Strobilomyces echinocephalus Gelardi & Vizzini sp. nov. is described as a new species, based on collections from Kunming (Yunnan Province, China). Macro-morphological and micro-morphological features, ecological data and analyses of RPB1 and ITS2 sequences support the establishment of this new taxon. Colour pictures of fresh basidiomata in habitat, scanning electron microscope (SEM) micro-photographs of the spores, and a line-drawing displaying the most relevant anatomical characters are provided in this paper. Strobilomyces zangii Gelardi nom. nov. is proposed to replace the name Heimiella nigricans M. Zang. A dichotomous key to the Strobilomyces species at global level is also provided at the end of this work.  相似文献

This study was focused on species of Peziza belonging to the “P. ammophila” complex, using both morphological and molecular approaches. Molecular and morphological analyses showed that several taxa are hidden under the name “ammophila” and, as a consequence, two additional species and one form are established: P. hellenica sp. nov., P. oceanica sp. nov. and P. ammophila f. megaspora f. nov. On the basis of the data that we have, these taxa seem to be related to some geographical areas; P. ammophila appears to be prevalent in northern, central and southern Europe, its f. megaspora is distributed in France and in the Netherlands, P. hellenica comes from Greece and P. oceanica from New Zealand. A cryptic species, not yet circumscribed satisfactorily, is described as P. deceptiva ad int. Moreover, the problem concerning the identification of P. ammophila is raised, the species is epitypified with a sequenced collection from Sicily (Italy) and a key to the species of the complex is provided.  相似文献

The purification, cloning, sequencing, molecular properties and expression of a fucose-binding lectin from the serum of Dicentrarchus labrax (DlFBL) have been previously reported. We now describe the distribution and expression of DlFBL during fish ontogeny. Immunohistochemistry and in situ hybridization assays were carried out at various developmental stages (from 10 days post-hatching larvae to juveniles). Another fucose-binding lectin, similar to DlFBL in biochemical, immunochemical and agglutinating properties, was extracted and purified from eggs and appeared to be localized in the embryo yolk sack residual. DlFBL was found in columnar and goblet cells of the intestinal epithelium of larvae (from 20 days post-hatching) and juveniles and in parenchymal tissue of juveniles. DlFBL mRNA and protein were detected in the intestinal epithelium and in hepatocytes. An amplification product from degenerate primers indicates that lectin isotypes with DlFBL epitopes are expressed in eggs and embryos. Whether the lectin fraction isolated from eggs and embryos includes DlFBL of maternal origin remains unclear.  相似文献

The aim of this work was to clarify the genetic structure of the ectomycorrhizal fungus, Tuber magnatum Pico, in a natural truffle ground located in north Italy. Ascomata of this population of T. magnatum were collected over a period of up to 5 years. For comparative analysis, T. magnatum fruit bodies of different geographical origin were also considered. We used single locus markers, such as the variable region of ribosomal genes (ITS), the beta-tubulin gene and sequence-characterized amplified regions (SCAR), as tools to identify single-nucleotide polymorphisms (SNPs). On the basis of the molecular results, which were indirectly supported by a karyological analysis, a self-fertilization mechanism is suggested. A SCAR region was polymorphic within the samples of the truffle ground, leading to the identification of two genotypes. In addition, both the SCAR and the ITS proved to be polymorphic among samples coming from different geographical regions, revealing a genetic differentiation in T. magnatum.  相似文献

A novel fucose-binding lectin, designated SauFBP32, was purified by affinity chromatography on fucose-agarose, from the serum of the gilt head bream Sparus aurata. Electrophoretic mobility of the subunit revealed apparent molecular weights of 35 and 30 kDa under reducing and non-reducing conditions, respectively. Size exclusion analysis suggests that the native lectin is a monomer under the selected experimental conditions. Agglutinating activity towards rabbit erythrocytes was not significantly modified by addition of calcium or EDTA; activity was optimal at 37 degrees C, retained partial activity by treatment at 70 degrees C, and was fully inactivated at 90 degrees C. On western blot analysis, SauFBP showed intense cross-reactivity with antibodies specific for a sea bass (Dicentrarchus labrax) fucose-binding lectin. In addition, the similarity of the N-terminal sequence and a partial coding domain to teleost F-type lectins suggests that SauFBP32 is a member of this emerging family of lectins.  相似文献

Four European Hodophilus species with an odour similar to naphthalene, a strong unpleasant odour similar to that of mothballs, are recognized based on sequence and/or morphological data. The traditional concept defines Ho. foetens as the only Hodophilus species with a naphthalene odour in Europe. This name is now assigned to one of the studied species based on morphological examination of the holotype specimen. A recently collected specimen is proposed as the epitype. The other three species with a naphthalene odour are described here as new: Ho. pallidus, Ho. subfoetens and Ho. tenuicystidiatus. They are distinguishable in the field based on a combination of lamellae number and colour of basidiomata. All four species are grouped in the Ho. foetens superclade, one of two superclades, together with the Ho. micaceus superclade, in the genus Hodophilus. All are different species from North American taxa with a naphthalene-like odour recognised in a previous study. The Ho. foetens superclade also includes one species identified as Ho. atropunctus that does not have a distinctive odour. The type collection of Ho. albofloccipes, a recently described European species with a naphthalene odour, is placed together with some collections without a distinctive odour in the Ho. micaceus superclade.  相似文献

Paxillus involutus is a model species for ecological or physiological studies of ectomycorrhizal agaricomycetes. Three to six groups or species linked to it have been ecologically and morphologically distinguished. Phylogenetic studies have revealed the existence of four species in Europe: Paxillus ammoniavirescens, Paxillus obscurisporus, P. involutus, and a fourth as yet not described species. We studied 47 collections from 24 French and Italian locations, supplemented with GenBank data, in order to genetically and taxonomically delineate these species. Phylogenetic analyses of three nuclear DNA regions (rDNA internal transcribed spacer (ITS), tef1-α, and gpd) confirmed the four European species. Morphology, culture, and ecology features allowed us to delineate species boundaries and to describe the fourth species we named Paxillus cuprinus since it turns coppery with age. As there is no existing original herbarium specimen for P. involutus, one of our collections was chosen as the epitype. The low genetic diversity found in P. cuprinus correlates with stable morphological traits (basidiome colour, ovoid–amygdaliform spores with an apical constriction) and with ecological preferences (association with Betulaceae in open and temperate areas). In contrast, P. ammoniavirescens is characterized by a high genetic diversity and a high variation of its morphological and ecological features.  相似文献

Musumecia, a new genus of Agaricales, is described to accommodate the new species Musumecia bettlachensis. Based on a combined ITS– and LSU–rDNA Bayesian, Maximum Likelihood and Maximum Parsimony analysis, Musumecia clearly clusters within the Tricholomatoid clade, where it is sister to Pseudoclitocybe. Musumecia is distinguished from allied genera by a unique combination of macro‐ and micromorphological characters, including basidiomes with a clitocyboid/hygrophoroid habit, emerging from a fleshy pseudosclerotial mass (pseudosclerotium), decurrent and thick lamellae, a brown darkening of both lamellae and stipe, whitish‐cream spore print, elongated non‐siderophilous basidia, smooth, acyanophilous and inamyloid basidiospores, and the absence of both cystidia and clamp‐connections.  相似文献
