Adrenal chromaffin cells secrete catecholamines in response to cholinergic receptor activation by acetylcholine (ACh). Characteristics of Ca(2+) transients induced by activation of nicotinic (nAChRs) and muscarinic (mAChRs) receptors were analyzed using Fura-2 fluorescent measurements on rat chromaffin cells. We first found two populations of chromaffin cells, which differently responded on AChR stimulation. In the first group (n-cells), consecutive ACh applications evoked persistent Ca(2+) transients, whereas desensitizing transients were observed in the other group (m-cells). The AChR agonists and antagonists precisely imitated or abolished the ACh action on n- and m-type cells, respectively. Cytochemical staining showed that n-cells contained adrenaline, whereas m-cells-noradrenaline. Thus, for the first time we found that nAChRs and mAChRs are differentially expressed in adrenergic and noradrenergic chromaffin cells, respectively. Our data suppose that chromaffin cells can be differentially regulated by incoming ACh signals and in such way release different substances-adrenaline and noradrenaline.  相似文献

Adult head structures are well known in the coleopteran suborders Archostemata and Adephaga, whereas the available information is very fragmentary in the megadiverse Polyphaga, including the successful superfamily Staphylinoidea. In the present study, the cephalic morphology of the cholevine species Catops ventricosus is described in detail and documented. The results were compared to conditions occurring in other polyphagan lineages, especially staphylinoid and scarabaeoid representatives. Specific external features documented in Catops and potential autapomorphies of Leiodidae include a five-segmented antennal club with a reduced eighth antennomere and the presence of periarticular grooves filled with sensilla on antennomeres 7, 9, and 10. The firm connection of the head and pronotum is possibly an apomorphy of Cholevinae. The monophyly of Cholevinae excluding Eucatopini and Oritocatopini is supported by the apical maxillary palpomere as long as or shorter than the subapical one, and the presence of cryptic pore plates on the surface of these palpomeres—a feature described and documented here for the first time. The internal cephalic structures of Catops are mostly plesiomorphic, as for instance the complete tentorium. The pattern of the muscles is similar to what is found in other staphylinoid taxa. The unusual maxillary muscle “Mx” is likely a groundplan apomorphy of the clade Staphyliniformia?+?Scarabaeoidea. M. hypopharyngomandibularis (M13) was identified in Catops and is ancestral for Coleoptera, even though it is often missing. The same applies to M. tentoriohypopharyngalis (M42).  相似文献

External and internal structures of the head of adults of Clambus are described and illustrated in detail. The results are compared with structural features found in the clambid genus Calyptomerus, in representatives of other scirtoid families, and also in species of other coleopteran suborders, notably Myxophaga. The results tentatively support the monophyly of Scirtoidea and a close relationship between Clambidae and Eucinetidae is suggested by one shared derived feature of the mandible, a long and slender apical tooth with a serrate edge. The monophyly of Clambidae is very strongly supported and Acalyptomerus is probably the sistergroup of a clade Calyptomerus + Clambinae. Potential scirtoid autapomorphies are the loss of the dorsal tentorial arms, a bulging gula, a strongly transverse labrum, and a ridge separating the mediostipes from the lacinia. However, all these features are homoplasious. The monophyly of Clambidae is supported by modifications of the head capsule which is strongly flattened and broadened, by a deep clypeofrontal incision enabling vertical antennal movements, and a series of antennal features. Synapomorphies of Clambinae + Calyptomerus (Clambidae excluding Acalyptomerus) are the conglobate body form with the ventral side of the head capsule in contact with the mesocoxae, and compound eyes integrated in the contour of the head. The completely subdivided eye is an autapomorphy of Clambus. An entire series of features is shared by Clambidae (or Scirtoidea) and Myxophaga. Most of them are apomorphies that apparently evolved independently in both groups. However, the presence of well‐developed maxillary and labial glands is arguably a retained groundplan feature of Coleoptera, with parallel loss in Archostemata, Adephaga and various groups of Polyphaga. The mouthparts and musculature of the head of Sericoderus lateralis are described. It is shown that the mouthparts of minute beetles preserve their complex structure and are not significantly affected by miniaturization. Such stability allows these features to be used in macrosystematics of groups with variable body size.  相似文献

The surface of unidentified cultured neurons and electrophysiologically identified units RPa1, RPa2, and LPa3 of the snailHelix pomatia was studied using a colloidal gold-labelled endogenous gonad lectin of this molluse (Helix pomatia agglutinin -HPA), which is specific to -anomer N-acetyl-D-galactosamine, and some other lectins. It was found that only two populations of cultured neurons with a specific ultrastructure typical of peptidergic cells and differing from the ultrastructure of other peptidergic and non-peptidergic neurons possessHPA receptors. These neurons average about 1% of cultured nerve cells. Using a variety of plant lectins withHPA-like specificity (LBA, VVA, SBA) showed that the surface of many peptidergic neurons contains -anomer-like structures, yet it does not bindHPA. These data were supported in the experiments on identified snall neurons with the use ofSBA. Gyrinidae is a group of beetles with a unique specialization of swimming on the water surface. Heterogyrus milloti Legros (Heterogyrinae) from Madagascar is a species with various preserved plesiomorphic features. The information on the morphology and biology was very limited until recently, and the thoracic anatomy remained largely unknown. Consequently, the aim of the present study is to describe external and internal thoracic features of Heterogyrus Legros in detail and to interprete them with respect to their phylogenetic and functional significance, with a special focus on the unusual flight apparatus of Gyrinidae. Characters documented with innovative techniques are compared to conditions found in other gyrinid genera and other groups of Adephaga, including characters of other body parts and larvae. A data matrix with 144 characters of adults, larvae and eggs was compiled and analysed cladistically. Gyrinidae excluding Spanglerogyrus Folkers (Heterogyrinae + Gyrininae) is supported by many apomorphies, mainly by a unique locomotor apparatus with paddle‐like middle and hind legs. The results confirm Heterogyrus as the earliest diverging branch in Gyrinidae except Spanglerogyrus, implying a sister‐group relationship between this genus and Gyrininae, a clade comprising Gyrinini, Dineutini and Orectochilini. The presence of an opening between the mesanepisternum and elytra, reduction of the lateral metafurcal arms, loss of the metathoracic M. furcacoxalis lateralis, and modifications of the head, including the dorsal shift of the upper subcomponent of the compound eyes, are synapomorphies of the three tribes. The monophyly of Gyrinini is moderately well‐supported, whereas Orectochilini is strongly supported by different characters including a highly simplified but functioning flight apparatus. A clade comprising Orectochilini and the dineutine genera is suggested by synapomorphies of adults and larvae. The monophyly of Dineutini was supported in a recent study, but not by the characters analysed here. Features of adults, larvae and eggs indicate that Gyrinidae are the sister group to the remaining adephagan families, as suggested in some earlier morphology‐based studies and recent analyses of large molecular datasets.  相似文献

The effects of thermophilic methane fermentation products on alfalfa (Medicago sativa L.) growth, nitrogen-fixing activity, and seed production were studied in pot and field experiments. Plant treatment with the preparation at the stages of stooling and flower bud formation resulted in an increase in the root nodule weight and their nitrogen-fixing activity, the accumulation of cytokinins (zeatin and zeatin riboside), and an increase in the green mass yield and protein content, especially at a low (0.25 of the full rate) mineral nitrogen rate. Crop seed production was considerably improved during the first (pot experiments) and second (field experiments) growing seasons. The conclusion was drawn that the causes for improved alfalfa seed production were the effects of group B vitamins, phytohormones, their interaction, and the switching of metabolic pathways toward cytokinin synthesis.  相似文献

Using a variety of colloidal gold-labelled lectins, the structure and topography of carbohydrate determinants of the surface membrane in different types of cultured glial cells of the snailHelix pomatia have been electron cytochemically investigated. Analysis of lectin binding having different sugar specificities have shown heterogeneity of carbohydrate pools between glial and nerve cells and among different types of glial cells. It was found that satellite glial cells displaying ultrastructural traits of intensive metabolism (type II cells) selectively bindGNA, which is specific for terminal -D-mannose residues, and do not interact (Con A) or slightly interact (LCA) with other mannose-specific lectins.GNA determinants remain during the whole period of cell growth and are absent in satellite type-I glial cells, fibrous glial cells, microglia, and neurons.LTA, PVA, andLABA do not bind to any glial cells.WGA determinants, which are abundant on the neurons, are completely absent onGNA-binding glial cells and single on other types of glial cells. The density ofPNA determinants on microglial cells is the highest, as compared with other types of glial cells or neurons. It is concluded that some lectin determinants (forRCA-1, PNA, LPA) are present on all types of glial cells, while another determinant (GNA) is specific for a certain type of glial cells only and can serve as a marker of these cells.
