The enigmatic arrow worms (Chaetognatha) are marine carnivores and among the most abundant planktonic organisms. Their phylogenetic
position has been heavily debated for a long time. Most recent molecular studies still provide a diverging picture and suggest
arrow worms to be some kind of basal protostomes. In an effort to understand the organization of the nervous system in this
clade for a broad comparison with other Metazoa we analysed the ultrastructure of the ventral nerve centre in Spadella cephaloptera by transmission electron microscopy. We were able to identify six different types of neurons in the bilateral somata clusters
by means of the cytoplasmic composition (regarding the structure of the neurite and soma including the shape and eu-/heterochromatin
ratio within the nucleus) as well as the size and position of these neurons. Furthermore, our study provides new insights
into the neuropil composition of the ventral nerve centre and several other fine structural features. Our second goal was
to examine if individually identifiable neurons are present in the ventral nerve centres of four chaetognath species, Sagitta setosa, Sagitta enflata, Pterosagitta draco, and Spadella cephaloptera. For that purpose, we processed whole mount specimens of these species for immunolocalization of RFamide-related neuropeptides
and analysed them with confocal laser-scanning microscopy. Our experiments provide evidence for the interspecific homology
of individual neurons in the ventral nerve centres of these four chaetognath species suggesting that the potential to generate

A wide range of plant lines has been propagated by farmers during crop selection and dissemination, but consequences of this crop diversification on plant-microbe interactions have been neglected. Our hypothesis was that crop evolutionary history shaped the way the resulting lines interact with soil bacteria in their rhizospheres. Here, the significance of maize diversification as a factor influencing selection of soil bacteria by seedling roots was assessed by comparing rhizobacterial community composition of inbred lines representing the five main genetic groups of maize, cultivated in a same European soil. Rhizobacterial community composition of 21-day-old seedlings was analysed using a 16S rRNA taxonomic microarray targeting 19 bacterial phyla. Rhizobacterial community composition of inbred lines depended on the maize genetic group. Differences were largely due to the prevalence of certain Betaproteobacteria and especially Burkholderia, as confirmed by quantitative PCR and cloning/sequencing. However, these differences in bacterial root colonization did not correlate with plant microsatellite genetic distances between maize genetic groups or individual lines. Therefore, the genetic structure of maize that arose during crop diversification (resulting in five main groups), but not the extent of maize diversification itself (as determined by maize genetic distances), was a significant factor shaping rhizobacterial community composition of seedlings. 相似文献

The present study investigates the effects of a chronic administration of diazepam, a benzodiazepine widely used as an anxiolytic, on locomotor activity and body core temperature rhythms in male Wistar rats housed under 12:12 light:dark (LD) cycle conditions. Diazepam was administered subcutaneously for 3 wks in a dosage of 3 mg/kg body weight/day, 1 h before the onset of darkness. Diazepam increased the level of locomotor activity from the first day until the end of treatment, and also increased the amplitude of the activity circadian rhythm, but only on the third wk of treatment. Diazepam exerted no effects on the length of the period and did not affect the phase of the locomotor activity rhythm. The body temperature rhythm of rats was affected neither by short-term (a single injection) nor by long-term (every day for 3 wks) diazepam treatment. Diazepam lacked effect on body core temperature even on the first day of administration, thereby ruling out the possibility of drug tolerance development. The fact that diazepam affects locomotor activity, but not core body temperature, suggests that different mechanisms mediate the actions of diazepam on locomotor activity and on core body temperature. 相似文献

Thecosomata is a marine zooplankton group, which played an important role in the carbonate cycle in oceans due to their shell composition. So far, there is important discrepancy between the previous morphological-based taxonomies, and subsequently the evolutionary history of Thecosomata. In this study, the remarkable planktonic sampling of TARA Oceans expedition associated with a set of various other missions allowed us to assess the phylogenetic relationships of Thecosomata using morphological and molecular data (28 S and COI genes). The two gene trees showed incongruities (e.g. Hyalocylis, Cavolinia), and high congruence between morphological and 28S trees (e.g. monophyly of Euthecosomata). The monophyly of straight shell species led us to reviving the Orthoconcha, and the split of Limacinidae led us to the revival of Embolus inflata replacing Limacina inflata. The results also jeopardized the Euthecosomata families that are based on plesiomorphic character state as in the case for Creseidae which was not a monophyletic group. Divergence times were also estimated, and suggested that the evolutionary history of Thecosomata was characterized by four major diversifying events. By bringing the knowledge of palaeontology, we propose a new evolutionary scenario for which macro-evolution implying morphological innovations were rhythmed by climatic changes and associated species turn-over that spread from the Eocene to Miocene, and were shaped principally by predation and shell buoyancy. 相似文献

Photosynthetic activity of developing leaves of Zea mays is less affected by heat stress than that of developed leaves 总被引:9,自引:0,他引:9
Various physiological and biochemical characters of a leaf change with stages of its ontogeny. It is likely that the photosynthetic functions of leaves of different ontogeny have different levels of heat tolerance. This study was initiated to analyze the photosynthetic heat tolerance of fully-developed, nearly-developed (more than 2/3 expanded) and developing (10–12 cm visible) leaves of two maize genotypes, F223 and F250. The results indicate that the photosynthetic CO2 assimilation rate (Pn ) of developing leaves was less affected by heat stress (42°C in the dark for 90 min) than that of developed leaves. The impaired Pn recovered within 24 h in the developing leaves, while the Pn of developed and nearly-developed leaves did not reach the non-stress level, even after 72 h. The Pn of the developed leaves of genotype F250 was less affected by heat stress than that of genotype F223. After heat stress, the slightly affected Pn of the developing leaf was associated with the almost unchanged photochemical efficiency of photosystem II (Fv /Fm ) and the quantum yield of photosystem II electron transport. The chlorophylls a and b were degraded by heat stress; the degradation was pronounced in the developed leaves. As a result of heat stress, the antheraxanthin and zeaxanthin of the xanthophyll cycle accumulated in both the nearly-developed and developed leaves but not in the developing leaves. Injury to the plasma membrane due to heat stress was much less severe in developing leaves than that in the developed leaves. From the physiological characters which we determined it would appear that the Pn functions of the developing leaves are more resistant to heat stress than those of nearly-developed and developed leaves. 相似文献

OBJECTIVE: To determine whether immunocytochemistry (ICC) for HER2 on ThinPrep (TP)-processed breast fine needle aspiration biopsies (Cytyc Corp., Boxborough, Massachusetts, U.S.A.) is comparable to the findings of immunohistochemistry on corresponding surgically removed tissue. STUDY DESIGN: Immunostaining was performed on 63 malignant breast fine needle aspirates and compared to immunostaining on paraffin sections (PSs) from the subsequent biopsies. The HercepTest (Dako, Carpinteria, California, U.S.A.) and TAB250 antibodies were utilized. Cases in which the TP and paraffin HER2 results did not correlate were further assessed for gene amplification by differential polymerase chain reaction (dPCR). RESULTS: HER2 overexpression was found in 9 of the 63 cases (14%). TAB250 had higher specificity on PS versus TP (P = .008), and TAB250 had higher specificity on PS versus the HercepTest on PS and TP (P = .004 and .0001, respectively). CONCLUSION: HER2 immunostaining with both the HercepTest and TAB250 on TP is unreliable due to low specificity (72% and 83% for HercepTest and TAB250, respectively). However, both antibodies have high sensitivity (89% and 100%, respectively); suggesting that this method may have some utility as a preliminary screening test for HER2 status. Negative HER2 staining by ICC is highly predictive of the absence of HER2 overexpression, whereas positive HER2 staining on TP would require further validation by either dPCR of fluorescence in situ hybridization. 相似文献
