The oxidation of methyl linoleate (ML) was studied in the presence of Fe(II) alone and its combination with either ascorbic acid (AsAH(2)) or hydrogen peroxide (H(2)O(2)) at different molar ratios. Reactions were carried out in micellar solutions of TTAB (tetradecyltrimethylammonium bromide) and SDS (sodium dodecyl sulfate), respectively, and were monitored by UV spectroscopy and electrospray ionization mass spectrometry (ESI-MS). Fe(II) alone was able to catalyze the oxidation of ML in micellar solutions of TTAB, but not in those of SDS. The combination of H(2)O(2) with Fe(II) showed catalytic effect only in the TTAB medium, leading to different ML and Fe(II) oxidation kinetics compared to the Fe(II)-only catalyzed reactions. The AsAH(2)/Fe(II) combination demonstrated to be a good catalyst for the oxidation of ML in SDS micellar solutions, but not in TTAB micellar solutions; the activity of the catalyst was dependent on the AsAH(2)/Fe(II) molar ratio. The obtained results confirm that, for the ML oxidation to be initiated, the presence of a Fe(II)/Fe(III) couple is essential, which is related to the pH of micellar solutions. The catalytic properties of the AsAH(2)/Fe(II) combination were explained by taking into account the anti-oxidant and pro-oxidant properties of AsAH(2), as well as the possible formation of an iron/ascorbate complex as the initiator of the ML oxidation.  相似文献

A new calanoid copepod species Scaphocalanus emine n. sp. is described for the first time from the Iskenderun Bay, northeastern Levantine Basin. S. emine is similar to S. affinis (Sars, 1905) having both identical orientation of the posterolateral corners of the cephalothorax and a median crest on the forehead. The new species is also similar in some features to S. affinis in the structure of Re1P1- lacking the outer marginal spine and by the presence of the rounded apex on the distal internal margin. It differs from S. affinis as the median crest on the forehead is smaller and due to the presence of a rudimentary endopod furnished with an apical spine on the P5. Originality in construction of the P1 and P5 distinguish the new species from other Scaphocalanus species.  相似文献

Cytosolic phospholipase A2 (cPLA2)-generated arachidonic acid (AA) has been shown to be an essential requirement for the activation of NADPH oxidase, in addition to its being the major enzyme involved in the formation of eicosanoid at the nuclear membranes. The mechanism by which cPLA2 regulates NADPH oxidase activity is not known, particularly since the NADPH oxidase complex is localized in the plasma membranes of stimulated cells. The present study is the first to demonstrate that upon stimulation cPLA2 is transiently recruited to the plasma membranes by a functional NADPH oxidase in neutrophils and in granulocyte-like PLB-985 cells. Coimmunoprecipitation experiments and double labeling immunofluorescence analysis demonstrated the unique colocalization of cPLA2 and the NADPH oxidase in plasma membranes of stimulated cells, in correlation with the kinetic burst of superoxide production. A specific affinity in vitro binding was detected between GST-p47phox or GST-p67phox and cPLA2 in lysates of stimulated cells. The association between these two enzymes provides the molecular basis for AA released by cPLA2 to activate the assembled NADPH oxidase. The ability of cPLA2 to regulate two different functions in the same cells (superoxide generation and eicosanoid production) is achieved by a novel dual subcellular localization of cPLA2 to different targets.  相似文献

Phycoerythrin-containing unicellular cyanobacteria Synechococcusspp. were studied for the first time during April–May,1994 and September–October, 1996, in the western and southernBlack Sea for pigments, size and abundance distribution viaspectrometry, epifluorescence microscopy and flow cytometry.Abundance distribution in the surface mixed layer in April–May,1994, revealed that cells were more concentrated in offshorewaters than in coastal regions under the direct influence ofthe river Danube. However, in the south, higher surface cellconcentrations were characteristic of the nearshore areas duringSeptember–October, 1996. A highly significant correlationwas observed between cell abundance and ambient physico-chemicalparameters with depth. Visual inspection of the individual cellsunder the epifluorescence microscope revealed that cells atthe subsurface, chlorophyll a maximum layer (SCML, based onin situ fluorometer readings) fluoresce more brightly and forlonger than those at the surface and at lower depths. Spectralproperties of a total of 64 Synechococcus spp. clonal isolatesfrom different depths within the euphotic layer (about the top60 m) in the southern Black Sea coast showed that all have type2 phycoerythrobilin in common, lacking phycourobilin. In vivofluorescence emission maxima for phycoerythrobilin were aboutthe same (~578 nm) for all isolates. All isolates had in vivoabsorption maxima at between 435 and 442 nm, and at about 681nm due to chlorophyll a. It was shown from the flow cytometermean forward light scatter data for size distribution that cellsat the surface mixed layer (0–10 m) were larger than cellsat lower depths (20–60 m). Based on in vivo fluorescencemeasurements, significant differences in the acclimated growthrates of clones from different depths were observed. Time versuscell count plots showed that cells of the cyanobacteria Synechococcusspp. are under grazing pressure, from midnight until noon, andslowly begin to rebuild their population in the afternoon bydividing throughout the evening.  相似文献

Species composition, community structure and spatial distribution of net phytoplankton in the southern Black Sea have been studied. In the fraction over 55 m, diatoms were found to be predominant in cell abundance and followed by dinoflagellates. Net phytoplankters were found to be more abundant in the southwestern sector, especially near the Bosporus and near Sakarya River mouth. MDS (Multi-Dimensional Scaling) analysis has projected four distinct patches along the coast, two of which appear to occupy smaller regions compared to the main western and eastern patches which have lower species numbers and diversity. The diatom species Pseudo-nitzschia seriata and Cerataulina pelagica and the dinoflagellates Noctiluca scintillans and Scrippsiella trochoidea dominate the west, while diatom species Synedra acus and Asterionella formosa together with Microspora sp. and Ankistrodesmus falcatus from chlorophyceae dominate the east.  相似文献
