A revised enzyme synthesis rate expression for cybernetic models of bacterial growth is presented. The rate expression, which is comprised of inducible and constitutive contributions, provides for a basal enzyme level that is necessary to predict certain types of commonly observed continuous culture transients. The response of a continuous culture to a step change in feed stream composition is simulated using both the old and new formulations, and the ramifications for the "matching-law" formulation are discussed.  相似文献

Macromolecules present in low concentrations in xylem fluid of Medicago sativa L. var DuPuits will increase the resistance to xylem liquid flow. This increase in resistance was found to be reversible by backflushing the xylem. Autoradiography showed that very large molecules do not pass through pit membrane pores. A comparison of pit membrane pore sizes to molecule sizes suggests that increased resistance to xylem flow is a result of plugging pit membrane pores. It was also found that pit membranes located in two parts of the plant differ in the apparent diameter of their pores and, thus, in their susceptibility to plugging by macromolecules. Macromolecules in xylem fluid may result from hostparasite interactions and may play a significant role in the outcome of the interaction.  相似文献

The design, construction, and characterization of a prototype-regenerable glucose biosensor based on the reversible immobilization of glucose oxidase (GOx) using cellulose binding domain (CBD) technology is described. GOx, chemically linked to CBD, is immobilized by binding to a cellulose matrix on the sensor-indicating electode. Enzyme immobilization can be reversed by perfusing the cellulose matrix with a suitable eluting solution. An autocavable sensor membrane system is employed which is shown to be practical for use in real microbial fermentations. The prototype glucose biosensor was used without failure or deterioration during fed-batch fermentations of Escherichia coli reaching a maximum cell density of 85 g (dry weight)/L. Medium glucose concentration based on sensor output correlated closely with off-line glucose analysis and was controlled manually at 0.44 +/- 0.2 g/L for 2 h based on glucose sensor output. The sensor enzyme component could be eluted and replaced without interrupting the fermentation. To our knowledge, no other in situ biosensor has been used for such an extended period of time in such a high-cell-density fermentation. (c) 1995 John Wiley & Sons, Inc.  相似文献

Increasing environmental legislation which controls the release and the levels of certain chemicals in the environment has created a need for reliable monitoring of these substances in air, soil and especially water. Conventional analytical techniques, although highly precise, suffer from the disadvantages of high cost, the need for trained personnel and the fact that they are mostly laboratory bound. Biosensors because of their specificity, fast response times, low cost, portability, ease of use and a continuous real time signal, can present distinct advantages in certain cases. Their biological base makes them ideal for toxicological measurements which are suited for health and safety applications. Over the last 3-4 years there has been an increase in the number of publications concerning biosensors for environmental monitoring, especially in the field of pesticide measurements.This paper reviews some of the more important developments over the past 3-4 years.  相似文献

Persistent atrial paralysis in a patient with complete heart block and mild mitral insufficiency is presented. Left atrial specimens obtained during implantation of a permanent cardiac pulse generator showed evidence of hypertrophy and fibrosis; subcellular degenerative changes ranged from near normal to irreversible, thus suggesting that atrial paralysis may be due to the replacement of normal atrial muscle with nonfunctional fibrous tissue.  相似文献

We have examined the response of the hormone-resistant mutants axr1 and axr2 of Arabidopsis thaliana to inoculation by Agrobacterium tumefaciens and Agrobacterium rhizogenes. Our results indicate that recessive mutations in the axr1 gene affect the frequency of tumor formation after inoculation with either Agrobacterium strain. In addition, tumors produced on axr1 plants were smaller than those growing on wild-type plants. These results indicate that the product of the AXR1 gene is important for both crown gall and hairy root tumor formation. In contrast, the dominant axr2 mutation has a more severe effect on the development of crown gall tumors than on hairy root tumors. Crown gall tumors produced on axr2 plants had a different morphology than wild-type tumors and did not grow when they were removed from the explant. In contrast, a large number of hairy root tumors were produced on wild-type and axr2 plants, and both types of tumors grew when they were removed from the explant. Like the roots of axr2 plants, roots produced on axr2 explants lacked root hairs.  相似文献

THE CHANGE IN MEAN FITNESS FOR TWO LOCI UNDER SELECTION CAN BE DESCRIBED BY FOUR TERMS: (i) the variance of fitness, (ii) a weighted between-gamete covariance, (iii) a function of recombination, linkage disequilibrium and the slope of the surface of mean fitness on disequilibrium, and (iv) a function of these two parameters and the curvature of the surface. Concurrent measurements of evaporation, leaf conductance, irradiance, leaf water potential, and osmotic potential of maize (Zea mays L. cv. Pa602A) in soil at either high or low soil water potential were compared at several hours on two consecutive days in July. Hourly evaporation, measured on two weighing lysimeters, was similar until 1000 hours Eastern Standard Time, but thereafter evaporation from the maize in the dry soil was always less than that in the wet soil; before noon it was 62% and by midafternoon, only 35% of that in the wet soil. The leaf water potential, measured with a pressure chamber, was between −1.2 and −2.5 bars and between −6.8 and −8 bars at sunrise (about 0530 hours Eastern Standard Time) in the plants in the wet and dry soil, respectively, but decreased quickly to between −8 and −13 bars in the plants in the wet soil and to less than −15 bars in the plants in the dry soil by 1100 to 1230 hours Eastern Standard Time. At this time, the leaf conductance of all leaves was less than 0.1 cm sec−1 in the maize in the dry soil, whereas the conductance was 0.3 to 0.4 cm sec−1 in the leaves near the top of the canopy in the wet soil. The osmotic potential, measured with a vapor pressure osmometer, also decreased during the morning but to a smaller degree than leaf water potential, so that by 1100 to 1230 hours Eastern Standard Time the leaf turgor potential was 1 to 2 bars in all plants. Thereafter, leaf turgor potential increased, particularly in the plants in soil at a high water potential, whereas leaf water potential continued to decrease even in the maize leaves with partly closed stomata. Evidently maize can have values of leaf conductance differing 3- to 4- fold at the same leaf turgor potential, which suggests that stomata do not respond primarily to bulk leaf turgor potential. Evidence for some osmotic adjustment in the plants at low soil water potential is presented. Although the degree of stomatal closure in the maize in dry soil did not prevent further development of stress, it did decrease evaporation in proportion to the decrease in canopy conductance.  相似文献
