To generate new chromosome 21 markers in a region that is critical for the pathogenesis of Down syndrome (D21S55-MX1), we used pulsed field gel electrophoresis (PFGE) to isolate a 600-kb NruI DNA fragment from the WA17 hybrid cell line, which has retained chromosome 21 as the only human material. This fragment, which contains the oncogene ETS2, was used to construct a partial genomic library. Among the 14 unique sequences that were isolated, 3 were polymorphic markers and contained sequences that are conserved in mammals. Five of these markers mapped on the ETS2-containing NruI fragment and allowed us to define an 800-kb high-resolution PFGE map. 相似文献

Thiopurine S-methyltransferase (TPMT) is an important enzyme that metabolizes thiopurine drugs. This enzyme exhibits a large number of interindividual polymorphism. TPMT?23 polymorphism has been reported in a few cases in the world in co-dominance with TPMT?3A. The phenotype has been reported to affect enzyme activity in vivo and in vitro. Its underlying structural basis is not clarified yet. In our study, the wild type (WT) protein structure was analyzed and the amino acids bordering water channels in thiopurine sites were identified. Molecular dynamics of both the WT and TPMT?23 mutation was carried out. In addition, the effects of this mutation, especially on the thiopurine site which is closed with a pincer like mechanism, were investigated. We focused on explaining how a locally occurred A167G substitution propagated through hydrogen bonds alteration to induce structural modification which affects both thiopurine and S-adenosylmethionine receptors. Finally, a genetic prediction of mutation functional consequences has been conducted confirming altered activity. Azoreductase, an enzyme catalyzing the reductive cleavage of the azo bond of methyl red (MR) and related dyes, was purified to electrophoretic homogeneity from Enterobacter agglomerans. This bacterial strain, isolated from dye-contaminated sludge, has a higher ability to grow, under aerobic conditions, on culture medium containing 100mg/L of MR. The enzyme was purified approximately 90-fold with 20% yield by ammonium sulfate precipitation, followed by three steps of column chromatography (gel-filtration, anion-exchange, and dye-affinity). The purified enzyme is a monomer with a molecular weight of 28,000 Da. The maximal azoreductase activity was observed at pH 7.0 and at 35 degrees C. This activity was NADH dependent. The K(m) values for both NADH and MR were 58.9 and 29.4 microM, respectively. The maximal velocity (V(max)) was 9.2 micromol of NADH min(-1)mg(-1). The purified enzyme is inhibited by several metal ions including Fe(2+) and Cd(2+). 相似文献

The partial sequence of the increasing capillary permeability protein (ICPP) purified from Vipera lebetina venom revealed a strong homology to vascular endothelial growth factor (VEGF)-A. We now report its complete amino acid sequence determined by Edman degradation and its biological effects on mouse and human vascular endothelial cells. ICPP is a homodimeric protein linked by cysteine disulfide bonds of 25115 Da revealed by mass spectrometry. Each monomer is composed of 110 amino acids including eight cysteine residues and a pyroglutamic acid at the N-terminal extremity. ICPP shares 52% sequence identity with human VEGF but lacks the heparin binding domain and Asn glycosylation site. Besides its strong capillary permeability activity, ICPP was found to be a potent in vitro angiogenic factor when added to mouse embryonic stem cells or human umbilical vein endothelial cells. ICPP was found to be as potent as human VEGF165 in activating p42/p44 MAPK, in reinitiation of DNA synthesis in human umbilical vein endothelial cells, and in promoting in vitro angiogenesis of mouse embryonic stem cells. All these biological actions, including capillary permeability in mice, were fully inhibited by 1 microm of a new specific VEGF receptor tyrosine kinase inhibitor (ZM317450) from AstraZeneca that belongs to the anilinocinnoline family of compounds. Indeed, up to a 30 times higher concentration of inhibitor did not affect platelet-derived growth factor, epidermal growth factor, FGF-2, insulin, alpha-thrombin, or fetal calf serum-induced p42/p44 MAPK and reinitiation of DNA synthesis. Therefore, we conclude that this venom-derived ICPP exerts its biological action (permeability and angiogenesis) through activation of VEGF receptor signaling (VEGF-R2 and possibly VEGF-R1). 相似文献

BackgroundThe disease course of human immunodeficiency virus (HIV) is often altered by existing or newly acquired coincident infections.Conclusions/SignificanceWe were unable to find a significant effect of W. bancrofti infection or its treatment on HIV clinical course or surrogate markers of HIV disease progression though we recognized that our study was limited by the smaller than predicted sample size and by the use of ART in half of the patients. Treatment of W. bancrofti coinfection in HIV positive subjects (as is usual in mass drug administration campaigns) did not represent an increased risk to the subjects, and should therefore be considered for PLWHA living in W. bancrofti endemic areas.

Influence of Cross-linked Waxy Maize Starch on the Aggregation Behavior of Casein Micelles During Acid-induced Gelation 总被引:1,自引:0,他引:1
The influence of cross-linked waxy maize starch on the aggregation behavior of casein micelles was investigated using a combination
of physico-chemical techniques. Milk was homogenized at two different temperatures (55 and 65 °C) and then heated at 95 °C
for 5 min in a pilot scale system. The possible interactions between modified starch and milk proteins during lactic acid
fermentation were evaluated. While 1% starch did not show differences in the whey protein complexes formed during heating
compared to milk with no starch (as measured by size exclusion chromatography), a higher (2.5%) concentration of starch clearly
showed an increased amount of heat-induced whey protein aggregates. The gelation pH also increased significantly with 2.5%
starch compared to that of the control samples. The storage modulus (G′) increased with increasing levels of starch, and confocal
microscopy confirmed that the microstructure of the casein gels was altered by the presence of modified starch. Milk-starch
mixtures preheated and homogenized at 55 or 65 °C exhibited similar physico-chemical behavior during acidification. The results
suggested a lack of interaction between starch granules and casein micelles during acidification, and scanning electron microscopy
images collected with a self-assembled monolayer technique also confirmed that starch granules were not attached to milk caseins
but only embedded in the protein gel matrix. 相似文献

We investigated the distribution pattern of centipedes (Chilopoda) in four primeval forests of the western Carpathians, central
Slovakia. The forests are located in two different mountain ranges (Kremnické vrchy and Pol’ana Mountains), which are exposed
on either the southern (Boky, Rohy) or northern slopes (Pol’ana, Badín). In these forests, the influence of coarse woody debris
(CWD) on centipede distribution was studied, by distinguishing sampling sites on the forest floor, close to CWD (c-CWD) and
distant from CWD (d-CWD). In total, we collected 2,706 individuals from 20 species of centipedes. Average species richness
and number of individuals per forest ranged from 8 to 12 species/m2 and from 244 to 486 individuals/m2. The oak forests on south facing slopes harboured several species, which did not occur in the more northern exposed fir-beech
forests. Number of species as well as individuals, however, varied more within than between individual forests. Increase of
species number and density was mainly caused by CWD and was more pronounced on the southern slopes (P<0.001), characterized by high temperatures and low precipitation, than on the northern slopes (n.s. to P<0.01), characterized by low temperatures and high precipitation. It was found that CWD did not generally increase (species)
