Summary We have constructed promoter probe vectors with the Escherichia coli galactokinase monitoring system that can be used in Bacillus subtilis. In vivo studies with these vectors demonstrated that the E. coli trp and tac(trp::lac) promoter regions could be utilized in B. subtilis. These promoter regions and the promoter region for the erythromycin resistance gene originating from Staphylococcus aureus were preferentially utilized during the stationary growth phase of B. subtilis, whereas the B. subtilis P21K and P29K promoters were utilized during the exponential growth phase and decreased rapidly during the stationary phase. The apparent strength of these promoters of E. coli in B. subtilis, in terms of galactokinase units, was comparable with those of the B. subtilis promoters.  相似文献

From a human fetal-brain cDNA library we isolated two novel genes encoding peptides containing six EGF-like repeats. Both showed significant homologies with nel, a gene strongly expressed in neural tissues of chicken. The cDNAs, designated NELL1 (nel-like, type 1) and NELL2 (nel-like, type 2), contained open reading frames encoding 810 and 816 amino acids, respectively. NELL2 is strongly expressed in brain of adult and fetus but only weakly in fetal kidney. NELL1 and NELL2 were mapped by FISH to chromosomal bands 11p15.1–p15.2 and 12q13.11–q13.12, respectively.  相似文献

Rhodococcus (opacus) erythropolis HL PM-1 grows on 2,4,6-trinitrophenol or 2,4-dinitrophenol (2,4-DNP) as a sole nitrogen source. The NADPH-dependent F420 reductase (NDFR; encoded by npdG) and the hydride transferase II (HTII; encoded by npdI) of the strain were previously shown to convert both nitrophenols to their respective hydride Meisenheimer complexes. In the present study, npdG and npdI were amplified from six 2,4-DNP degrading Rhodococcus spp. The genes showed sequence similarities of 86 to 99% to the respective npd genes of strain HL PM-1. Heterologous expression of the npdG and npdI genes showed that they were involved in 2,4-DNP degradation. Sequence analyses of both the NDFRs and the HTIIs revealed conserved domains which may be involved in binding of NADPH or F420. Phylogenetic analyses of the NDFRs showed that they represent a new group in the family of F420-dependent NADPH reductases. Phylogenetic analyses of the HTIIs revealed that they form an additional group in the family of F420-dependent glucose-6-phosphate dehydrogenases and F420-dependent N5,N10-methylenetetrahydromethanopterin reductases. Thus, the NDFRs and the HTIIs may each represent a novel group of F420-dependent enzymes involved in catabolism.  相似文献

An enzyme, dUTPase, that catalyzes the conversion of dUTP to dUMP and PPi, was partially purified from regenerating rat livers. The molecular weight was estimated by gel filtration to be 60,000. The apparent Km for dUTP was 12 μM. No other deoxyribonucleoside triphosphates served as a substrate. This enzyme is active in the absence of added divalent cations or sulfhydryl reagents; the activity could be inhibited by EDTA and shows a broad pH optimum with no decrease in activity from pH 7 to 11. The specific activity of dUTPase in rat liver begins to rise 16 h after partial hepatectomy and reaches a maximum about 24 h after the operation, rising to at least 5 to 6 times the normal level.  相似文献

In Arabidopsis thaliana the ANGUSTIFOLIA (AN) gene regulates the width of leaves by controlling the diffuse growth of leaf cells in the medio‐lateral direction. In the genome of the moss Physcomitrella patens, we found two normal ANs (PpAN1‐1 and 1‐2). Both PpAN1 genes complemented the A. thaliana an‐1 mutant phenotypes. An analysis of spatiotemporal promoter activity of each PpAN1 gene, using transgenic lines that contained each PpAN1‐promoter– uidA (GUS) gene, showed that both promoters are mainly active in the stems of haploid gametophores and in the middle to basal region of the young sporophyte that develops into the seta and foot. Analyses of the knockout lines for PpAN1‐1 and PpAN1‐2 genes suggested that these genes have partially redundant functions and regulate gametophore height by controlling diffuse cell growth in gametophore stems. In addition, the seta and foot were shorter and thicker in diploid sporophytes, suggesting that cell elongation was reduced in the longitudinal direction, whereas no defects were detected in tip‐growing protonemata. These results indicate that both PpAN1 genes in P. patens function in diffuse growth of the haploid and diploid generations but not in tip growth. To visualize microtubule distribution in gametophore cells of P. patens, transformed lines expressing P. patens α‐tubulin fused to sGFP were generated. Contrary to expectations, the orientation of microtubules in the tips of gametophores in the PpAN1‐1/1‐2 double‐knockout lines was unchanged. The relationships among diffuse cell growth, cortical microtubules and AN proteins are discussed.  相似文献

Temperature-dependent regulation of affinity binding between bioactive ligands and their cell membrane receptors is an attractive approach for the dynamic control of cellular adhesion, proliferation, migration, differentiation, and signal transduction. Covalent conjugation of bioactive ligands onto thermoresponsive poly(N-isopropylacrylamide) (PIPAAm)-grafted surfaces facilitates the modulation of one-on-one affinity binding between bioactive ligands and cellular receptors by changing temperature. For the dynamic control of the multivalent affinity binding between heparin and heparin-binding proteins, thermoresponsive cell culture surface modified with heparin, which interacts with heparin-binding proteins such as basic fibroblast growth factor (bFGF), has been proposed. Heparin-functionalized thermoresponsive cell culture surface induces (1) the multivalent affinity binding of bFGF in active form and (2) accelerating cell sheet formation in the state of shrunken PIPAAm chains at 37°C. By lowering temperature to 20°C, the affinity binding between bFGF and immobilized heparin is reduced with increasing the mobility of heparin and the swollen PIPAAm chains, leading to the detachment of cultured cells. Therefore, heparin-functionalized thermoresponsive cell culture surface was able to enhance cell proliferation and detach confluent cells as a contiguous cell sheet by changing temperature. A cell cultivation system using heparin-functionalized thermoresponsive cell culture surface is versatile for immobilizing other heparin-binding proteins such as vascular endothelial growth factor, fibronectin, antithrombin III, and hepatocyte growth factor, etc. for tuning the adhesion, growth, and differentiation of various cell species.  相似文献

A bacterium, (El7), has been found to form new amino acid, when grown in well aerated medium containing ethanol, n-propanol or n-butanol as a sole source of carbon. The new compounds were isolated and purified and their structures were investigated by elementary analyses, infrared spectra, NMR analyses, mass spectra and determination of their degradation products. The amino acids formed from ethanol, n-propanol and n-butanol have now been shown to be O-ethyl-l-homoserine, O-propyl-l-homoserine and O-butyl-l-homoserine, respectively.  相似文献

Three new labdane type diterpenes; 4-epiagathadiol (named kayadiol), 18-hydroxymanool (named torreferol), 18-hydroxy-13-epimanool (named 13-epitorreferol), were isolated from the non-steam-volatile fraction of leaves of Torreya nucifera Sieb. et Zucc. (Taxaceae, Japanese name “Kaya”).

This is the first reported isolation these three diterpenes in a natural source.  相似文献

Strains with lac fused to each of the arylsulfatase (ats) and tyramine oxidase (tyn) operons in Escherichia coli were isolated. Synthesis of β-galactosidase in strains with tyn:: lac fusions was induced by tyramine, histamine, tryptamine, dopamine and octopamine, and the induction of the tyn operon was subject to catabolite and ammonium repressions. These repressions were relieved when the cells were grown with a poor carbon or nitrogen source. No arylsulfatase activity is detected in E. coli strains. Synthesis of β-galactosidase in strains with ats:: lac fusions was repressed by sulfur compounds. The repression was relieved by monoamine compounds, which induced tyramine oxidase synthesis. The inhibition of tyramine oxidase activity by cysteine resulted in a decrease of the derepressed synthesis of β-galactosidase in the ats:: lac fusion. Repressing and derepressing conditions for the tyn operon prevented and stimulated, respectively, expression of the ats operon. Thus, the expression of latent arylsulfatase in E. coli seems to be regulated by expression of the tyn operon.  相似文献

1. A cytoplasmic nucleotidase, which preferably hydrolyzed 5'-dUMP, was investigated in rat spleen. 2. Total activity of the nucleotidase increased about 3-fold in the spleen of anemic rat was caused by phenylhydrazine administration. This increase was repressed by the injection of methotrexate, an inhibitor of DNA synthesis. 3. Activities of heme oxygenase or acid phosphatase did not correlate to the change of the nucleotidase activity. 4. The nucleotidase catalyzed dephosphorylation of 3'(5')-dUMP, 3'(5')-dTMP and 3'-UMP more readily, in the presence of Mg2+. Its optimum pH was around 6.0-6.5. It was stimulated by the addition of deoxyinosine. 5. These catalytic properties and tissue distribution of the enzyme, abundant in the thymus, spleen and intestine, were similar to that of 5'(3')-nucleotidase in rat liver [Fritzson P. (1978) Adv. Enzym. Regul. 16, 43-61]. 6. A possible physiological significance of the nucleotidase is in reutilization of preformed pyrimidine nucleotides.  相似文献
