Sunflower cultivation is affected seriously by Sclerotinia sclerotiorum (Lib.) de Bary in Iran, particularly north-western areas. Because of economic and environmental harms by chemical control, it is necessary to develop cultivars with adequate genetic resistance for reduction of yield losses. The purpose of this study was to find an effective method of inoculation with S. sclerotiorum under field evaluations. Three stem-inoculation techniques including: 1 – mycelium plug, 2 – oxalic acid solution (OAS) and 3 – infested wheat seeds with Sclerotinia mycelium were employed under field conditions. Four genotypes including Ghalami (local variety in market), Confeta, Allstar and Master were used in this study. The lesion length, lesion width and lesion as up and down leading on the stem from inoculation site were measured after 3, 7, 10 and 14 days of inoculation. The analysis of variance showed significant difference between all employed techniques and incubation days after inoculation. Mycelial plug (MP) inoculation technique produced significantly more developed lesions on the treated stems. In spite of this effect, Master variety demonstrated reasonable resistance reaction against the disease. The progress of disease in wounded treatments was also faster than the non-wounded ones. And, the shortest time to obtain significant differences between varieties was 10?days after inoculation. By comparison of results of lesion length at flowering and seed-filling stages, the more obvious effectiveness of the disease was observed at the second stage. Finally, there were negative correlations between mean temperature and mean lesion length in all three inoculation methods.  相似文献

Two novel Aspergillus species, one belonging to the section Terrei and the other to section Flavipedes, were isolated from hypersaline soils of The National Park of Lake Urmia (Iran) and are here described as Aspergillus iranicus and Aspergillus urmiensis. A polyphasic taxonomic approach comprising extrolite profiles, phenotypic characters and molecular data (beta-tubulin, calmodulin and ribosomal polymerase II second largest subunit gene sequences) was applied to determine their novel taxonomic status. Aspergillus iranicus (CBS 139561T) is phylogenetically related to A. carneus, A. niveus, A. allahabadii and A. neoindicus, and it can be differentiated from those species by a unique extrolite pattern (citrinin, gregatins, and a terrequinone) and its conidial colour. Aspergillus urmiensis (CBS 139558T) shares a most recent common ancestor with A. templicola. The former species produces globose vesicles, and those of A. templicola are predominantly elongate. The Aspergillus urmiensis isolates produce several uncharacterized extrolites. Two other strains obtained during this study reside in a clade, together with the type strain of A. movilensis (CCF 4410T), and are identified accordingly. Based on the phylogenetic data presented in this study, A. frequens is reduced to synonymy with A. micronesiensis and A. mangaliensis is considered to be a synonym of A. templicola.  相似文献

In this study, sensitivities of 156 Sclerotinia sclerotiorum isolates collected from sunflower fields of West Azarbaijan province, Iran, were assessed to carbendazim and iprodione, and the baseline sensitivities were established for azoxystrobin and tebuconazole. Resistance to carbendazim and iprodione was observed in 53.85% and 4.49% of the isolates, respectively. The 50% effective concentration (EC50) values of azoxystrobin for the isolates ranged from 0.017 to 3.515 μg/ml with a mean of 0.330 μg/ml, and 8.97% of the strains showed low levels of resistance to the fungicide. However, in the presence of salicylhydroxamic acid, all isolates were sensitive to azoxystrobin and EC50 values ranged from 0.015 to 0.263 μg/ml with a mean of 0.086 μg/ml. All isolates were found to be sensitive to tebuconazole, and EC50 values ranged from 0.003 to 0.177 μg/ml with a mean of 0.036 μg/ml. Among the multiple-resistant isolates, the strains exhibiting resistance to both carbendazim and iprodione were detected in the highest frequency (4.49%). No correlation was observed between mycelial growth and aggressiveness with fungicide sensitivity of the isolates suggesting the absence of fitness cost associated with resistance to the studied fungicides. The results indicated that iprodione, azoxystrobin and tebuconazole could be effectively used in rotation or mixture in spray programmes to manage S. sclerotiorum in the region. The baselines established for azoxystrobin and tebuconazole would be useful in monitoring the fungal populations in the province to assess possible shifts in fungicide sensitivity of S. sclerotiorum isolates in the future.  相似文献

Leptinotarsa decemlineata is a serious potato pest throughout the world that has developed resistance to many insecticides. This study investigated the virulence of two indigenous isolates of Beauveria bassiana, consisting of DEBI007 and IR1217C. Five different concentrations of each isolate, 1 × 107, 3.1 × 107, 1 × 108, 3.1 × 108 and 1 × 109 conidia ml?1, were applied in bioassay using dipping method on second instar larvae. Control insects were treated with distilled water containing 0.01% Tween-80 (Sigma, Germany) as surfactant. The results showed that mortality percentages due to DEBI007 and IR1217C isolates in the highest concentration were 60% and 57.77%, respectively. There was a significant difference between efficacies of different conidial concentrations; therefore, with rise in conidial concentration, the mortality percentage was increased. The DEBI007 isolate that was previously isolated from the soil was more effective than IR1217C isolate. Calculated LC50 for this effective isolate was 1.4 × 107 conidia ml?1, which, respectively, is the highest concentration.  相似文献

In order to replace the conventional chemical pesticides, extensive researches have been done on entomopathogenic fungi. Entomopathogenic fungus Beauveria bassiana is an important biocontrol agent against major economic pests and is being employed in Integrated pest management (IPM) along with synthetic pesticides. Cabbage aphid Brevicoryne brassicae L. is one of the important pests of Brassicaceae family. Therefore, in this research, the virulence isolate of B. brassicae (IRAN 429C) was investigated on adults of cabbage aphid under laboratory conditions. The experiments were conducted at 25 ± 2 °C, 60 ± 10 R. H. and a photoperiod of 16:8 (L: D). After preliminary experiments, the adult aphids were treated with fungal concentrations of 1 × 103 to 1 × 107 spores/ml. Probit analysis was conducted to calculate LC50 and LC95 values for the isolate. Positive correlation was observed between concentrations and pest mortality. LC50 and LC95 values calculated for IRAN 429C isolate are 2.04 × 105 and 1.82 × 108, respectively. The mortality was counted one day after the treatment and then continued for 14 days. Cumulative mortality for 14 days after treatment varied from 54% for IRAN 429C at low concentration (103 conidia/ml) to 83% at high concentration (107 conidia/ml). The lowest LT50 was obtained at 7.67 days for IRAN 429C isolate at concentration 1 × 107 spore/ml. According to the insecticidal activity of mentioned fungi on cabbage aphid, it can be used in biocontrol programmes of B. brassicae.  相似文献

Shot hole disease is one of the most important diseases of stone fruit trees in Iran. The disease is wide spread among orchards of Prunus spp. During spring and summer of 2007, 80 monoconidial isolates of the pathogen were recovered from infected leaves, fruits and twigs of different Prunus spp. in West Azerbaijan, Tehran, Ghazvin and Razavi Khorasan provinces of Iran and were studied taxonomically. Based on morphological and physiological characteristics and growth optimal temperature, all isolates were identified as Wilsonomyces carpophilus. Seedlings of stone fruits (apricot, almond, peach, nectarine, plum, sweet cherry and sour cherry) were used for pathogenicity tests. All seedlings were susceptible to the fungal isolates and showed disease symptoms on twigs, leaves, buds and petioles. Genetic diversity of 28 selected fungal isolates was investigated based on DNA fingerprinting by random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR), using four random primers. Based on cluster analysis of the PCR results from the four primers, 10 fingerprinting groups (clonal lineages) and 27 haplotypes were identified. Clonal lineages “C”, “D” and “E”, each with six haplotypes formed the biggest clonal lineages, but other clonal lineages (“B”, “F”, “G”, “H”, “I” and “J”) included only one isolate. No correlation was detected among clonal lineages with the location of selected isolates and their host species. A correlation was found between the substrate (fruit, twig or leaf) and clonal lineages, particularly in “C” clonal lineage. The results showed that the fungus population had high genetic diversity which is distributed among the different areas of Iran.  相似文献

The interaction of arbuscular mycorrhizal fungi (Glomus etunicatum, Glomus intraradices, and Glomus versiforme) with a wilt-causing soil-borne pathogen, Verticillium dahliae, was studied in cotton. It was found that establishment by arbuscular mycorrhizal fungi reduced disease index. In diseased cotton plants colonised by G. etunicatum, the disease index was less than other diseased mycorrhizal and non-mycorrhizal ones. In diseased cotton plants, chlorophyll content was lower than others. Three Glomus species significantly increased content of sugar and protein in shoot and root. Pathogen-infected plants had higher proline concentration in shoot and root than healthy plants. On the other hand, the increased content of proline as stress sensor showed that Verticillium accelerates senescence and reduces yield. These results suggest that the beneficial effects of mycorrhiza can alleviate the pathogenesis effects of V. dahliae partly, and also there is a competitive interaction between the pathogenic and symbiotic fungi.  相似文献

During the spring and summer of 2014 and 2015, wheat and barley fields in the Iranian provinces of Golestan and Alborz showed a high incidence of symptoms of black (sooty) head mold of wheat and barley. The isolation results revealed that Alternaria was associated with these symptoms. One hundred and forty isolates were collected and morphologically characterized based on the development of conidial chains with primary, secondary, and tertiary branching patterns, consistent with the three-dimensional sporulation complexity of members of Alternaria in sections Infectoriae and Pseudoalternaria. Subsequently, 16 Alternaria isolates exhibiting high morphological diversity were characterized based on extensive morphological and molecular comparisons. Phylogenetic analyses of three loci [ITS, glyceraldehyde 3-phosphate dehydrogenase (gpd), and plasma membrane ATPase (ATPase)] revealed that 15 isolates belonged to section Infectoriae but could not be assigned to phylogenetic species and one isolate represents a new species, Alternaria kordkuyana sp. nov., in section Pseudoalternaria. Morphological assessments revealed a high degree of variation among section Infectoriae isolates and that A. kordkuyana has significant morphological differences as compared to the three other species currently described in section Pseudoalternaria.  相似文献
