The effect of (2RS, 3RS)-1-(4-Chlorophenyl)-4, 4-dimethyl-2-(1H-1,2,4 triazol-1-yl) pentan-3-ol (PP333) on the growth and transpiration of normal and root pruned colt rootstocks was measured. PP333 reduced plant height, stem diameter increment, leaf number, area and weight and stem weight. Root pruning reduced root, leaf and stem weight, and plant height in control plants. PP333 reduced both total water use and transpiration per unit leaf area and increased stomatal resistance. In control plants root pruning also reduced total water use and increased stomatal resistance. 15 days after the beginning of the experiment half the plants in all treatments were allowed to dry out. The effects of drought, i.e. reduced transpiration, growth and leaf water potentials, were smaller in PP333 treated than in control plants.  相似文献

Site-specific electrostatic measurements have been limited to soluble proteins purified for in vitro spectroscopic characterization or proteins of known structure; however, comparable measurements have not been made for functional membrane bound proteins. Here, using an electrochromic fluorophore, we describe a method to monitor localized electric field changes in a voltage-gated potassium channel. By coupling the novel probe Di-1-ANEPIA to cysteines in Shaker and tracking field-induced optical changes, in vivo electrostatic measurements were recorded with submillisecond resolution. This technique reports dynamic changes in the electric field during the gating process and elucidates the electric field profile within Shaker. The extension of this method to other membrane bound proteins, including transporters, will yield insight into the role of electrical forces on protein function.  相似文献

Parrot populations in Africa and Madagascar are declining and the need for conservation actions to address threats is increasingly recognised. Effective conservation requires a robust knowledge base on which decisions over appropriate actions can be made, yet at present there is no current and readily accessible synthesis of the status of populations, the threats they face and knowledge gaps. Here we begin to address this shortfall for the larger species in the region belonging to the genera Coracopsis, Poicephalus, Psittacus and Psittacula. We review developments since the production of the IUCN Parrot Action Plan published in 2000, identify areas where critical knowledge is lacking and highlight opportunities to address them. While advances have been made over the last decade, progress has not been evenly spread, with a strong bias towards populations in southern Africa. To date much research has focused on describing aspects of ecology and behaviour and there remains a need for studies determining the current status of populations and the factors limiting distributions and abundance. This review aims to provide a basis upon which progress towards an improved understanding of the conservation needs of the larger parrots of Africa and Madagascar can be made.  相似文献

Background

Despite a history of conflicts and widespread human rights violation in sub-Saharan Africa, little is known about the prevalence of interpersonal violence among the population in this region. Evidence from high-income countries suggests that exposure to violence has mental health consequences and violence also has associations with experiences of sexual coercion.

Aims

This study sought to investigate the prevalence of physical and perceived threats of violence among university students in Uganda and to assess the possible relationship between such violence, sexual coercion, and symptoms of anxiety, depression, and psychoticism, respectively.

Method

In 2005, 980 Ugandan university students responded to a self-administered questionnaire (response rate 80%) that assessed socio-demographic factors, social capital, importance of religion, mental health, experience of violence and sexual coercion, and sexual behaviour factors. Logistic regression analysis was applied as the main analytical tool.

Results

Of those who responded, 28% reported perceived threats/threats of violence and 10% exposure to actual physical violence over the previous 12 months, with no significant gender differences in exposure history. Exposure to violence was significantly associated with the experience of sexual coercion among both males and females. Sexual coercion and threats/threats of violence were both significantly associated with poor mental health in males and females, but only males showed a strong association between exposure to physical violence and poor mental health.

Conclusion

The current study suggests that in terms of general exposure, both males and females in the study population are equally exposed to sexual coercion and interpersonal violence, and both male and female students show generally similar mental health effects of exposure to such violence. The prevalence of interpersonal violence found in our study population may have long-term negative health implications. Our findings may serve as a baseline for interventions and continuing research aimed at preventing interpersonal violence.  相似文献

Background

Malaria still remains a major health issue in Ghana despite the introduction of Artemisinin-based combination therapy (ACT) coupled with other preventative measures such as the use of insecticide treated nets (ITNs). The global quest for eradication of malaria has heightened the interest of identifying drugs that target the sexual stage of the parasite, referred to as transmission-blocking drugs. This study aimed at assessing the efficacy and gametocydal effects of some commonly used herbal malaria products in Ghana.

Methodology/Principal Findings

After identifying herbal anti-malarial products frequently purchased on the Ghanaian market, ten of them were selected and lyophilized. In vitro drug sensitivity testing of different concentrations of the herbal products was carried out on asexual and in vitro generated gametocytes of the 3D7 strain of Plasmodium falciparum. The efficacies of the products were assessed by microscopy. Cultures containing low dose of RT also produced the least number of late stage gametocytes. Two of the herbal products CM and RT inhibited the growth of late stage gametocytes by > 80% at 100 μg/ml whilst KG was the most inhibitory to early stage gametocytes at that same concentration. However at 1 μg/ml, only YF significantly inhibited the survival of late stage gametocytes although at that same concentration YF barely inhibited the survival of early stage gametocytes.

Conclusions/Significance

Herbal product RT (Aloe schweinfurthii, Khaya senegalensis, Piliostigma thonningii and Cassia siamea) demonstrated properties of a highly efficacious gametocydal product. Low dose of herbal product RT exhibited the highest gametocydal activity and at 100 μg/ml, RT exhibited >80% inhibition of late stage gametocytes. However inhibition of asexual stage parasite by RT was not optimal. Improving the asexual inhibition of RT could convert RT into an ideal antimalarial herbal product. We also found that generally C. sanguinolenta containing herbal products exhibited gametocydal activity in addition to high asexual efficacy. Herbal products with high gametocydal activity can help in the fight to reduce malaria transmission.  相似文献

Evidence is presented suggesting, for the first time, that the protein foldability metric σ = (Tθ − Tf) / Tθ, where Tθ and Tf are, respectively, the collapse and folding transition temperatures, could be used also to measure the foldability of RNA sequences. These results provide further evidence of similarities between the folding energy landscapes of proteins and RNA. The importance of σ is discussed in the context of the in silico design of rapidly foldable RNA sequences.  相似文献

Forest reserves within the Ghanaian rain forest have been classified into categories (Condition I-VI) on the basis of their botanical importance and status of the vegetation. We present data on the distribution and abundance of avifauna of 28 Condition II and III forest reserves in southern Ghana based on line transect counts and mist-net captures. A total of 227 species were recorded for all the sites; species records for individual sites ranged from 78–119. The dominant groups in the overall survey were Muscicapidae (represented by 40 species), Pcynonotidae (21) and Cuculidae (12). The most abundant bird species in the Ghanaian forests were Yellow-whiskered Greenbul, Green Hylia, Green Pigeon and Olive Sunbird. Species encounter rates ranged from 16.7 to 50.7 species per km with individual bird encounter rates of 27.9 to 172.0 birds per km. Capture rates in mist-nets ranged from 1.2 to 3.9 birds per 100 metre net-hr (mnh). A total of 183 species were recorded in Condition II forest as compared with 167 in Condition III forests. The effects of forest condition and vegetation types on the total number of species recorded were, however, not statistically significant. The Similarity Index for bird communities within Condition II and III forests was 0.72. Bird species composition in the two forest types also differed: e.g., seven primary forest species, as well as 40% of the species associated with primary and matured secondary forest which were recorded in Condition II forests, were absent from Condition III forests. Bird communities of Moist Evergreen, Upland Evergreen and Moist semi-Deciduous were more similar than communities in the Wet Evergreen forest type. The implication of the results for conservation of the Ghana's forests is discussed.  相似文献

Adenosine is an endogenous signaling molecule that is highly up-regulated in inflammatory states. Adenosine acts through the A2b receptor, a G protein-coupled receptor that couples positively to Galpha(s) and activates adenylate cyclase. This leads to cAMP-mediated electrogenic chloride secretion in intestinal epithelia. To better understand the regulation of the A2b receptor in intestinal epithelia, we studied the effects of interferon-gamma (IFN-gamma), a potent immunomodulatory cytokine, in the T84 cell line. Pretreatment of cells with 500 units/ml IFN-gamma for 12 h inhibited an adenosine-induced short circuit current (Isc) without affecting the transepithelial resistance. Under these conditions, IFN-gamma did not inhibit the protein expression or membrane recruitment of the A2b receptor, shown to be essential for its function. Interestingly, IFN-gamma inhibited cAMP levels as well as its downstream signaling pathway as shown by the inhibition of adenosine-induced phosphorylation of cAMP response element-binding protein and protein kinase A activity. Similar studies with forskolin, a direct activator of adenylate cyclase, also demonstrated inhibition of cAMP and its downstream response by IFN-gamma. However, IFN-gamma did not affect secretory responses to the calcium-dependent secretagogue carbachol or cAMP analog 8-bromo-cAMP, indicating that normal secretory responses to adequate second messengers in IFN-gamma-treated cells are achievable. Moreover, IFN-gamma inhibited the expression of adenylate cyclase isoforms 5 and 7. In conclusion, we demonstrate that IFN-gamma down-regulates adenosine-mediated signaling possibly through the direct inhibition of adenylate cyclase expression. We propose that IFN-gamma may acutely affect global cAMP-mediated responses in the intestinal epithelia, thereby decreasing secretory responses, which may consequently aggravate inflammatory processes.  相似文献
