A new genus and species combination are proposed for Urocryptum tortum n. gen., n. comb., a scuticociliate with a polymorphic life cycle. This marine ciliate was isolated from a sample taken at Gokasho Bay in Mie Prefecture (Japan). Specimens from different phases of the growth cycle were examined in vivo and with two silver staining techniques. Three life-history stages were observed: an exponential growth phase stage (trophont), a stationary phase stage (tomite), and finally a resting stage (cyst). The exponential growth form is laterally flattened and ovoid; it has 20-24 somatic kineties (SK) and a typical complement of scuticociliate oral structures. Polykinetid 1 (Pk1) has two longitudinal files of 6 kinetosomes (Ks); sometimes one or two additional kinetosomes are located anteriorly. Polykinetid 2 (Pk2) has two files of 6 or 7 Ks; a third file of three widely spaced kinetosomes is located on its right side, as well as a small curved row of 5 Ks positioned on the right hand side of the posterior end of Pk2. Polykinetid 3 (Pk3) has 3 rows of 4, 5, and 7 Ks's perpendicular to the haplokinety (Hk) or paroral membrane. The stationary growth phase cell is spindle-shaped and has a similar number of SK that are much more closely spaced than in exponentially growing specimens. Oral infraciliary structures are reduced in size, having fewer kinetosomes and being positioned more anteriorly in the cell. Pk1 is composed of two files of 5-Ks, Pk2 has only two files of 6-7 Ks, and Pk3 has two to three rows of 3-4 Ks. The Hk is displaced anteriorly and becomes straight, losing the shape typical of exponential growth phase cells. Cysts are pyriform with a wide cyst wall; no infraciliary structures were visible.  相似文献

ABSTRACT. The morphology and morphogenesis of two species of the genus Lembadion, L. lucens and L. bullinum , are described. In both species, left and right ventral kineties converge behind the mouth forming a postoral suture. Buccal infraciliature is formed by one polykinety and two very close paroral kineties (inner and outer). During stomatogenesis, the new oral structures originate from the paroral kineties. The inner paroral kinety forms the new adoral polykinety and regenerates the outer paroral kinety of the proter, while the paroral kineties of the opisthe originate from the outer paroral kinety of the parental cell. Somatic proliferation starts before the stomatogenesis at the equatorial level of the cell, and extends towards the poles forming an equatorial band. Two large invariant zones, anterior and posterior, remain in the dividing cell. Moreover, the kinetodesmal fibers disappear in the proliferation band during the bipartition (fission) process.  相似文献

ABSTRACT. Marine goniomonads have a worldwide distribution but ultrastructural information has not been available so far. An isolate of the heterotrophic marine nanoflagellate Goniomonas (G. aff. amphinema) from North Wales (UK) has been studied, providing information on its morphology and cellular structure using video, electron, laser scanning confocal microscopy (LSCM), and atomic force microscopy. Here, we describe a new feature, a granular area, potentially involved in particle capture and feeding. The binding of the lectin wheat germ agglutinin to the granular area of cells with discharged ejectisomes indicates the adhesive nature of this novel feature. The presence of a microtubular intracellular cytopharynx, apparently also used for feeding, has been revealed by LSCM. The small subunit rRNA gene of the isolate has been sequenced (1,788 bp). Phylogenetic results corroborate significant genetic divergence within the marine members of Goniomonas. This work highlights the need for integrated morphological, ultrastructural, and molecular investigation when describing and studying heterotrophic nanoflagellates.  相似文献

The somatic and buccal infraciliature of Lagynus elegans are described, and aspects of its division and conjugation are reported. Its somatic infraciliature is made up of 37–46 meridianal kineties composed of isolated kinetosomes that have thick and long kinetodesmal fibers. In the anterior zone of the cell, the circumoral infraciliature can be observed: it is composed of short, slightly oblique kinetal segments, which are formed of three kinetosomes each. The brosse of this species consists of 3 or 4 groups that possess 4 to 6 ciliated kinetosomes each; these kinetosomes lack kinetodesmal fibers. On the apical pole of the cell, surrounding the oral opening, a crown of nematodesmata is observed; these nematodesmata are connected to each other by a fibrillar structure. Taking into account these features, we propose that this genus be transferred from the order Prostomatida to a new family, Lagynidae, of the order Prorodontida.  相似文献

The infraciliature, argyrome, and myonemic system of Opisthonecta henneguyi have been examined after impregnation with the Fernández-Galiano silver impregnation method. The oral infraciliature is formed by one haplokinety and three polykineties while the aboral infraciliature is formed by the trochal band—six kinetosomes wide—and the scopula. The trochal band has three fibrillar systems: orally directed fibers, aboral fibers, and oblique fibers. The argyrome is formed by 90 to 135 fine circular striations, which cover the whole organism with the exception of the peristome. The myonemic system is located in two regions: the oral and the aboral ones. The oral myonemic system is formed by a fibrous ring which surrounds the peristome underneath the spiral of the peristomial infraciliature and by a group of fibers which depart from the peristomial disc. The aboral myonemic system is much simpler, and it is formed by fibers which extend radially from the scopula. Biometrical characterizations of the vegetative cells and living cysts were also made, and our data are compared with the results obtained previously by other authors.  相似文献

A new method for the enrichment of cultures of Serratia marcescens for auxotrophic mutants has been developed. The method is based on the formation of filaments by growing cells in minimal medium M70 containing azthreonam. Auxotrophic mutants unable to grow in M70 do not form filaments. Mutants are collected from the culture by filtration.  相似文献

Opisthonecta matiensis n. sp. was isolated from the inlet water of a wastewater treatment plant near Madrid, Spain, and studied in vivo, with silver methods, and using electronic and indirect immunofluorescence microscopy. This new species shows an amphora-like cell shape and has a size of 45-73 microns (x 58.2) x 25-40 microns (x 31.3). The oral infraciliature is formed by one haplokinety, three polykineties, and a short row of kinetosomes (epistomial membrane). The aboral infraciliature is made up of the trochal band and the scopula. From the trochal band arise three fibrillar systems: oral fibers, aboral fibers, and oblique fibers. The myoneme system is composed of a delicate peristomial ring, longitudinal branched fibers that reach the trochal band and of radial fibers extending from the scopula to the trochal band. The silverline system consists of an average of 147 lines. This new species is separated from other known forms by its smaller size, the presence of one single vacuole, and its higher number of silverlines.  相似文献
