Oxidative stress has been linked to the pathogenesis of diabetic nephropathy, the complication of diabetes in the kidney. NADPH oxidases of the Nox family, and in particular the homologue Nox4, are a major source of reactive oxygen species in the diabetic kidney and are critical mediators of redox signaling in glomerular and tubulointerstitial cells exposed to the diabetic milieu. Here, we present an overview of the current knowledge related to the understanding of the role of Nox enzymes in the processes that control mesangial cell, podocyte and tubulointerstitial cell injury induced by hyperglycemia and other predominant factors enhanced in the diabetic milieu, including the renin-angiotensin system and transforming growth factor-β. The nature of the upstream modulators of Nox enzymes as well as the downstream targets of the Nox NADPH oxidases implicated in the propagation of the redox processes that alter renal biology in diabetes will be highlighted. 相似文献

It has been shown that lens regeneration from the iris of the newt Notophthalmus viridescens is dependent on the presence of neural retinal tissue in organ culture and in vivo. The recent discovery of various eye-derived growth factors (EDGFs) in the bovine retina [14] prompted us to investigate whether one of these factors may be involved in the stimulation of lens regeneration. Dorsal irises were cultured for 20 days in serum-supplemented diluted Eagle's medium. Growth factors from bovine retina of various degrees of purification were added. Lens regeneration was assessed on the basis of morphological lens-regeneration stages and by immunofluorescent detection of a lens-specific marker protein, alpha-crystallin. Crude isotonic retinal extract at 80-800 micrograms/ml significantly augmented lens regeneration. Very similar results were obtained when EDGF III, the nonretained retinal factor after heparin-affinity chromatography, was present at 2-20 micrograms/ml. Lens regeneration was also significantly increased when EDGF II, the retinal form of acidic fibroblast growth factor (aFGF) at 50-500 ng/ml was added to the cultures. On the other hand, EDGF I at 4-40 ng/ml and brain basic FGF at 5-50 ng/ml did not seem to significantly stimulate lens regeneration under the conditions used. Our results suggest that at least two retina-derived growth factors (EDGF II and III) can stimulate lens regeneration. A selection of normal human tissues was investigated for the presence of lamins B1, B2, and A-type lamins, using a panel
of antibodies specific for the individual lamin subtypes. By use of immunoprecipitation and two-dimensional immunoblotting
techniques we demonstrated that these antibodies do not cross-react with other lamin subtypes and that a range of different
phosphorylation isoforms is recognized by each antibody. The lamin B2 antibodies appeared to decorate the nuclear lamina in
all tissues examined, except hepatocytes, in which very little lamin B2 expression was observed. In contrast to previous studies,
which suggested the ubiquitous expression of lamin B1 in mammalian tissues, we show that lamin B1 is not as universally distributed
throughout normal human tissues as was to be expected from previous studies. Muscle and connective tissues are negative, while
in epithelial cells lamin B1 seemed to be preferentially detected in proliferating cells. These results correspond well with
those obtained for lamin B1 in chicken tissues. The expression of A-type lamins is most prominent in well-differentiated epithelial
cells. Relatively undifferentiated and proliferating cells in epithelia showed a clearly reduced expression of A-type lamins.
Furthermore, most cells of neuroendocrine origin as well as most hematopoietic cells were negative for A-type lamin antibodies.

Fractional anisotropy (FA) is an effective marker of motor outcome at the chronic stage of stroke yet proves to be less efficient at early time points. This study aims to determine which diffusion metric in which location is the best marker of long-term stroke outcome after thrombolysis with diffusion tensor imaging (DTI) at 24 hours post-stroke. Twenty-eight thrombolyzed patients underwent DTI at 24 hours post-stroke onset. Ipsilesional and contralesional FA, mean (MD), axial (AD), and radial (RD) diffusivities values were calculated in different Regions-of-Interest (ROIs): (1) the white matter underlying the precentral gyrus (M1), (2) the corona radiata (CoRad), (3) the posterior limb of the internal capsule (PLIC) and (4) the cerebral peduncles (CP). NIHSS scores were acquired at admission, day 1, and day 7; modified Rankin Scores (mRS) at 3 months. Significant decreases were found in FA, MD, and AD of the ipsilesional CoRad and M1. MD and AD were also significantly lower in the PLIC. The ratio of ipsi and contralesional AD of the CoRad (CoRad-rAD) was the strongest diffusion parameter correlated with motor NIHSS scores on day 7 and with the mRS at 3 months. A Receiver-Operator Curve analysis yielded a model for the CoRad-rAD to predict good outcome based on upper limb NIHSS motor scores and mRS with high specificity and sensitivity. FA values were not correlated with clinical outcome. In conclusion, axial diffusivity of the CoRad from clinical DTI at 24 hours post-stroke is the most appropriate diffusion metric for quantifying stroke damage to predict outcome, suggesting the importance of early axonal damage. 相似文献

The population ecology of the beetle Speonomus hydrophilus , occurring both in caves (reduced fluctuations in many abiotic parameters) and under the deepest layer of soil in mountains (MSS, more exposed to climatic variations), was studied in four habitats in the French central Pyrenees We have assessed some of the characteristics of the environment where these populations occur c g physical data (altitude and exposure), geologic data (nature of the parent-rock) and abiotic parameters (temperature with its seasonal fluctuations) and we investigated the relative importance of environmental structure and ecological characteristics on the temporal organization of S hydrophilus and the troglobitic fauna which cohabits The climatic study shows the existence of an annual thermal cycle which is regular and well marked for the MSS habitats but slightly out of phase with the surface cycle These periodic variations however slight may be stressful for troglobitic species In the MSS populations, the phenology of the entire community is reflected in the pattern seen in Speonomus The analysis of faunal profiles shows that samples follow the same seasonal succession during the annual cycle A potential seasonal rhythm of emergence may reflect a seasonal rhythm of vitellogenesis which produces a rhythm of egg-laying 相似文献

Congenital amusia is a neurogenetic disorder of music processing that is currently ascribed to a deficit in pitch processing. A recent study challenges this view and claims the disorder might arise as a consequence of a general spatial-processing deficit. Here, we assessed spatial processing abilities in two independent samples of individuals with congenital amusia by using line bisection tasks (Experiment 1) and a mental rotation task (Experiment 2). Both amusics and controls showed the classical spatial effects on bisection performance and on mental rotation performance, and amusics and controls did not differ from each other. These results indicate that the neurocognitive impairment of congenital amusia does not affect the processing of space. 相似文献

Dealing with in situ sediments is also dealing with our own identity. Our established confidence in scientific technical and economic progress and prosperity is undergoing a painful awakening. After all, sediments and pollution are a symbol of the destruction of our environment rather than an improvement of a quality of life. We are starting to see the wholeness of the issue. While in the past, manipulation of the environment left a few human concerns which were treated as residues of a view of progress, we are now faced with the fact that all environmental issues are human issues and that preoccupations with values, aspirations and interdependency need to be at center stage.Pollution issues become a disturbance of the perfect harmony between human beings and nature, and between different human groups. The restoration of the harmony requires a revision of paradigms, values and the interactivity of systems. Ultimately, the choices will be made on the basis of very many concerns. The rational and the irrational will combine to form a series of remedial measures that may not satisfy the traditional view of progress nor the newly acquired vision of a restorated environment. However, if communities are strengthened by the experience, if physical and social sciences as well as humanities find a new vision of their interdependency in the process, maybe the present state of high apathy, isolation and powerlessness will have been replaced with a better set of values with which to assess the problem and establish our common goal.If scientists, technocrats and decision-makers can come together in other places like this gathering, I believe that the momentum will lead to bringing together communities. Any such interaction inevitably creates its own dynamics so that interacting becomes the mechanism by which one experiences and through which one's choices are molded by each other's. Summary We constructed a cDNA library for the beetle,Tribolium castaneum. This library was screened using a cloned amylase gene fromDrosophila melanogaster as a molecular probe. Beetle amylase cDNA clones were isolated from this bank, and the nucleotide sequence was obtained for a cDNA clone with a coding capacity for 228 amino acids. Both the nucleotide sequence and predicted amino acid sequence were compared to our recent results forD. melanogaster alpha-amylases, along with published sequences for other alpha-amylases. The results show that animal alpha-amylases are highly conserved over their entire length. A borader comparison, which includes plant and microbial alpha-amylase sequences, indicates that parts of the gene are conserved between prokaryotes, plants, and animals. We discuss the potential importance of this and other enzyme-coding genes for the construction of molecular phylogenies and for the study of the general question of molecular clocks in evolution. 相似文献
