Pneumocystis carinii is a pathogen which, causes fatal pneumonia in patients with the acquired immune deficiency syndrome (AIDS). To facilitate the basic study of P. carinii , we have analyzed its major surface proteins by both immunochemical and biochemical methods. The major protein components of both cysts and trophozoites are a group of proteins called "P115" with apparent masses of 105–120 kd. It includes 6 isoelcclric variants. A monoclonal antibody raised against cysts recognizes all 6 variants and reacts with epitopes located in the cell wall indicating that P115 is an immunorcactive surface component. The isoelectric variants contain identical or closely related protein components and they are mannose-rich glycoproteins. The isoelectric variation may be due primarily to differences in glycosylation. The majority of sera from humans with diagnosed pneumocystosis that were tested reacted strongly with the P115 proteins. To develop probes for DNA diagnosis and to facilitate molecular studies, a genomic DNA library of P. carinii has been constructed. Some of these clones were used for DNA hybridization analysis of rat and human lungs.  相似文献

Summary 1. To examine spatial heterogeneity of trophic pathways on a small scale (<5 m diameter), we conducted dual stable isotope (δ13C and δ15N) analyses of invertebrate communities and their potential food sources in three patchy habitats [sphagnum lawn (SL), vascular‐plant carpet (VC) and sphagnum carpet] within a temperate bog (Mizorogaike Pond, Kyoto, Japan). 2. In total, 19 invertebrate taxa were collected from the three habitats, most of which were stenotopic, i.e. collected from a single habitat. Amongst the habitats, significant variation was observed in the isotopic signatures of dominant plant tissues and their detrital matter [benthic particulate organic matter (BPOM)], both of which were potential organic food sources for invertebrates. Site‐specific isotopic variation amongst detritivores was found in δ13C but not in δ15N, reflecting site‐specificity in the isotopic signatures of basal foods. The eurytopic hydrophilid beetle Helochares striatus was found in all habitats, but showed clear site variation in its isotopic signatures, suggesting that it strongly relies on foods within its own habitat. 3. The most promising potential foods for detritivores were the dead leaf stalks of a dominant plant in the VC and BPOM in the SL and carpet. An isotopic mixing model (IsoSource version 1.3.1) estimated that aquatic predators rely on unknown trophic sources with higher δ13C than detritus, whereas terrestrial predators forage on allochthonous as well as autochthonous prey, suggesting that the latter predators might play key roles in coupling between habitats. 4. Our stable isotope approach revealed that immobile detritivores are confined to their small patchy habitats but that heterogeneous trophic pathways can be coupled by mobile predators, stressing the importance of habitat heterogeneity and predator coupling in characterising food webs in bog ecosystems.  相似文献

Abstract:  A new genus and species of an elasmosaurid plesiosaur, Futabasaurus suzukii , is described based on a partial skeleton from the Inoceramus amakusensis Zone (Lower Santonian, Upper Cretaceous) of the Irimazawa Member of the Tamayama Formation, Futaba Group, in Fukushima Prefecture, Japan. The new taxon is characterized by a number of characters such as the wide space between the orbit and external naris, posterior extension of the interclavicle, relatively long humerus and prominent femoral muscle scar. The holotype includes a partial skull and mandible, posterior cervicals to sacrals, ribs, clavicular arch, pelvic girdle and four limbs. The remains are mostly in articulation, and exhibit evidence of predation/scavenging by sharks. The distribution of elasmosaurid species in the circum-Pacific region remains unclear due to the lack of diagnostic materials. The occurrence of F. suzukii is geographically and stratigraphically significant, because it allows species-level comparison; as a diagnosable elasmosaurid specimen, F. suzukii is the first and the oldest from the northern Pacific.  相似文献

An acid α-glucosidase (EC 3.2.1.20) was purified to homogeneity from the culture medium of Tetrahymena thermophila CU 399. Its general molecular, catalytic and immunological properties were compared to those of the T. pyriformis W enzyme. The enzyme from T. thermophila was a 105-kD monomer and the N-terminus (25 amino acid residues) displayed some homology with that of T. pyriformis enzyme. The purified enzyme was most active at 56° C and showed resistance to thermal inactivation. The acid α-glucosidase appears to have α-1,6-glucosidase as well as α-1,4-glucosidase activity. The Km values determined with p-nitrophenyl-α-glucopyranoside, maltose, isomaltose and glycogen were 0.7 mM, 2.5 mM, 28.5 mM and 18.5 mg/ml, respectively. The enzyme was antigenically distinct from T. pyriformis acid α-glucosidase.  相似文献
