Summary The relative efficiences of the biparental mating systems and selfing series in connection with phenotypic and geno-phenotypic selection procedures were evaluated for yield improvement in a cross HP1102 X CPAN 1681 of wheat. Two selection cycles having a 4 per cent selection intensity for grain yield were carried out following both selection procedures under the two mating systems. Following these selection procedures, a greater improvement for grain yield could be achieved with the biparental mating system than with the selfing series. During the first selection cycle, the geno-phenotypic selection procedure had an edge over phenotypic selection procedure. The realized response due to the second cycle of selection and the predicted response for the third selection cycle indicated that the phenotypic selection procedure is more efficient than the geno-phenotypic selection procedure. It is suggested that selection following intermating in early segregating generations is able to overcome several inherent limitations of the simple pedigree method as it is possible to increase genetic variation and to concentrate favourable genes and gene combinations for grain yield. An increase in grain yield was, in general, accompanied by an increase in plant height, peduncle length, 100 grain weight, tiller number and biological yield. Therefore, it is suggested that an index comprised of grain yield, plant height, tiller number, grain weight and biological yield could be used for selecting high yielding genotypes of suitable height.  相似文献

通过对2018年河南省环境污水中肠道病毒(Enterovirus,EV)的持续监测,了解河南省环境污水中脊灰病毒(PV)和非脊灰肠道病毒(NPEV)的血清型分布及流行情况.选取了河南省东、西、南、北、中部的五个城市,对每个城市有10万~30万人共同使用的下水管网的地区进行污水采样,每两个月采集1次,每次采集两份污水样品,采集持续1年,对污水样品进行浓缩,将浓缩液接种至RD细胞、L20B细胞和HEp-2细胞进行病毒分离.对病毒分离物进行VP1区核苷酸序列测定分析,采用最大似然法构建系统进化树对EV进行分子定型,并进行核苷酸相似性分析.共采集污水样品60份,分离到EV阳性毒株16株(26.67%),分别为8株CVB5、3株E7、3株E11、1株CVB5+E11混合株;1株PVIII;环境监测标本与急性弛缓性麻痹(Acute flaccid paralysis,AFP)病例标本,在济源市和周口市分离到的CVB5比例有一致性,郑州市和济源市分离到的E7比例有一致性;污水标本中阳性毒株的检出时间要早于AFP病例标本中同种毒株型别的检出时间1~3个月.来源于AFP病例监测和环境监测的CVB5之间的核苷酸相似性为94.7%~99.6%,来源于AFP病例监测和环境监测的E7之间的核苷酸相似性为92.5%~100.0%.来源于AFP病例监测和环境监测的CVB5属于同一个基因型,E7也是同一个基因型,甚至是同一个病毒传播链.河南省2018年建立了环境监测方法,是AFP病例监测的有益的补充,也可对人类肠道病毒病流行或暴发进行预测和预警.  相似文献

患者,女,61岁,确诊肺结节病、口服糖皮质激素7月,双手背及腕部红色斑块3个月。组织病理示表皮轻度棘层增厚,真皮浅、中层中性粒细胞、淋巴、组织细胞、浆细胞、多核巨细胞混合性浸润,可见散在双轮廓厚壁酵母样结构。六铵银染色见不规则肿胀菌丝及酵母样厚壁孢子。真菌培养见棕/灰褐色菌落,镜下见棕色分隔孢子。ITS区序列分析鉴定为Alternaria alternate。口服伊曲康唑胶囊200 mg/d,联合外用硝酸舍他康唑乳膏2次/d,连续用药4个月后皮损完全消退,随访半年,未见复发。  相似文献

Abstract

The interaction between bacteria and minerals is very complicated and has been intensively studied in the laboratory and the field in the last few decades, but the processes and mechanisms of biomineralization and mineral precipitation are still not fully understood and need to be explored further. In the present work, biomineralization experiments were undertaken using Klebsiella pneumoniae LH1, collected from a natural surface environment in an area of outcrops of Cambrian dolomite, in a culture medium with various Mg/Ca molar ratios (0, 3, 6 and 12). The mineral precipitates obtained were analyzed by X-ray diffraction (XRD), scanning electron microscope (SEM), energy dispersive spectrometer (EDS), Fourier transform infrared spectrometer (FTIR), laser scanning confocal microscopy (LSCM) and X-ray photoelectron spectroscopy (XPS). Cells were analyzed with a scanning transmission electron microscope (STEM), high resolution transmission electron microscope (HRTEM) and selected area electron diffraction (SAED). The composition of amino acids in extracellular polymeric substances (EPS) was also determined. In the experiments it was found that the production of ammonia and the presence of carbonate anhydrase promoted the increase of the medium pH and that minerals are nucleated on the EPS, which consist chiefly of amino acids and negatively-charged organic functional groups. With increasing Mg/Ca ratios, the mineral phases changed, including calcite (100%) at Mg/Ca molar ratio of 0, monohydrocalcite (36.05%) + dypingite (63.95%) at Mg/Ca molar ratio of 3, monohydrocalcite (29.72%) + dypingite (15.48%) + nesquehonite (54.80%) at Mg/Ca molar ratio of 6, and monohydrocalcite (14.2%) + dypingite (1.0%) + nesquehonite (84.80%) at Mg/Ca molar ratio of 12. Some intracellular amorphous calcium- and magnesium-rich inclusions were also detected in K. pneumoniae LH1, suggesting intracellular biomineralization accompanying the extracellular mineral precipitation. This study provides further understanding of the biomineralization processes of microorganisms.  相似文献

This article presents a study of struvite formation in liquid medium induced by the sulfate-reducing bacterium Acinetobacter calcoaceticus SRB4, a strain isolated from river sediment. We identified the bacterial strain A. calcoaceticus SRB4 and analyzed its micromorphology. The minerals formed were studied with an electroprobe microanalyzer, Fourier transform infrared spectroscopy, high-resolution transmission electron microscopy, selected-area electron diffraction, X-ray diffraction, thermogravimetry, differential thermogravimetry, and differential scanning calorimetry. Acinetobacter calcoaceticus SRB4 was found to induce struvite precipitation, whereas sterile control cultures did not. Many transparent stick-shaped struvite precipitates were distributed at the bottom of the conical flasks in the experimental group. Most bacteria were spherical and a large quantity of spherical struvite particles (less than 200 nm in diameter) adhered to the bacterial surface. An electron probe microanalysis showed that the precipitates contained C, O, P, Mg, and other elements. Fourier transformation infrared spectra showed that the precipitates contained crystalline water, NH4+, and PO43? groups. X-ray diffraction spectra showed that the precipitates were struvite crystals, with preferential orientation and lattice distortion. Thermogravimetry showed that the weight loss was caused by the evaporation of crystalline water at temperatures lower than 136°C and the release of ammonia from struvite at temperatures of 136–228.5°C. In this article, we discuss the possible mechanism of struvite formation and the possible role played by A. calcoaceticus SRB4. Our study extends our understanding of the phosphate biomineralization mechanism and should prove useful in recycling phosphorus in wastewater.  相似文献

A rat pheochromocytoma cell line (PC12), when transfected with beta1,4-N-acetylglucosaminyltransferase III (GnT-III), which catalyzes the formation of a bisecting GlcNAc structure in N-glycans, resulted in the suppression of neurite outgrowth induced by costimulation of epidermal growth factor (EGF) and integrins. The neurite outgrowth was restored by the overexpression of a constitutively activated mitogen- or extracellular signal-regulated kinase kinase-1 (MEK-1). Consistent with this, the EGF receptor (EGFR)-mediated ERK activation was blocked in GnT-III transfectants. Conversely, the overexpression of dominant negative MEK-1 or treatment with PD98059, a specific inhibitor of MEK-1, inhibited neurite outgrowth in controls transfected with mock. Furthermore GnT-III activity is required for these inhibitions, because the overexpression of a dominant negative GnT-III mutant (D321A) failed to reduce neurite outgrowth and EGFR-mediated ERK activation. Lectin blot analysis confirmed that EGFR from wild-type GnT-III transfectants had been modified by bisecting GlcNAc in its N-glycan structures. This modification led to a significant decrease in EGF binding and EGFR autophosphorylation. Collectively, the results constitute a comprehensive body of evidence to show clearly that the overexpression of GnT-III prevents neurite outgrowth induced by costimulation of EGF and integrins through the Ras/MAPK activation pathway and indicates that GnT-III may be an important regulator for cell differentiation in neural tissues.  相似文献

Quantitative trait loci (QTL) analysis for pre-harvest sprouting tolerance (PHST) in bread wheat was conducted following single-locus and two-locus analyses, using data on a set of 110 recombinant inbred lines (RILs) of the International Triticeae Mapping Initiative population grown in four different environments. Single-locus analysis following composite interval mapping (CIM) resolved a total of five QTLs with one to four QTLs in each of the four individual environments. Four of these five QTLs were also detected following two-locus analysis, which resolved a total of 14 QTLs including 8 main effect QTLs (M-QTLs), 8 epistatic QTLs (E-QTLs) and 5 QTLs involved in QTL × environment (QE) or QTL × QTL × environment (QQE) interactions, some of these QTLs being common. The analysis revealed that a major fraction (76.68%) of the total phenotypic variation explained for PHST is due to M-QTLs (47.95%) and E-QTLs (28.73%), and that only a very small fraction of variation (3.24%) is due to QE and QQE interactions. Thus, more than three-quarters of the genetic variation for PHST is fixable and would contribute directly to gains under selection. Two QTLs that were detected in more than one environment and at LOD scores above the threshold values were located on 3BL and 3DL presumably in the vicinity of the dormancy gene TaVp1. Another QTL was found to be located on 3B, perhaps in close proximity to the R gene for red grain colour. However, these associations of QTLs for PHST with genes for dormancy and grain colour are only suggestive. The results obtained in the present study suggest that PHST is a complex trait controlled by large number of QTLs, some of them interacting among themselves or with the environment. These QTLs can be brought together through marker-aided selection, leading to enhanced PHST.  相似文献
