Forty-two white-rot fungi isolated in South America were incubated with long fibre sugar cane bagasse (LFB). The residual composition of LFB was determined after white-rot decay at 30 and 60 days. The ratio of residual lignin to residual lignin to residual cellulose (RL/RC) of untreated material (LFB) was 0.48. After white-rot-decay, the residual material with lower RL/RC ratios indicated that mainly lignin was degraded. In only 30 days, Phlebia sp. MVHC 5535, Athelia sp. MVHC 5509 and Spongipellis pachyodon MVHC 5019 caused a decrease in the RL/RC ratio to 0.36, 0.37 and 0.38, respectively, while it took 60 days for Ganoderma applanatum MVHC 5347, Hyphodontia sp. MVHC 5544, Panus tigrinus MVHC 5400, Stereum sp. MVHC 5113, Phellinus punctatus MVHC 5346 and MVHC 6388 to reach a ratio lower than 0.40. No correlation was found between the amount of some ligninolytic enzymes secreted and the residual composition of bagasse after white-rot fungi fermentation. Most of the fungal strains caused an increase in the relative amount of residual cellulose, indicating that hemicellulose was the preferred energy source. 相似文献

Summary Lactic acid produced by continuous culture of L.casei in an upflow packed bed reactor, was recovered with Amberlite IRA 400 in a fluidized bed column. Bed expansions of 1.25 and 2.25 were applied. Reutilization did not alter the capability of net recovery of 0.048 ± 0.01 g lactic acid/g resin. When 2200 cm/h of ascensional velocity was used, (bed expansion of 2.25), the resin adsorbed 39.3% of the initial lactic acid and 63.5% was eluted. This resin supported the highest exchange capacity of 0.126 g lactic acid/g resin. Applying high flow rates, the process has potential industrial applications due to the short time employed. 相似文献

The influence of environmental pH on the regulation of glucose catabolism by Lactobacillus reuteri was examined in anaerobic batch cultures. Under acidic conditions both glucose consumption and end-products formation were low. Maximum biomass was reached at pH 5·0, with a specific growth rate of µ= 0·78 h-1 . The shift in pH values from 4.3 to 6.5 reflected an increase in glucose uptake as well as in the yield ( Y p/x ) of acetate, lactate and ethanol after 12 h of incubation. Ethanol was the major metabolite produced at all pH values assayed. 相似文献

Summary The Castellani's Method for the preservation of Sclerotium rolfsii in sterile distilled water was tested. Culturing on Potato Dextrose Yeast extract (PDY) slants, the current system used, was also evaluated. Preservation of sclerotia according to the Castellani's method allowed the strain survival for more than two years. Comparing with the strain periodically activated, a critical decrease (about 80%) in -glucan synthesizing capacity was detected for mycelium preserved either on PDY slants or in water. Activation of stored sclerotia followed by subculturing in liquid Production Medium (PM) allowed preparation of homogeneous suspensions for batch fermentations, and scleroglucan concentrations achieved were similar to those with the strain periodically activated. 相似文献

5.5 and 37 °C and a second stage at pH 6.0, a temperature change from 40 °C to 45 °C in the second stage resulted in a 100%
substrate consumption at an overall dilution rate of 0.05 h−1. To increase the cell mass in the system, an adhesive strain of L. casei was used to inoculate two packed-bed reactors, which operated with two mixed feedstock streams at the optimal conditions
found above. Lactic acid fermentation started after a lag period of cell growth over foam glass particles. No significant
amount of free cells, compared with those adhering to the glass foam, was observed during continuous lactic acid production.
The extreme values, 57.5 g l−1 for lactic acid concentration and 9.72 g l−1 h−1 for the volumetric productivity, in upflow packed-bed reactors were higher than those obtained for free cells (48 g l−1 and 2.42 g l−1 h−1) respectively and the highest overall l(+)-lactic acid purity (96.8%) was obtained in the two-chemostat system as compared with the immobilized-cell reactors (93%).

Bacillus sp. MIR32 has been isolated using xylan as the only carbon source, and one of its xylanolytic enzymes has been extensively studied. Biochemical analysis first related this strain to Bacillus amyloliquefaciens, but further studies based on a comparison of 16S rDNA sequences, G+C content, and DNA-DNA hybridization showed that strain MIR32 should be classified as a member of the species Bacillus halodurans. This change is also supported by the typical phenotype observed and by the results of PCR amplification directed toward spacers in rDNA and tDNA genes, which were assayed and compared with those of B. halodurans DSM 497(T). Although among alkaliphilic bacilli competence development has not been experimentally demonstrated, in this work both B. halodurans MIR32 and DSM 497(T) were transformed according to a simple procedure developed in our laboratory, reaching 10(2)-10(3) stable transformants per microgram of plasmid DNA. 相似文献

An integrative vector was constructed to allow expression of heterologous proteins into the adhB locus of Zymomonas mobilis. As a reporter gene, the ORF of a bright variant of green fluorescent protein from Aequorea victoria (GFPuv) was fused to the adhB strong promoter from Z. mobilis by using a two-step PCR strategy. Z. mobilis recombinant strains that were stably marked by precise gene replacement at adhB locus with a single chromosomal copy of gfpuv. Protein expression was confirmed by fluorescence microscopy and measured by fluorescence spectroscopy, showing high expression levels (12 to 30 times higher than those obtained in E. coli) without affecting the host growth. 相似文献

The feasibility of applying the UASB concept for the anaerobic treatment of stillage of distilleries in the sugar producing area of Argentina was subject to study. Results obtained in a 100-L UASB reactor treating stillages with COD values between 35 and 100 g COD/L are presented. Loading rates of up to 24 g COD/L/day were applied with an average COD removal of 75% and a biogas production of more than 9 L/L/day, with an average methane content of 58%. The settling velocity distribution of sludge particles would indicate a good formation of biomass pellets. System interruptions of months without feed and at ambient temperature (20-24 degrees C) were well tolerated. 相似文献

Acinetobacter johnsonii A2 isolated from the natural community of Laguna Azul (Andean Mountains at 4,560 m above sea level), Serratia marcescens MF42, Pseudomonas sp. strain MF8 isolated from the planktonic community, and Cytophaga sp. strain MF7 isolated from the benthic community from Laguna Pozuelos (Andean Puna at 3,600 m above sea level) were subjected to UV-B (3,931 J m−2) irradiation. In addition, a marine Pseudomonas putida strain, 2IDINH, and a second Acinetobacter johnsonii strain, ATCC 17909, were used as external controls. Resistance to UV-B and kinetic rates of light-dependent (UV-A [315 to 400 nm] and cool white light [400 to 700 nm]) and -independent reactivation following exposure were determined by measuring the survival (expressed as CFU) and accumulation of cyclobutane pyrimidine dimers (CPD). Significant differences in survival after UV-B irradiation were observed: Acinetobacter johnsonii A2, 48%; Acinetobacter johnsonii ATCC 17909, 20%; Pseudomonas sp. strain MF8, 40%; marine Pseudomonas putida strain 2IDINH, 12%; Cytophaga sp. strain MF7, 20%; and Serratia marcescens, 21%. Most bacteria exhibited little DNA damage (between 40 and 80 CPD/Mb), except for the benthic isolate Cytophaga sp. strain MF7 (400 CPD/Mb) and Acinetobacter johnsonii ATCC 17909 (160 CPD/Mb). The recovery strategies through dark and light repair were different in all strains. The most efficient in recovering were both Acinetobacter johnsonii A2 and Cytophaga sp. strain MF7; Serratia marcescens MF42 showed intermediate recovery, and in both Pseudomonas strains, recovery was essentially zero. The UV-B responses and recovery abilities of the different bacteria were consistent with the irradiation levels in their native environment. 相似文献
