the end of the fermentation. Biomass production in alginate gels is proposed as a method of obtaining concentrated cell suspensions without centrifugation or filtration.  相似文献
