Lycopene beta-cyclase (Lyc-B) is the key enzyme in the catalysis of linear lycopene to form cyclic beta-carotene, an indispensable part of the photosynthetic apparatus and an important source of vitamin A in human and animal nutrition. Studies showing that the microalga Dunaliella salina can accumulate a high level of beta-carotene are lacking. We hypothesize that D. salina is closely involved with the catalytic mechanism of Lyc-B and the molecular regulation of its gene. In this study, we used RT-PCR and RACE-PCR to isolate a 2475 bp cDNA with a 1824 bp open reading frame, encoding a putative Lyc-B, from D. salina. Homology studies showed that the deduced amino acid sequence had a significant overall similarity with sequences of other green algae and higher plants, and that it shared the highest sequence identity, up to 64%, with Lyc-B of Chlamydomonas reinhardtii. Codon analysis showed that synonymous codon usage in the enzyme has a strong bias towards codons ending with adenosine. Two motifs were found in the Lyc-B sequence, one at the N terminus, for binding the hypothetical cofactor FAD, and the other was a substrate carrier motif in oxygenic organisms shared by an earlier carotenogenesis enzyme, phytoene desaturase, and Lyc-B. A tertiary structure prediction suggested that the catalytic or binding site structure within LycB from D. salina is superior to that of both H. pluvialis and C. reinhardtii. The LycB protein from D. salina was quite removed from that of H. pluvialis and C. reinhardtii in the phylogenetic tree. Taken as a whole, this information provides insight into the regulatatory mechanism of Lyc-B at the molecular level and the high level of beta-carotene accumulation in the microalga D. salina.  相似文献

One hundred and seventy-two strains of Escherichia coli isolated from diarrheal patients in Beijing, P. R. China, were analyzed for plasmid DNA profile, HEp-2 cell adherence ability and reactivity to 10 previously described DNA probes. They had not been recognized as pathogenic E. coli in China. Of the 110 strains tested, 76 (69%) contained one or multiple large plasmids. Of the 71 strains with the large plasmids 64 could adhere to HEp-2 cells. Of the 172 strains, 102 (59.3%) were hybridized with at least one of the 10 probes. Of those, seven strains hybridized with enteroaggregative E. coli (EAggEC) probe. Their serotypes were O128 (two strains), O6 (one strain), and O111 (one strain). Three strains were untypable. Six and three strains were hybridized with enteropathogenic E. coli (EPEC) attaching and effacing genes (eae) or EPEC adherence factor (EAF) probe, respectively. Two non-O157: H7 strains hybridized with enterohemorrhagic E. coli (EHEC) probe. Seventy-two strains (41.9%) hybridized with shiga-like toxin 2 or 1 (SLT2 or SLT1) probes. Among the SLT1 or SLT2 probe-positive strains, 54 hybridized with invasive (INV) plasmid probe developed for identification of enteroinvasive E. coli (EIEC) and Shigella species. The INV and SLT probe-positive strains might represent a new variety of verotoxin-producing E. coli (VTEC).  相似文献

Identifying the factors that cause reproductive isolation and their relative importance in species divergence is crucial to our understanding of speciation processes. In most species, natural selection is commonly considered to play a large role in driving speciation. Based on whole genome re-sequencing data from 27 Populus alba and 28 Populus adenopoda individuals, we explored the factors related to reproductive isolation of these two closely related species. The results showed that the two species diverged ~5–10 million years ago (Ma), when the Qinghai–Tibet Plateau reached a certain height and the inland climate of the Asian continent became arid. In highly differentiated genomic regions, the relative divergence (FST) and absolute divergence (dxy) were significantly higher than the genomic background, θπ and shared polymorphisms decreased whereas fixed differences increased, which indicated that natural selection played a key role in the reproductive isolation of the two species. In addition, we found several genes that were related to reproduction that may be involved in explaining the reproductive isolation. Using phylogenetic trees resolved from haplotype data of Populus tomentosa and P. adenopoda, the maternal origin of P. tomentosa from P. adenopoda was likely to be located in Hubei and Chongqing Provinces.  相似文献

The chemical shift assignments and secondary structure of a murine–human chimera,MH35, of leukaemia inhibitory factor (LIF), a 180-residue protein of molecular mass 20 kDa,have been determined from multidimensional heteronuclear NMR spectra acquired on auniformly 13C,15N-labelled sample. Secondary structure elements were defined on the basisof chemical shifts, NH-CH coupling constants, medium-range NOEs and the location ofslowly exchanging amide protons. The protein contains four -helices, the relativeorientations of which were determined on the basis of long-range, interhelical NOEs. The fourhelices are arranged in an up-up-down-down orientation, as found in other four-helical bundlecytokines. The overall topology of MH35-LIF is similar to that of the X-ray crystallographicstructure for murine LIF [Robinson et al. (1994) Cell, 77, 1101–1116]. Differencesbetween the X-ray structure and the solution structure are evident in the N-terminal tail, wherethe solution structure has a trans-Pro17 compared with the cis-Pro17 found in the crystalstructure and the small antiparallel -sheet encompassing residues in the N-terminus andCD loop in the crystal structure is less stable.  相似文献

以17个建兰(Cymbidium ensifolium)品种为材料,采用改良的丙酮法提取叶绿素,再通过Arnon丙酮法公式计算光合色素含量,利用捷克FluorCam开放式叶绿素荧光仪测定不同品种的叶绿素荧光参数。结果表明,17个建兰品种的光合色素和叶绿素荧光参数具有不同程度的差异,其中‘铁骨素’(C. ensifolium ‘Tiegusu’)、‘逸红双娇’(C. ensifolium ‘Yihongshuangjiao’)和‘闽南黄蝶’(C. ensifolium ‘Minnanhuangdie’)的光合色素含量高于其他品种,表明这3个品种具有良好的光合效率,吸收光能的能力较强;‘铁骨素’最大荧光产量(Fm)、Kautsky诱导效应最大荧光(Fp)、PS Ⅱ原初光能转化效率(Fv/Fm)和非光化荧光淬灭系数(NPQ)均为最高。综上可知,‘铁骨素’的光合生理特性优于其他品种,可作为优良建兰品种进行种植推广。  相似文献

为揭示小叶锦鸡儿(Caragana microphylla)天然居群叶形态性状的变异规律及其生态适应性特征,该研究以10个小叶锦鸡儿天然居群为对象,通过多重比较、巢式方差分析、相关性分析、聚类分析和主成分分析等方法,对7个叶形态性状进行分析。结果表明:(1)小叶锦鸡儿叶形态性状在居群内和居群间均存在极显著差异(P < 0.01),平均变异系数为10.13%,不同性状的变异幅度为6.23%~12.78%;平均叶形态性状的表型分化系数为43.62%,居群内变异(30.09%)大于居群间变异(24.91%),说明居群内是其叶形态性状变异的主要来源。(2)相关性分析表明,环境因子对小叶锦鸡儿的叶形态性状变异有很大的影响,在地理空间上主要呈现出沿海拔梯度的变异模式;主成分分析的结果显示,小叶宽、叶柄宽和叶柄长对小叶锦鸡儿叶形态变异起主导作用;利用欧式距离对小叶锦鸡儿居群进行UPGMA聚类分析结果显示,基于叶形态性状和环境因子可分别将小叶锦鸡儿10个居群分为3类和2类,Mantel检验结果表明,小叶锦鸡儿的叶形态性状变异不存在地理连续性。研究结果为小叶锦鸡儿的适应性进化和开发利用提供了理论依据。  相似文献

水杨酸(SA)和硫化氢(H2S)在调控非生物胁迫下植物生长发育和生理代谢中均起着非常重要的作用,但二者作为信号分子在调控低温弱光下黄瓜光合作用中的互作关系还不清楚。本试验以黄瓜幼苗为试材,分别用SA和硫氢化钠(NaHS,H2S供体)及其清除剂或抑制剂喷撒叶面,以适宜温光下去离子水处理为对照(CK),研究低温(8 ℃/5 ℃,昼/夜)弱光(100 μmol·m-2·s-1)下SA和H2S对黄瓜幼苗光合作用的调控及互作关系。结果表明: SA可明显增强L-/D-半胱氨酸脱巯基酶(LCD、DCD)活性及其mRNA表达,促进内源H2S产生;NaHS对苯丙氨酸解氨酶和异分支酸合成酶活性、mRNA表达量及内源SA含量影响不大。SA和NaHS可使低温弱光下黄瓜幼苗的光合速率、气孔导度和蒸腾速率明显提高,胞间CO2浓度显著降低;同时增强核酮糖-1,5-二磷酸羧化酶、Rubisco活化酶、景天庚酮糖-1,7-二磷酸酯酶和果糖-1,6-二磷酸醛缩酶活性及其mRNA表达,促进光合碳同化;提高光下PSⅡ实际光化学效率和暗下PSⅡ最大光化学效率,从而减轻低温弱光胁迫对黄瓜幼苗的光合机构的损伤和生长量的影响。H2S清除剂次牛磺酸(HT)可使SA对低温弱光下黄瓜幼苗的光合作用和生长促进效应明显减弱,而SA抑制剂多效唑和氨基茚磷酸对H2S诱导的黄瓜幼苗光合机构对低温弱光的耐受性无显著影响,说明H2S作为SA的下游信号,参与调控低温弱光下黄瓜幼苗的光合作用。  相似文献

阿尔泰山的北方森林是中亚以及全球的生态系统的重要组成部分, 其生长动态可以影响到全球范围的热辐射、碳平衡等。因此, 探究影响阿尔泰山树木径向生长的主要因素至关重要。该研究以新疆喀纳斯国家级自然保护区的西伯利亚五针松(Pinus sibirica)为研究对象, 建立西伯利亚五针松年表, 通过分析不同时间间隔累年生长量、竞争指数以及气候因子之间的关系, 运用线性混合效应模型、相关分析等方法, 探究竞争和气候对新疆阿尔泰山西伯利亚五针松树木径向生长的影响。结果表明: (1)线性混合效应模型结果显示竞争树胸径和与西伯利亚五针松过去30年的累年生长量之间的拟合效果最好; (2)标准年表与3月的平均气温、平均最高气温、平均最低气温之间有显著正相关关系; (3)累年生长量最高值出现在气温0-5 ℃, 竞争指数低于100的时候。累年生长量最低时, 气温达到-10 ℃, 竞争指数也超过了300。目标树的树木径向生长受到竞争树胸径和及生长季前期气温的影响, 两者共同作用。但相较于气候因子而言, 竞争对西伯利亚五针松的树木径向生长有更大的影响作用。  相似文献

针对白蚁防治药剂联苯菊酯传统加工剂型存在的安全性差、持效期短等缺点,采用溶剂蒸发法制备联苯菊酯微胶囊.通过粒径大小、外观形貌、包封率以及载药量筛选出最佳芯壁比、乳化剂用量和剪切时间,并对微胶囊理化特性及释放性能进行表征,同时考察微胶囊对白蚁的杀灭效果和持效性.研究结果表明,芯壁比为1:1.5,乳化剂用量为7%,剪切时间为6 min时制备的联苯菊酯微胶囊粒径适中(97.6μm),包封率高达70.5%,缓释性能良好,与市售乳油相比,对白蚁的杀灭效果相当,但持效性能优异.该研究获得的联苯菊酯微胶囊为安全、高效防治白蚁提供了技术手段.  相似文献
