Abstract The prolylendopeptidase (PEP) is the proteolytic enzyme, which plays an essential role in the regulation of some processes in central nervous system, such as memory, learning and behavior. It was shown that PEP activity changes at different diseases, like Parkinsons or Alzheimer's diseases, and some PEP inhibitors are used in therapy. At present time the discovery of new types of PEP inhibitors are the actual task. In this study the structure of PEP active site was analyzed by 3D-QSAR with CoMFA methods using of 12 PEP substrates. The designed pharmacophore model assumes that substrates interact with PEP active site by pyrrolidol ring of proline residue and by hydrogen bonding. The 3-D-QSAR + CoMFA model of PEP substrates propose that the hydrophobic bonds play the essential role in substrate interaction with enzyme. This model reveals the important steric and electrostatic areas around the molecules and the presence of substituents controls the PEP activity for substrates. Analysis of obtained data allows to assume, that substrate binding in PEP active site causes essential perturbations of substrate structure. This effect mainly depends on chemical nature of the amino acid side chain, located near to proline. 相似文献

Based on materials of trawl studies performed from 1992 to 2002 off the Pacific coast of Kamchatka and the northern Kuril
Islands in the framework of the integrated (VNIRO, KamchatNIRO, Sakh-NIRO) program on the study of fishery resources of the
continental slope, data on the distribution and biology of rock greenling Hexagrammos lagocephalus were analyzed. It was elucidated that this species is extremely eurybathic; the range of its seasonal vertical migrations
embraces depths of 1–2 m in summer, and in the spawning period and up to 665 m in winter. The spatial distribution has a mosaic
pattern, maximum concentrations were recorded at the slope and shelf of the Shumshu Island and the southern extremity of Kamchatka.
The size, weight, and sex composition, and the maturation and feeding of the rock greenling are considered. Maximum fish sizes
in trawl catches are 58 cm and 2.63 kg; sex composition is characterized by the domination of females in all size groups (79%).
The bulk of feeding consisted of Cephalopoda, small crustaceans—Amphipoda and Isopoda—as well as the eggs of fish and Gastropoda.
With an increase in sizes, the greenling passes from feeding on small benthic organisms to larger movable forms. 相似文献

Data on the occurrence, spatial-bathymetric distribution, and size composition of skilfish Erilepis zonifer are given on the basis of the materials collected at the long-line catches on the Emperor Ridge seamounts. This fish is a large representative of fam. Anoplopomatidae (maximal length 188 cm; weight 88.5 kg). From June to July 2009, the species was found on all five sampled underwater mountains (Nintoku, Jingu, Ojin, Koko, Northern Koko). The fish was found within the depth range 370–1040 m. The initial biomass of skilfish at the studied seamounts calculated by the Leslie method is approximately 203.5 tons. The structure of the range and possible ways of the dispersion of this species in the waters of the continental slope of the North-Pacific rim are discussed. 相似文献

A portable optoelectronic reflectometric photocolorimeter was designed. The photocolorimeter implements two operating modes
(transmission and reflection), and its performance was tested in two systems of enzyme immunoassay for testosterone: microplate
ELISA and membrane dot-ELISA. The detection threshold for microplate ELISA and membrane dot-ELISA was 0.1 and 0.6 ng/ml, respectively.
The coefficient of correlation between the photocolorimeter readings and conventional photometric methods is r=0.999. Relative
standard deviation of the results of photocolorimetric measurements (n = 4) within the optical density range of 0.03 to 1.00
ranged from 3.4 to 0.7%. The simple and versatile design of the photocolorimeter makes it appropriate for testing various

Rats with experimental parkinsonism induced by intraperitoneal daily administration of rotenone (2.75 mg/kg) during seven days are characterized by an increased activity of prolyl endopeptidase (PEP; ЕС 3.4.21.26) in serum and a decreased activity of adenosine deaminase (ADA; EC 3.5.4.4) in serum and in the prefrontal cortex. PEP and ADA activities in other brain structures (in the striatum, hypothalamus, and hippocampus) as well as activity of dipeptidyl peptidase IV (DPP-4, CD26; ЕС 3.4.14.5) in serum and in all the brain structures investigated remained unchanged. Afobazole and levodopa, which exhibit antiparkinsonian activity in this model of parkinsonism, decreased the elevated PEP activity in serum and increased ADA activity reduced in the prefrontal cortex of rats with the experimental pathology. At the same time, treatment with these drugs resulted in a decrease of ADA activity in the other brain structures. 相似文献

The effect of antioxidant (AO) of the 3-hydroxypyridin class on microcirculatory system (MC) disorders of rabbits with dyslipoproteinemia (DLP) and its alimentary correction (the standard ration--9 months) was studied. Aorta, the intestinal mesentery microcirculatory bed, the microvessels adrenergic innervation, erythrocyte morphology were examined. The various lipoprotein fractions and lipid peroxidation (LP) products were studied in venous blood. After AO including into atherogenic diet (ATD) the lipid homeostasis disorders, the LP activation, MC disturbances, anomalous erythrocyte form appearance and atherosclerosis changes in aorta were less pronounced. Under the AO influence when DLP was corrected the initial level of lipid metabolism and LP restored, the MC disorders and the atherosclerosis changes in aorta regressed in a shorter period of time. The interrelation was observed between the DLP level, the structural and functional MC disorders and the extent of aorta atherosclerosis. Thus, DLP and LP correction at the early stages of pathological process can lead to the disappearance of MC disorders, what seems important for the prevention of atherosclerosis. The role of the LP activation in MC disorders in DLP and mexidol effect on them, especially in complex with DLP correction were evaluated. 相似文献
