Therapeutic monoclonal antibodies have several advantages over small molecule drugs and small proteins and peptides, including a long serum half-life. The long serum half-life of IgG is due, in part, to its molecular weight (150kDa) and its ability to bind FcRn. Both the CH2 and CH3 domains of Fc are involved in FcRn binding. Antibody fragments and antibody-like scaffolds have improved penetration into tissues due to their small size, yet suffer from a short serum half-life of less than one hour. The human CH2 domain (CH2D) of IgG1 retains a portion of the FcRn binding site, is amenable to modification for target binding, and may represent the smallest antibody-like scaffold retaining a relatively long serum half-life. Here we describe the generation of a dimeric CH2D (dCH2D) and determination of its pharmacokinetics (PK), as well as the PK of wild-type monomeric CH2D (mCH2D) and a short stabilized CH2D variant (ssCH2D) in normal B6 mice, human FcRn transgenic mice and cynomolgus macaques. The elimination half-life of dCH2D was 9.9, 10.4 and 11.2 hours, and that of ssCH2D was 13.1, 9.9 and 11.4 hours, in B6 mice, hFcRn mice and cynomolgus macaques, respectively. These half-lives were slightly longer than that of mCH2D (6.9 and 8.8 hours) in B6 and hFcRn mice, respectively. These data demonstrate that engineered CH2D-based variants have relatively long serum half-lives, making them a unique scaffold suitable for development of targeted therapeutics.  相似文献

C2H2+Ar处理医用涤纶材料的细菌粘附   总被引:3,自引:0,他引:3  
目的:对最常用心脏血管替代材料涤纶片作最新发展的具有全方位表面改性特征的混合等离子体浸没注入,以观察经处理后的涤纶片抑制细菌粘附的效果。方法:用多功能全方位等离子体浸没及离子注入机(PⅢ),用射频电源建立气体等离子体,对涤纶材料作全方位乙炔和氩气混合离子(C2H Ar)注入获取表面改性涤纶片。用金黄色葡萄球菌,表皮葡萄球菌,大肠杆菌,绿脓杆菌,白色念珠菌制取细菌悬液并作5-^125I-2’-脱氧尿嘧啶核苷(^125I-UDR)标记,再对改性涤纶材料作体外细菌动态粘附实验。结果:表面改性涤纶材料改变了亲水性和表面能,降低了水分子接触角。与未改性材料相比,改性涤纶材料抗细菌粘附能力有较明显提高。结论:混合离子(C2H2 Ar)表面改性涤纶片有良好的抗细菌和血小板粘附能力。  相似文献

长鞘卷叶甲Leptispa longipennis(Gestro)是为害绿竹等丛生竹的新害虫,该虫在福建永定一年发生1代,以成虫在卷起的竹叶内越冬。翌年4月上旬开始交尾产卵,卵期10无;5月中旬开始化蛹,蛹期8无:5月中下旬第一代成虫开始羽化。以幼虫及成虫在卷起的竹叶内取食及繁殖。在对其生物学特性进行系统研究的基础上,找到了该虫生活史中的薄弱环节,提出了在7月份新羽化成虫在卷起的竹叶内取食盛期喷洒灭多粉、1605粉荆;初孵化幼虫喷洒白僵菌粉剂、25%灭幼脲粉剂、灭多粉粉荆、1605粉剂的防治技术。  相似文献

运用形态学标记和分子标记对江苏省文蛤良种场红壳色文蛤F1代养殖群体(父母本为江苏野生红壳色群体)进行遗传多样性分析。用文蛤壳长、壳宽、壳高和体重4个可量性状进行形态学数据的聚类分析,显示可量性状变异系数在38.48%-82.95%。运用7个引物微卫星基因座的多态性进行了养殖群体F1代的分子标记评估,结果表明,7个微卫星基因位点的平均等位基因数3.857 1,平均有效等位基因数2.583 0,平均观察杂合度0.565 3,平均期望杂合度0.565 3,Shannon指数平均数1.050 1,多态信息含量平均数0.522 5。综合形态学数据和分子标记的研究结果表明,红壳色文蛤江苏养殖群体F1代的遗传多样性处于中度偏高水平,具有较高的遗传改良潜力。  相似文献

The shell color polymorphism widely exists in economic shellfish, which not only results in a better visual perception but also shows great value as an economic trait for breeding. Small numbers of reddish-orange shell Yesso scallops, Patinopecten yessoensis, were found in cultured populations compared to the brown majority. In this study, a genome-wide association study was conducted to understand the genetic basis of shell color. Sixty-six 2b-RAD libraries with equal numbers of reddish-orange and brown shell individuals were constructed and sequenced using the Illumina HiSeq 2000 platform. A total of 322,332,684 high-quality reads were obtained, and the average sequencing depth was 18.4×. One genomic region on chromosome 11 that included 239 single-nucleotide polymorphisms (SNPs) was identified as significantly associated with shell color. After verification by high-resolution melting in another population, two SNPs were selected as specific loci for reddish-orange shell color. These two SNPs could be used to improve the selective breeding progress of true-breeding strains with complete reddish-orange scallops. In addition, within the significantly associated genomic region, candidate genes were identified using marker sequences to search the draft genome of Yesso scallop. Three genes (LDLR, FRIS, and FRIY) with known functions in carotenoid metabolism were identified. Further study using high-performance liquid chromatography proved that the relative level of carotenoids in the reddish-orange shells was 40 times higher than that in the brown shells. These results suggested that the accumulation of carotenoids contributes to the formation of reddish-orange shells.  相似文献

The protein-tyrosine kinase substrate annexin II is a growth regulated gene whose expression is increased in several human cancers. While the precise function of this protein is not understood, annexin II is proposed to be involved in multiple physiological activities, including DNA synthesis and cell proliferation. Targeted disruption of the annexin II gene affects calcium signaling, tyrosine phosphorylation and apoptosis, indicating the important physiological role of this protein. We used a transient co-transfection assay to regulate annexin II expression in human HeLa, 293 and 293T cells, and measured the effects of annexin II down regulation on DNA synthesis and proliferation. Transfection of cells with an antisense annexin II vector results in inhibition of cell division and proliferation, with concomitant reduction in annexin II message and protein levels. Cellular DNA synthesis is significantly reduced in antisense transfected cells. Replication extracts made from antisense transfected cells have significantly reduced efficiency to support SV40 in vitro DNA replication, while the extracts made from sense transfected cells are fully capable of replication. Our results indicate an important role of annexin II in cellular DNA synthesis and cell proliferation.  相似文献

湿地生态健康评价对于掌握湿地的健康状况、理解人类活动干扰对其影响及实施生态预警等有重要意义。鄱阳湖对于维持其流域甚至长江中下游的生态平衡十分重要,目前尚未建立起其较完善的生态健康评价指标体系。大型底栖无脊椎动物完整性指数(benthic macroinvertebrate-based index of biotic integrity, B-IBI)是广泛应用的湿地生态健康评价方法。基于30个采样点(7个参照点,23个受损点)的大型底栖无脊椎动物采样数据,构建鄱阳湖湿地的B-IBI指数,采用自然断点法划分非常健康、健康、一般、差和极差5个健康等级标准,据此评价湿地生态健康状况。研究表明:(1)基于B-IBI指数的鄱阳湖湿地生态健康评价结果为一般;(2)就采样点B-IBI分值而言,呈现出西部健康状况优于东部的格局,其中国家级自然保护区内状况较好,而工业区、城镇、农田及河流入湖口附近状况较差;(3)B-IBI指数与前期构建的景观发展强度指数(LDI)、栖息地环境质量指数(QHEI)以及植被完整性指数(V-IBI)具有显著相关性,表明基于不同指数的评价结果较为一致。本研究构建的B-IBI指数能为鄱阳湖湿地的生态健康评价和监测提供重要方法。  相似文献

Microtubules are composed of αβ-tubulin heterodimers and have been treated as highly attractive targets for antitumor drugs. A broad range of agents bind to tubulin and interfere with microtubule assembly, including colchicine binding site inhibitors (CBSIs). Tubulin Polymerization Inhibitor I (TPI1), a benzylidene derivative of 9(10H)-anthracenone, is a CBSI that inhibits the assembly of microtubules. However, for a long time, the design and development of anthracenone family drugs have been hindered by the lack of structural information of the tubulin-agent complex. Here we report a 2.3 Å crystal structure of tubulin complexed with TPI1, the first structure of anthracenone family agents. This complex structure reveals the interactions between TPI1 and tubulin, and thus provides insights into the development of new anthracenone derivatives targeting the colchicine binding site.  相似文献
