Reports suggest that microglia play a key role in spinal nerve ligation (SNL)-induced neuropathic pain, and toll-like receptor 3 (TLR3) has a substantial role in the activation of spinal microglia and the development of tactile allodynia after nerve injury. In addition, ketamine application could suppress microglial activation in vitro, and ketamine could inhibit proinflammatory gene expression possibly by suppressing TLR-mediated signal transduction. Therefore, the present study was designed to disclose whether intrathecal ketamine could suppress SNL-induced spinal microglial activation and exert some antiallodynic effects on neuropathic pain by suppressing TLR3 activation. Behavioral results showed that intrathecal ketamine attenuated SNL-induced mechanical allodynia, as well as spinal microglial activation, in a dose-dependent manner. Furthermore, Western blot analysis displayed that ketamine application downregulated SNL-induced phosphorylated-p38 (p-p38) expression, which was specifically expressed in spinal microglia but not in astrocytes or neurons. Besides, ketamine could reverse TLR3 agonist (polyinosine-polycytidylic acid)-induced mechanical allodynia and spinal microglia activation. It was concluded that intrathecal ketamine depresses TLR3-induced spinal microglial p-p38 mitogen-activated protein kinase pathway activation after SNL, probably contributing to the antiallodynic effect of ketamine on SNL-induced neuropathic pain.  相似文献

A sensitive, rapid liquid chromatographic-electrospray ionization mass spectrometric method for determination of erythromycylamine in human plasma was developed and validated. Erythromycylamine in plasma (0.2 mL) was extracted with ethyl acetate, the organic phase was transferred to another clear 1.5 mL Eppendorf tube and evaporated to dryness under gentle nitrogen stream at 45 degrees C, and the residue was dissolved in 100 microL of mobile phase. The samples were separated using a Thermo Hypersil HyPURITY C18 reversed-phase column (150 mm x 2.1 mm I.D., 5 microm). A mobile phase containing 10 mM of ammonium acetate (pH = 6.4)-acetonitrile-methanol (50:10:40, v/v/v) was used isocratically eluting at a flow rate of 0.2 mL/min. Erythromycylamine and its internal standard (IS), midecamycin, were measured by electrospray ion source in positive selective ion monitoring mode. The method demonstrated that good linearity ranged from 4.5 to 720 ng/mL with r = 0.9997. The limit of quantification for erythromycylamine in plasma was 4.5 ng/mL with good accuracy and precision. The mean extraction recovery of the method was higher than 75.1% and 72.7% for erythromycylamine and IS, respectively. The intra-day and inter-day precision ranged from 5.2% to 6.4% and 5.6-9.3% (relative standard deviation, RSD), respectively. The established method has been successfully applied to a bioequivalence study of two dirithromycin formulations for 18 healthy volunteers.  相似文献

Agouti 有多方面的意义.首先,它是分属几种不同物种的哺乳动物的俗称.其次,它是一些啮齿类动物的属名.第三,它代表了一种啮齿类动物特有的毛色.第四,它是一个与黑素皮质素受体信号转导途径的调控有关,存在于从人(Homo sapiens)到啮齿类动物,乃至斑马鱼(Danio rerio)的一个特定基因的名称,又是该基因产生的一种特定信号蛋白的名称.Agouti和Agouti相关蛋白是肿瘤、肥胖与糖尿病等人类疾病的研究热点之一.Agouti黄色肥胖小鼠最近成了表观遗传学研究的一个重要模型动物.本文阐述并区分了与agouti有关的一些重要概念与科学术语,并综述了相关领域的研究进展.  相似文献

为了确定豆科作物作为林果行间套种作物的适宜性, 于2014年通过田间试验和盆栽试验对4种豆科作物的耐阴能力及其光合生理进行了研究。测定全光和弱光处理(全光的48%)下豆科作物在初花期的叶片光合特征参数、光合色素含量与核酮糖-1,5-二磷酸羧化酶(RuBPCase)活性, 监测其生长发育对弱光的响应。结果表明: 弱光使供试作物的最大净光合速率、光饱和点、光补偿点、表观量子效率和暗呼吸速率不同程度地向耐阴植物变化; 弱光还使供试作物的净光合速率、气孔导度、蒸腾速率、瞬时水分利用效率和RuBPCase活性不同程度地下降, 而胞间CO2浓度显著上升; 遮阴后, 乌豇豆(Vigna cylindrica)和绿豆(Vigna radiata)的叶绿素(Chl) a和Chl b含量显著增加, Chl a/b值显著降低, 大猪屎豆(Crotalaria assamica)和望江南(Senna occidentalis)的光合色素含量也有不同程度的变化; 弱光使供试作物茎变细, 侧枝数减少, 生物量和干物质积累效率降低, 根冠比降低, 根瘤量减少, 叶片变小变薄, 叶片数减少, 叶面积指数降低; 弱光胁迫下, 大猪屎豆不开花, 望江南只开花不结实, 而绿豆和乌豇豆开花数减少, 花期缩短, 种子产量显著降低。根据供试作物的光合特性和生长发育对弱光的响应, 得出它们的耐阴能力排序是: 乌豇豆>绿豆>望江南>大猪屎豆, 其中乌豇豆和绿豆适宜与林果套种, 而望江南和大猪屎豆不适宜套种。  相似文献

Enkephalinergic (ENKergic) neurons have been proposed to play crucial roles in pain modulation in the trigeminal subnucleus caudalis (Vc). To assist an advance in the research of ENKergic neurons, here we used preproenkephalin-green fluorescent protein (PPE-GFP) transgenic mice, in which all ENKergic neurons were fluorescent. We first performed fluorescent in situ hybridization combined with immunofluorescent histochemistry to confirm the specificity of this transgenic mouse and its advantages in showing ENKergic neurons in the Vc. Then based on this useful transgenic mouse, we examined the phenotypic diversity of PPE-GFP neurons by immunostaining for several markers that characterize ENKergic neuron subtypes. About 25.9±1.9% of GFP-positive neurons were regarded as immunoreactive for glutamic acid decarboxylase (GAD)(67) mRNA and 14.7±1.4% of GFP-positive neurons were positive for γ-aminobutyric acid. The proportions of calbindin-, calretinin-positive cells among the ENKergic neurons were 8.4±1.2% and 7.3±1.7%, respectively. Only 1.1±0.1% of GFP-positive neurons colocalized with parvalbumin and no GFP-positive neurons were found to co-express neuronal nitric oxide synthase. We then injected retrograde tracer into the thalamic regions and observed that a small number of ENKergic neurons in the Vc were retrogradely labeled with the tracer. The present results provide a detailed morphological evidence of the neurochemical features of ENKergic neurons. These results have broad implications for understanding the functional roles of ENKergic neurotransmission in the Vc.  相似文献

Diabetic neuropathic pain (DNP) is one of the most common clinical manifestations of diabetes mellitus (DM), which is characterized by prominent mechanical allodynia (DMA). However, the molecular mechanism underlying it has not fully been elucidated. In this study, we examined the spatio-temporal expression of a major nociceptive channel protein transient receptor potential vanilloid 1 (TRPV1) and analyzed its functional involvement by intrathecal (i.t.) application of TRPV1 antagonists in streptozocin (STZ)-induced DMA rat models. Western blot and immunofluorescent staining results showed that TRPV1 protein level was significantly increased in the soma of the dorsal root ganglion (DRG) neurons on 14 days after STZ treatment (DMA 14 d), whereas those in spinal cord and skin (mainly from the central and peripheral processes of DRG neurons) had already been enhanced on DMA 7 d to peak on DMA 14 d. qRT-PCR experiments confirmed that TRPV1 mRNA level was significantly up-regulated in the DRG on DMA 7 d, indicating a preceding translation of TRPV1 protein in the soma but preferential distribution of this protein to the processes under the DMA conditions. Cell counting assay based on double immunostaining suggested that increased TRPV1-immunoreactive neurons were likely to be small-sized and CGRP-ergic. Finally, single or multiple intrathecal applications of non-specific or specific TRPV1 antagonists, ruthenium red and capsazepine, at varying doses, effectively alleviated DMA, although the effect of the former was more prominent and long-lasting. These results collectively indicate that TRPV1 expression dynamically changes during the development of DMA and this protein may play important roles in mechanical nociception in DRG neurons, presumably through facilitating the release of CGRP.  相似文献

通过长双岐杆菌TTF活菌、菌体破碎物和发酵上清液3种处理物灌胃正常小鼠和免疫功能低下小鼠试验发现,长双歧杆菌TTF的3种受试物对环磷酰胺(Cy)造成的免疫功能低下小鼠模型(IDMM)3项免疫指标均有显著(P〈0.05)或极显著(P〈0.01)影响,且在本试验选取的3个剂量范围内基本呈量效关系。B.longum TTF的3种受试物对IDMM的非特异性免疫功能的影响普遍大于对小鼠的细胞免疫和体液免疫的影响;对正常小鼠免疫功能影响的水平低于其对IDMM各项免疫指标的影响,且无论活菌、菌体破碎物、发酵上清液均不会改变小鼠自身正常的免疫功能。急性毒性试验表明,B.longum,TTF3种处理物对健康小鼠无急性毒性。就3种处理物来讲,发酵上清液组的效果高于菌体破碎物和活菌。  相似文献

单链构象多态性(SSCP)技术是一种分析环境微生物遗传多态性的有效方法,具有快速、简便、灵敏等特点.但是,该技术用于分析复杂环境微生物群落结构时需要有针对性地优化操作条件.本研究针对聚丙烯酰胺凝胶电泳(PAGE)胶浓度、上样缓冲液中去离子甲酰胺浓度、电泳温度,电泳时间等确定了PCR-SSCP技术的优化条件,并以自养脱硫反硝化反应器启动期的活性污泥样品对此优化条件进行了检验,证明了16S rDNA V1~V3区为靶对象时,丙烯酰胺与甲叉的质量比49:1,质量分数12%的聚丙烯酰胺凝胶,上样缓冲液中去离子甲酰胺的体积分数1/3,在4℃,300 V,电泳18 h是最优的操作条件,可以获较理想的SSCP图谱.  相似文献
