Six Arcanobacterium haemolyticum strains isolated from six patients of two hospitals in Denmark were identified phenotypically, also including matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis, and by genotypic methods. The latter were performed by sequencing 16S rDNA and glyceraldehyde 3-phosphate dehydrogenase encoding gene gap and by amplification of an A. haemolyticum specific region of 16S–23S rDNA intergenic spacer region and 23S rDNA. The six A. haemolyticum strains were further investigated for the presence of seven potential virulence genes encoding arcanolysin, phospholipase D, hemolysin A, CAMP factor family protein, collagen binding protein, neuraminidase A and neuraminidase H which appeared to be present in two (seven virulence genes), two (six virulence genes) and two strains (four virulence genes), respectively. The phenotypic and genotypic properties described in the present study might help to reliably identify and further characterize A. haemolyticum isolated from human patients, a species which seems to be of increasing importance.  相似文献

Increased generation of oxidants and (or) reduced endogenous antioxidant defense mechanisms are associated with the etiology of diabetic vascular complications. The aim of the present study was to evaluate whether curcumin supplementation increases the vasodilatory effect of cilostazol in streptozotocin induced diabetic rat aorta. Cumulative addition of cilostazol caused concentration-dependent relaxations of thoracic aorta rings. The sensitivity and the maximal response to cilostazol were significantly higher in control than those in diabetic animals. Treatment with curcumin in control rats increased the sensitivity to cilostazol. Further, in aortic rings from diabetic rats treated with curcumin, the responses to cilostazol were significantly increased in comparison to the response in aorta from untreated diabetic rats. It can be conclude, that curcumin increases the cilostazol-induced vasodilation in diabetic rat aorta.  相似文献

The objective of the study was to determine the effects of repeated bouts of supramaximal exercise on plasma adiponectin, interleukin (IL)-6 and tumor necrosis factor (TNF)-α levels in sedentary men. Fourteen healthy, nonsmoking, and sedentary men aged between 18.4 and 21.4 years participated in the study. All the subjects performed 5 Wingate tests (WTs) with 75 g per kilogram body weight load with 2-minute intervals between the tests. Blood samples were collected at preexercise, immediately after, 15 and 60 minutes after the fifth WT. Serum and plasma samples were stored at -80°C until the time of analysis for myoglobin, adiponectin, IL-6, and TNF-α. Plasma adiponectin levels decreased, whereas IL-6 levels increased postexercise compared with that preexercise. The TNF-α levels were not changed with supramaximal exercise. In conclusion, repeated bouts of supramaximal exercise cause an inflammatory response in exercised muscle and increase in plasma IL-6 levels and decrease in adiponectin concentrations.  相似文献

We recently reported an unconventional mechanism by which miRNAs inhibit HIV-1 viral production. This occurs when miRNAs bind nonspecifically to the viral structural protein Gag, interfering with viral RNA-mediated Gag assembly at the plasma membrane. Consequently, misassembled viral complexes are redirected into the endocytic pathway where they are delivered to lysosomes for degradation. In this study, we demonstrate that autophagy is a critical mediator of the viral degradation pathway and that this pathway is not HIV-1 specific. Misassembled viral complexes were found to colocalize extensively with LC3 and p62 in late endosomes/lysosomes, demonstrating a convergence of autophagy with functional degradative compartments. Knocking down autophagosome formation machineries reduced this convergence, while treatment with autophagy-inducer rapamycin enhanced the convergence. Furthermore, similar autophagy-dependent nonspecific miRNA inhibition of murine leukemia virus (MLV) assembly was shown. Overall, these results reveal autophagy as a crucial regulator of the retroviral degradation pathway in host cells initiated by nonspecific miRNA-Gag interactions. These findings could have significant implications for understanding how cells may regulate retroviral complex assembly by miRNA expression and autophagy, and raise the possibility that similar regulations can occur in other biological contexts.  相似文献

The main purpose of the present study was to examine the relationships between measures of fitness [estimated peak oxygen consumption (V(O2) peak) and handgrip strength] and forearm vascular function in 55 young (22.6 +/- 3.5 yr) adults. In addition, the present study considered methodological and technical aspects regarding the examination of the venous system using mercury in-Silastic strain-gauge plethysmography (MSGP). Forearm venous capacitance and outflow were examined using five different [7, 14, 21, 28, and 35 mmHg < diastolic blood pressure (DBP)] venous occlusion pressures and after a 5- and 10-min period of venous occlusion. A pressure of 7 mmHg < DBP and a period of 10 min venous occlusion produced the greatest (P < 0.05) venous capacitance and outflow, without altering arterial indexes. Reproducibility of forearm arterial and venous indexes were evaluated at rest and after 5 min of upper arm arterial occlusion at 240 mmHg on three different occasions within 10 days with the interclass correlation coefficient ranging from 0.70 and 0.94. Estimated V(O2) peak correlated with postocclusion arterial inflow (r = 0.54, P = 0.012) and resting venous outflow (r = 0.56, P = 0.016). Finally, handgrip strength was associated with venous capacitance (r = 0.57, P = 0.007) and outflow (r = 0.67, P = 0.001). These results indicate that the examination of forearm vascular function using MSGP is reproducible. Moreover, the data show the importance of careful consideration of the selection of venous occlusion pressure and period when implementing these measures in longitudinal trials. Finally, the associations between fitness and venous measures suggest a link between venous function and exercise performance.  相似文献

Background

Computer Networks have a tendency to grow at an unprecedented scale. Modern networks involve not only computers but also a wide variety of other interconnected devices ranging from mobile phones to other household items fitted with sensors. This vision of the "Internet of Things" (IoT) implies an inherent difficulty in modeling problems.

Purpose

It is practically impossible to implement and test all scenarios for large-scale and complex adaptive communication networks as part of Complex Adaptive Communication Networks and Environments (CACOONS). The goal of this study is to explore the use of Agent-based Modeling as part of the Cognitive Agent-based Computing (CABC) framework to model a Complex communication network problem.

Method

We use Exploratory Agent-based Modeling (EABM), as part of the CABC framework, to develop an autonomous multi-agent architecture for managing carbon footprint in a corporate network. To evaluate the application of complexity in practical scenarios, we have also introduced a company-defined computer usage policy.

Results

The conducted experiments demonstrated two important results: Primarily CABC-based modeling approach such as using Agent-based Modeling can be an effective approach to modeling complex problems in the domain of IoT. Secondly, the specific problem of managing the Carbon footprint can be solved using a multiagent system approach.  相似文献

Background

Living systems are associated with Social networks — networks made up of nodes, some of which may be more important in various aspects as compared to others. While different quantitative measures labeled as “centralities” have previously been used in the network analysis community to find out influential nodes in a network, it is debatable how valid the centrality measures actually are. In other words, the research question that remains unanswered is: how exactly do these measures perform in the real world? So, as an example, if a centrality of a particular node identifies it to be important, is the node actually important?

Purpose

The goal of this paper is not just to perform a traditional social network analysis but rather to evaluate different centrality measures by conducting an empirical study analyzing exactly how do network centralities correlate with data from published multidisciplinary network data sets.

Method

We take standard published network data sets while using a random network to establish a baseline. These data sets included the Zachary''s Karate Club network, dolphin social network and a neural network of nematode Caenorhabditis elegans. Each of the data sets was analyzed in terms of different centrality measures and compared with existing knowledge from associated published articles to review the role of each centrality measure in the determination of influential nodes.

Results

Our empirical analysis demonstrates that in the chosen network data sets, nodes which had a high Closeness Centrality also had a high Eccentricity Centrality. Likewise high Degree Centrality also correlated closely with a high Eigenvector Centrality. Whereas Betweenness Centrality varied according to network topology and did not demonstrate any noticeable pattern. In terms of identification of key nodes, we discovered that as compared with other centrality measures, Eigenvector and Eccentricity Centralities were better able to identify important nodes.  相似文献

The objective of the study was to explore the changes in melatonin and zinc levels in rats with induced hypothyroidism. Thirty adult male rats used in the study were allocated to three groups with equal numbers. Group 1: General control group which was not subjected to any procedure. Group 2: Sham-hypothyroidism group to which was administered 10 mg kg(-1) intraperitoneal (i.p.) physiologic saline (0.09% NaCl) for 4 weeks. Group 3: Hypothyroidism group which was supplemented with intraperitoneal 10 mg kg(-1) propylthiouracil (PTU) for 4 weeks. Blood samples collected from all animals at the end of the study by decapitation were analysed for serum Total T4 (TT4), Total T3 (TT3), Free T4 (FT4), Free T3 (FT3) (ELISA) as well as for melatonin (RIA) hormones and zinc levels (atomic emission). Comparison of the study groups in terms of thyroid hormones, melatonin and zinc levels showed that TT4, TT3, FT4, FT3, melatonin and zinc levels in group 3 were lower than those in groups 1 and 2 (p < 0.01). These parameters were not different in groups 1 and 2. The results of the study demonstrate that PTU supplementation for 4 weeks results in a significant inhibition in both melatonin and zinc levels. Inhibited melatonin levels may result from the decrease in zinc levels.  相似文献
